ZusammenfassungDer Artikel gibt einen Überblick über die aktuellen Therapieoptionen des metastasierten hormonrezeptorpositiven und HER2-negativen Mammakarzinoms. Im Fokus stehen Kombinationstherapien z. B. mit CDK4/6-Inhibition im Vergleich zu rein endokrin basierten Therapien in Prä- und Postmenopause mit Darlegung der neuesten Studienergebnisse. Multiple independent variants at the TERT locus are associated with telomere length and risks of breast and ovarian cancer

Journal article TERT-locus SNPs and leukocyte telomere measures are reportedly associated with risks of multiple cancers. Using the Illumina custom genotyping array iCOGs, we analyzed ~480 SNPs at the TERT locus in breast (n = 103,991), ovarian (n = 39,774) and BRCA1 mutation carrier (n = 11,705) cancer cases and controls. Leukocyte telomere measurements were also available for 53,724 participants. Most associations cluster into three independent peaks. Treatment of Advanced Hormone Receptor-Positive (HR+) HER2-negative Breast Cancer

AbstractThe article gives an overview of current treatment options for metastatic hormone receptor-positive and HER2-negative breast cancer. The focus is on combined therapies, e.g., with CDK4/6 inhibition compared with purely endocrine-based therapies in the pre- and postmenopause, presenting the latest study results. The addition of a CDK4/6 inhibitor to endocrine-based therapy with an aromatase inhibitor or fulvestrant leads to a marked improvement in progression-free survival and is independently beneficial whether palbociclib, ribociclib or abemaciclib is involved.
