Effect of Dietaryn-3Polyunsaturated Fatty Acids on Oxidant/Antioxidant Status in Macrosomic Offspring of Diabetic Rats

The aim of this work was to determine the effect of dietaryn-3PUFA on oxidant/antioxidant status,in vitrovery low and low density lipoprotein (VLDL-LDL), and VLDL-LDL-fatty acid composition in macrosomic pups of diabetic mothers. We hypothesized thatn-3PUFA would improve oxidative stress in macrosomia. Diabetes was induced in female Wistar rats fed with the ISIO diet (control) or with the EPAX diet (enriched inn-3PUFAs), by streptozotocin. Iridoid extracts from Ajuga iva increase the antioxidant enzyme activities in red blood cells of rats fed a cholesterol-rich diet.

The lyophilized aqueous extract of Ajuga iva (Ai) is able to reduce oxidative stress, which may prevent lipid peroxidation in hypercholesterolemic rats. Iridoids (I) were isolated from Ai. We hypothesized that the antioxidant defense status in red blood cells (RBC) and tissues in rats fed a cholesterol-rich diet and treated with Ai may be correlated to these compounds. Male Wistar rats (n = 32) weighing 120 +/- 5 g were fed a diet containing 1% cholesterol for 15 days. Abstract The aim of the study was to investigate the effect of aqueous extract of Ajuga iva ( Ai ) on serum and tissues lipid peroxidation as well as antioxidant enzymes activities in red blood cells (RBC) and tissues, in high hypercholesterolemic rats (HC). Male Wistar rats ( n =12) were fed on 1% cholesterol-enriched diet for 15 d. Mitochondria from females exhibit higher antioxidant gene expression and lower oxidative damage than males

We have investigated the differential mitochondrial oxidative stress between males and females to understand the molecular mechanisms enabling females to live longer than males. Mitochondria are a major source of free radicals in cells. Those from female rats generate half the amount of peroxides than those of males. This does not occur in ovariectomized animals. Estrogen replacement therapy prevents the effect of ovariectomy. Mitochondria from females have higher levels of reduced glutathione than those from males. The hemodynamic and anti-ischemic effects of nitroglycerin (GTN) are rapidly blunted as a result of the development of nitrate tolerance. Hydralazine has been shown to prevent tolerance in experimental and clinical studies, all of which may be at least in part secondary to antioxidant properties of this compound. Involvement of endogenous nitric oxide in the inhibition by endotoxin and interleukin-1 beta of gastric acid secretion.

Administration of Escherichia coli endotoxin abolished the acid secretory response induced by a bolus injection of pentagastrin in the continuously perfused stomach of the anaesthetized rat. Likewise, acid secretion stimulated by the continuous intravenous perfusion of pentagastrin was inhibited by administration of interleukin-1 beta (IL-1 beta). In both cases pretreatment with NG-nitro-L-arginine methyl ester (L-NAME) but not dexamethasone or indomethacin substantially restored the secretory responses to pentagastrin. Effects of fluoxetine on behavioral deficits evoked by chronic social stress in rats.

Recently, we described an advanced model of chronic social stress in male rats based on the resident intruder paradigm. In this model, rats subjected to daily social stress for 5 weeks showed behavioral changes resembling anhedonia and motivational deficits in humans. In the present study, male Wistar rats were subjected to 5 weeks of daily social defeat by an aggressive conspecific and concomitant treatment with the antidepressant drug fluoxetine (10 mg/kg) after the first week of stress.
