Polymorphonuclear leukocyte membrane fluidity and cytosolic Ca2+ content at baseline and after activation in essential hypertension

Obiective: To examine in essential hypertension (EH) the polymorphonuclear leukocyte (PMN) membrane fluidity and cytosolic Ca2+ content at baseline and after in vitro activation. Subjects and methods: In 45 hypertensive subjects (27 men and 18 women; mean age 45.8 +/- 11.3 years) we evaluated the PMN membrane fluidity, obtained by labeling PMNs with 1-[4-(trimethyImino)phenyl]-6phenyl-1,3,5-hexatriene (TMA-DPH) and considering the TMA-DPH polarization degree, inversely related to the membrane fluidity, and the PMN cytosolic Ca2+ content, obtained by marking PMNs with Fura 2-AM. These PMN parameters were examined at baseline and after in vitro activation (prolonged for 5 min) with two agents…

Vascular atherosclerotic disease: evaluation of the whole-blood filterability and red cell membrane microrheological pattern after intravenous administration of a single dose of pentoxifylline.

SummaryA study was carried out in 10 patients with vascular atherosclerotic disease to investigate the effects of a single dose of 300 mg pentoxifylline, given by intravenous infusion over 30 minutes, on various macrorheological and nzicrorheological parameters. Measurements were made at baseline, at the end of the intravenous infusion and again 30 minutes later of whole-blood filterability, erythrocyte membrane fluidity (using pyrene as probe) and transverse fluidity gradient (using fluorescent fatty acids as probes). The results showed that there was a significant variation in whole-blood filterability and the red cell membrane fluidity gradient after pentoxifylline. No difference, howeve…

Polymorphonuclear leukocyte membrane fluidity and cytosolic Ca2+ concentration in subjects with vascular atherosclerotic disease subdivided according to the extent.

An abnormal activation state of polymorphonuclear leukocytes (PMN) plays a key role in organ injury induced by vascular atherosclerotic disease (VAD) and diabetes mellitus (DM). PMN membrane fluidity and cytosolic Ca2+ content can be considered markers of PMN activation. In this research we evaluated the PMN membrane fluidity and cytosolic Ca2+ content in VAD subjects with and without type 2 DM and examined the association between these parameters and the mono- or polyvascular localization. We enrolled 155 VAD subjects, including 92 non-diabetic (group A: mean age 63.6 +/- 9.2 years) and 63 diabetic patients (group B: mean age 65.4 +/- 7.8 years). Among group A 63 patients had monovascular …

Prevalence of renal failure and use of antithrombotic prophylaxis among medical inpatients at increased risk of venous thromboembolic events.

Abstract Background Evidence-based guidelines recommend the use of antithrombotic prophylaxis in medical patients at risk of venous thromboembolism (VTE). Low molecular weight heparins (LMWHs) are usually preferred to unfractionated heparin. However, when prophylactic doses of LMWH are administered, patients with renal failure (RF) are exposed to the risk of excessive accumulation, and thus to an increased risk of bleeding. We aimed to assess the prevalence of RF among medical inpatients at increased risk of VTE and the use and dosage of antithrombotic prophylaxis in these patients. Methods In a cross sectional study carried out at three different hospitals, information on all medical inpat…

Membrane fluidity, membrane lipid pattern, and cytosolic Ca2+ content in platelets from a group of type II diabetic patients with macrovascular complications

OBJECTIVE To evaluate platelet membrane fluidity and some platelet metabolic parameters in type II diabetic patients with macrovascular complications. RESEARCH DESIGN AND METHODS In a group of 21 type II diabetic patients with macrovascular complications, we evaluated platelet membrane fluidity [marking intact resting platelets with the fluorescent probe 1,4-(trimethylamino)-phenyl-4-phenylhexatriene (TMA-DPH)], platelet membrane lipid pattern (cholesterol :phospholipid [C:PL] ratio and individual phospholipids), and platelet cytosolic Ca2+ content (marking intact resting platelets with the fluorescent probe Fura 2AM). RESULTS Platelet membrane fluidity is decreased in type II diabetic pat…

Hemorheologic profile of hyperlipidemic patients treated with gemfibrozil

Abstract We measured the plasma lipid levels and the macrorheologic (whole-blood, plasma, and serum viscosity levels; fibrinogen; hematocrit; mean erythrocyte aggregation; and whole-blood filterability) and microrheologic (erythrocyte membrane fluidity and red cell membrane protein lateral mobility) determinants in a group of 19 hyperlipidemic patients (Fredrickson's classification phenotype IIa, 12 patients; phenotype IV, 4; phenotype IIb, 3) at baseline and after 45 and 90 days of treatment with gemfibrozil (900 mg orally once a day). At baseline we noted statistically significant increases in plasma viscosity level, fibrinogen, and mean erythrocyte aggregation, as well as a statistically…

Polymorphonuclear leukocyte: rheology, metabolism and integrin pattern in vascular atherosclerotic disease and in type 2 diabetes mellitus

Leukocytes, and in particular polymorphonuclear cells (PMN), play a role in the organ injury that characterizes the progression of vascular atherosclerotic disease (VAD) and diabetes mellitus (DM). We enrolled subjects with VAD, subjects with type 2 DM (DM2) and subjects with VAD and DM2. We evaluated the initial relative flow rate (IRFR) of PMN, using the St. George Filtrometer, the PMN membrane fluidity, labelling intact PMN cells with the fluorescent probe 1.4-(trimethylamino)-phenyl-4-phenylhexatriene (TMA-DPH), the PMN cytosolic Ca2+ content marking the cells with the fluorescent probe Fura 2-AM and the PMN integrin profile using the flow cytofluorimetry. All these evaluations were eff…

Brain histamine and oleoylethanolamide restore behavioral deficits induced by chronic social defeat stress in mice.

The physiological mechanisms underlying the complex interplay between life stressors and metabolic factors is receiving growing interest and is being analyzed as one of the many factors contributing to depressive illness. The brain histaminergic system modulates neuronal activity extensively and we demonstrated that its integrity is necessary for peripheral signals such as the bioactive lipid mediator oleoylethanolamide (OEA) to exert its central actions. Here, we investigated the role of brain histamine and its interaction with OEA in response to chronic social defeat stress (CSDS), a preclinical protocol widely used to study physio-pathological mechanisms underlying symptoms observed in d…

Relationship between elastase and total antioxidant status in young subjects with recent myocardial infarction

In a group of young subjects with acute myocardial infarction (AMI) (68 men and 7 women; mean age 39.6 ± 5.7 years) we examined the plasma concentration of elastase, the thiobarbituric acid-reactive substances (TBARS) and the total antioxidant status (TAS) at the initial stage of AMI. In this group we found an increase of elastase (p <0.001) and TBARS (p < 0.001) and a decrease of TAS (p < 0.001). A statistical correlation was observed in the whole group of AMI patients between plasma elastase and TAS (p < 0.01) and this correlation was more statistically significant in patients with more risk factors and not in those with more involved vessels.

CLINICAL AND LABORATORY FINDINGS OF BOUTONNEUSE FEVER IN SICILIAN CHILDREN

The spectrum of signs and symptoms of 645 consecutive children diagnosed from 1984 to 1996 with boutonneuse fever (BF), a mild rickettsial disease caused by Rickettsia conorii endemic in the Mediterranean basin, are reported. The major clinical features were fever (97.2%), exanthema (96.1%) and “tache noire” (71.8%). The large series examined permitted the authors to observe some rare or disregarded clinical features of the disease: cases with papulovesicular exanthema, reported previously only in adults who had been infected by R. conorii in Africa; and cases in which the only symptom was an isolated lymphadenopathy. Conclusion R. conorii infection should be considered in patients with lym…
