Mosaic Study: Actualization of Overall Survival (Os) with 10 Years Follow Up and Evaluation of Braf. By Gercor and Mosaic Investigators

ABSTRACT Aim: The MOSAIC study (Andre T, N Engl J Med, 2004) has demonstrated in patients with stage II/III resected colon cancer (CC) a benefit of oxaliplatin added to 5FU and LV (LV5FU) in 3-year disease-free survival (DFS) and in overal survival (OS). LV5FU and oxaliplatin (FOLFOX4) has also shown a benefit in deficient MisMatch Repair (dMMR) CC pts (Flejou JF, J Clin Oncol 31: 2013;suppl; abstr 3524). We report here 1) Results of MOSAIC after 10 year follow-up, and 2) results in the BRAF evaluable population. Molecular markers and biological targeted therapies in metastatic colorectal cancer: expert opinion and recommendations derived from the 11th ESMO/World Congress on Gastrointestinal Cancer, Barcelona, 2009

The article summarizes the expert discussion and recommendations on the use of molecular markers and of biological targeted therapies in metastatic colorectal cancer (mCRC), as well as a proposed treatment decision strategy for mCRC treatment. The meeting was conducted during the 11th ESMO/World Gastrointestinal Cancer Congress (WGICC) in Barcelona in June 2009. The manuscript describes the outcome of an expert discussion leading to an expert recommendation. The increasing knowledge on clinical and molecular markers and the availability of biological targeted therapies have major implications in the optimal management in mCRC. 21 Suppl 6 vi1 10

The evolving role of oxaliplatin in the management of colorectal cancer

The introduction of oxaliplatin into the chemotherapy of advanced colorectal cancer has substantially increased the frequency and magnitude of clinical response compared with that achieved using 5-FU/leucovorin, and has extended progression-free and overall survival. Research is now in progress on several fronts to determine how oxaliplatin-based therapy can be optimized. A phase III multicentre trial recently compared the efficacy and safety of the FUFOX regimen, based on high dose infusional 5-FU/leucovorin and 50 mg/m2 oxaliplatin given weekly for 4 weeks in a 5-week cycle (n = 123) with the Mayo clinic 5-FU/leucovorin regimen (n = 129) in the first-line therapy of metastatic disease.
