Exogenous makisterone A accelerates early embryonic development in the milkweed bugOncopeltus fasciatus

Reproducing females ofOncopeltus fasciatus which were treated with exogenous makisterone A and 20-hydroxyecdysone laid eggs with considerably elevated ecdysteroid contents. The early embryonic development was markedly accelerated when the mother was treated with makisterone A, whereas 20-hydroxyecdysone had no influence.

Effects of a new IGR-ovicide on a heteropteran (Oncopeltus fasciatusDallas) and lepidopteran (Manduca sextaJohansen) insect

A new insect ovicide (Ov. 165049), 4-methoxy-benzoic acid propargylester, synthesized by BASF (Ludwigshafen) was tested on Oncopeltus fasciatus and Manduca sexta. In the case of O. fasciatus also the effects on last-instar larvae and adult bugs were studied. Topical application of 40 μg/larva dissolved in acetone caused a mortality exceeding 50% at day 1 after treatment. The few animals which developed into imagines did not reproduce. A treatment with 4 μg/larva had no severe impact on metamorphosis and reproduction. Adult O. fasciatus received doses of 40 μg, 20 μg and 4 μg per animal. The presence of neurosecretory cells in the frontal ganglion (FG) has been demonstrated only in the Lepidoptera Bombyx (BOUNHIOL et al., 1953), Manduca (BORG et al., 1973; BELL et al., 1974), and Diatraea (YIN and CHIPPENDALE, 1975). In Manduca and Diatraea the neurosecretory material of the FG is apparently involved in the regulation of diapause.

It has been tested whether juvenile hormone plays a role in the larval-adult transformation of lateral oviducts in the milkweed bug. The transformation is ecdysteroid-dependent, as was reported previously2. Application of precocene or juvenile hormone III proved that the absence of juvenile hormone is required.

We have recently shown that the aggregation factor (AF) from the sponge Geodia cydonium stimulates DNA synthesis in quiescent, dissociated cells from the same organism; this event was correlated with the release of the two second messengers: inositol trisphosphate and diacylglycerol. Here we describe that after binding of the AF to the plasma membrane-bound aggregation receptor, a rapid and drastic increase in the incorporation of 32Pi into a series of proteins in the pore complex-lamina fraction occurs. Effects of azadirachtin on reproduction and egg development of the heteropteranOncopeltus fasciatusDallas

Azadirachtin injected into newly hatched adults of Oncopeltus fasciatus affected longevity, fecundity and hatchability of eggs from treated parents. There were marked differences between males and females. Whereas mortality was higher in females, reproduction was interrupted at lower doses in males: 0.125 μg caused impotency. Topical treatment of normal eggs had no demonstrable effect on egg hatch. The studies showed that in respect to a possible use of azadirachtin in insect pest managment, the interruption of reproduction and adult development is of no less importance then growth interruption in larvae (Dorn et al. 1986). Ovicidal Activity of Maternally Applied Ecdysteroids in the Large Milkweed Bug (Heteroptera: Lygaeidae)

Exogenous ecdysteroids (ecdysone, 20-hydroxyecdysone and makisterone A) administered to reproducing females of Oncopelus fasciatus (Dallas) reduced egg hatch drastically. The ovicidal activity apparently stemmed from ecdysteroids—unchanged hormone or metabolites—transferred from mother into oocyte. Different stages of embryogenesis were affected. Makisterone A, the predominant ecdysteroid in normal embryos, is by far the most effective ecdysteroid.

Studies on protein kinases involved in regulation of nucleocytoplasmic mRNA transport

The rate of energy-dependent nucleoside triphosphatase (NTPase)-mediated nucleocytoplasmic translocation of poly(A)-containing mRNA [poly(A)+mRNA] across the nuclear envelope is thought to be regulated by poly(A)-sensitive phosphorylation and dephosphorylation of nuclear-envelope protein. Studying the phosphorylation-related inhibition of the NTPase, we found that phosphorylation of one polypeptide of rat liver envelopes by endogenous NI- and NII-like protein kinase was particularly sensitive to poly(A).
