Biomolecular conjugation inside synthetic polymer nanopores via glycoprotein-lectin interactions
We demonstrate the supramolecular bioconjugation of concanavalin A (Con A) protein with glycoenzyme horseradish peroxidase (HRP) inside single nanopores, fabricated in heavy ion tracked polymer membranes. Firstly, the HRP-enzyme was covalently immobilized on the inner wall of the pores using carbodiimide coupling chemistry. The immobilized HRP-enzyme molecules bear sugar (mannose) groups available for the binding of Con A protein. Secondly, the bioconjugation of Con A on the pore wall was achieved through its biospecific interactions with the mannose residues of the HRP enzyme. Logic gates using nanofluidic diodes based on conical nanopores functionalized with polyprotic acid chains.
Single-track conical nanopores functionalized with polyprotic acid chains have pH-sensitive fixed charge groups and show three levels of conductance that allow integrating several functions on a single nanofluidic diode. Nanometer-scaled pores have previously been employed in separation and sensing but not in logic devices, despite the fact that biological ion channels with pH-dependent fixed charges are known to be responsible for information processing in biophysical structures. As a preliminary application, we propose a logic gate scheme where binary and multivalued logical functions are implemented.
Precision Laser Spectroscopy of the Ground State Hyperfine Splitting of HydrogenlikeBi82+209
The first direct observation of a hyperfine splitting in the optical regime is reported. The wavelength of the $M1$ transition between the $F=4$ and $F=5$ hyperfine levels of the ground state of hydrogenlike $^{209}\mathrm{Bi}^{82+}$ was measured to be ${\ensuremath{\lambda}}_{0}=243.87(4)$ nm by detection of laser induced fluorescence at the heavy-ion storage ring ESR at GSI. In addition, the lifetime of the laser excited $F=5$ sublevel was determined to be ${\ensuremath{\tau}}_{0}=0.351(16)$ ms. Hydrogen peroxide sensing with horseradish peroxidase-modified polymer single conical nanochannels.
Inspired from the funtioning and responsiveness of biological ion channels, researchers attempt to develop biosensing systems based on polymer and solid-state nanochannels. The applicability of these nanochannels for detection/sensing of any foreign analyte in the surrounding environment depends critically on the surface characteristics of the inner walls. Attaching recognition sites to the channel walls leads to the preparation of sensors targeted at a specific molecule. There are many nanochannel platforms for the detection of DNA and proteins, but only a few are capable of detecting small molecules. Laser cooling of stored high-velocity ions by means of the spontaneous force
A longitudinal laser cooling of ion beams at about 5% of the velocity of light has been performed at the Heidelberg Test Storage Ring with various cooling schemes employing the spontaneous force. For a 7.29-MeV $^{9}\mathrm{Be}^{+}$ beam with an initial longitudinal temperature of 2700 K, the main characteristics of laser cooling in a storage ring are discussed. When undamped, the transverse betatron oscillations of the coasting ions limit the longitudinal temperature after laser cooling to typically 1 K. After damping the transverse motion by precooling the ions with an electron cooler, longitudinal temperatures of below 30 mK have been obtained in the subsequent laser cooling. In this cas…
First laser cooling of relativistic ions in a storage ring
The first successful laser cooling of ions at relativistic energies was observed at the Heidelberg TSR storage ring. A $^{7}\mathrm{Li}^{+}$-ion beam of 13.3 MeV was oberlapped with resonant copropagating and counterpropagating laser beams. The metastable ions were cooled from 260 K to a longitudinal temperature of below 3 K and decelerated by several keV. The longitudinal velocity distribution was determined by a fluorescence method. After laser cooling a strongly enhanced narrow peak appeared in the Schottky noise spectrum in addition to the uncooled ion distribution.
Ionic conduction, rectification, and selectivity in single conical nanopores
Modern track-etching methods allow the preparation of membranes containing a single charged conical nanopore that shows high ionic permselectivity due to the electrical interactions of the surface pore charges with the mobile ions in the aqueous solution. The nanopore has potential applications in electrically assisted single-particle detection, analysis, and separation of biomolecules. We present a detailed theoretical and experimental account of the effects of pore radii and electrolyte concentration on the current-voltage and current-concentration curves.
