International audience; This study explores the mechanisms responsible for the fatty liver setup in mice fed trans-10,cis-12 conjugated linoleic acid (t10c12 CLA), hypothesizing that an induction of low density lipoprotein receptor (LDLR) expression is associated with lipid accumulation. To this end, the effects of t10c12 CLA treatment on lipid parameters, serum lipoproteins, and expression of liver lipid receptors were measured in LDLR(-/-) apoB(100/100) mice as a model of human familial hypercholesterolemia itself depleted of LDLR. Abstract Peroxisomal ABC transporters encoded by the ABCD genes are thought to participate in the import of specific fatty acids in the peroxisomal matrix. ABCD1 deficiency is associated with X-linked adrenoleukodystrophy (X-ALD), the most frequent peroxisomal disorder which is characterized by the accumulation of saturated very-long-chain fatty acids (VLCFA). AbstractConjugated linoleic acids (CLA) have recently been recognized to reduce body fat and plasma lipids in some animals. This study demonstrated that the steatosis accompanying the fat loss induced by trans-10,cis-12-C18:2 (CLA2) and not cis-9,trans-11-C18:2 (CLA1) isomer in C57BL/6j mice was not due to an alteration of the liver lipoprotein production that was even increased. The 3-fold decrease in plasma triacylglycerol contents and the induction of mRNA expression of low-density lipoprotein receptors concomitantly observed in CLA2-fed mice suggested an increase in the lipoprotein clearance at the level of the liver itself. Incorporation and metabolism of dietary trans isomers of linolenic acid alter the fatty acid profile of rat tissues

To study the influence on lipid metabolism and platelet aggregation of the fatty acid isomerization that occurs during heat treatment, weanling rats were fed for 8 wk a diet enriched with 5% isomerized (experimental group) or normal (control group) canola oil. Trans isomers of polyunsaturated fatty acids (PUFA) are formed during heat treatment of oils. In the present work, the effect of dietary geometrical isomers of α-linolenic acid (18:3 n-3) on the Δ6 desaturation of all cis 18:3 n-3 was investigated, using rat liver microsomes. The desaturation rates were higher in microsomes from animals fed a cis n-3 deficient diet, as compared to those from the control group or those from rats fed 18:3 n-3 and its trans isomers. Fatty acid composition of french butters, with special emphasis on conjugated linoleic acid (CLA) isomers

Conjugated linoleic acids (CLA) are fatty acids primarily found in dairy products. These fatty acids may have potential biological properties interesting for human health. Update of French food composition data banks was needed to determine CLA levels in French dairy products and to evaluate CLA intake by the French population. As part of this investigation, we analyzed 54 butters from local producers at different times of the year. The average CLA levels in butter were 0.45 g CLA/100 g butter in winter, 0.58 in spring, and 0.80 in summer. In situ hybridization of dihydroxyacetone phosphate acyltransferase, the regulating enzyme involved in plasmalogen biosynthesis

International audience; In situ hybridization can be carried out using different methods. The experimenter has to choose various parameters: the type of tissue fixation, the time of incubation, and the duration of the exposure time. All these parameters are determinant for the sensitivity and the resolution of this technique. This publication of technical aspects described different experiments performed for in situ hybridization on liver tissue. We may conclude on the parameters to optimize each step of the hybridization procedure. Moreover, this technique could be transposed to the brain and applied to little structures with a light expression of DHAP-AT.

Acute in vivo administration of a fish oil-containing emulsion improves post-ischemic cardiac function in n-3-depleted rats

Bioavailability of dietary omega-3 DHA and lutein for the retina: results from a preclinical study in mini-pigs with encapsulated nutrients

Purpose: Epidemiological studies support the hypothesis that dietary omega-3 long chain polyunsaturated fatty acids and carotenoids prevent from Age-related Macular Degeneration (AMD). AREDS2 failed to find benefits of omega-3 supplementation on the progression to advanced AMD. However, lower risk progression was reported in AMD patients of the NAT2 study whom omega-3 incorporation in red blood cells and plasma was the highest. Modification of the monoaminergic neurotransmitters in frontal cortex and hippocampus by dietary trans alpha linolenic acid in piglets

International audience; The effect of partial isomerization of dietary α-linolenic acid on the monoaminergic neurotransmitters in piglets was studied. After feeding the animals for 14 days with diets containing or not trans α-linolenic acid, neurotransmitters related to the monoaminergic function were quantified in the frontal cortex and in the hippocampus. The partial isomerization of dietary α-linolenic acid resulted in increasing endogenous monoamine levels in the frontal cortex (+55% for dopamine) and was related to a very low incorporation of trans polyunsaturated fatty acids.
