2021

The main goal of this article is to provide evidence in favor of the phonological character of some intonational contrasts in Catalan. First, the article shows that tonal alignment differences with respect to metrically strong positions are crucial for the distinction between a yes-no question and a wh-question (cf. "¿Que l’ha llogada?" vs. "¿Qui l’ha llogada?") and between a command and a request ("¡Vine!" vs. "¡Vine!") in Central Catalan. Second, the article reports the major results from the application of the Categorical Perception Paradigm to a pitch height contrast in Majorcan Catalan (Vanrell 2006a). Experimental resuls show that a small variation in the pitch height of the pretonic …

The beaker phenomenon and the Genomic transformations of Northwest Europe

2018

Bell Beaker pottery spread across western and central Europe beginning around 2750 BCE before disappearing between 2200–1800 BCE. The mechanism of its expansion is a topic of long-standing debate, with support for both cultural diffusion and human migration. We present new genome-wide ancient DNA data from 170 Neolithic, Copper Age and Bronze Age Europeans, including 100 Beaker-associated individuals. In contrast to the Corded Ware Complex, which has previously been identified as arriving in central Europe following migration from the east, we observe limited genetic affinity between Iberian and central European Beaker Complex-associated individuals, and thus exclude migration as a signific…

An exploratory study of two Caco-2 cell models for oral absorption: A report on their within-laboratory and between-laboratory variability, and their…

2010

In 2005, the European Centre for the Validation of Alternative Methods (ECVAM) sponsored a study aimed at evaluating the reproducibility (between-laboratory and within-laboratory variability) and the predictive capacity of two in vitro cellular systems — the Caco-2/ATCC parental cell line and the Caco-2/TC7 clone — for estimating the oral fraction absorbed (Fa) in humans. Two laboratories, both of which had experience with Caco-2 cultures, participated in the study. Ten test chemicals with documented in vivo oral absorption data were selected. Atenolol, cimetidine and propranolol were included as reference compounds for low, medium and high intestinal absorption, respectively. Transport ex…

Use of flow cytometry and confocal microscopy techniques to investigate early CdCl(2)-induced nephrotoxicity in vitro.

2001

CdCl(2) is a well-known toxic compound for the kidney in vivo and in vitro. We report here part of the results of an ECVAM (European Centre for the Validation of Alternative Methods) contract study, aimed at establishing and assessing several flow cytometric and confocal microscopic endpoints for use in an in vitro nephrotoxicity model. Three renal tubule cell lines, OK (opossum, proximal tubule origin), LLC-PK1 (pig, proximal tubule origin) and MDCK (dog, distal tubule origin) were exposed for 1, 5 and 24 h to 25 microM and 100 microM CdCl(2). The results obtained for mitochondrial membrane potential showed a decrease in all the cell lines after 5 h of treatment with both CdCl(2) concentra…

Comparative analysis of eight cytotoxicity assays evaluated within the ACuteTox Project.

2013

A comparative analysis of eight cytotoxicity assays [the 3T3 and normal human keratinocytes Neutral Red Uptake (NRU) assay, the primary rat hepatocytes, human HepG2 and 3T3 MU assay, and the human A.704, SH-SY5Y and HepG2 cells propidium iodide (PI) assay] included in several work packages of the EU Integrated Project ACuteTox, has been carried out. The aim was to evaluate whether cells originating from liver, kidney and brain provided different in vitro acute toxicity results, and the influence of primary liver cells versus cell lines originated from the same tissue. Spearman rank correlation analysis and Hierarchical Cluster Analysis were performed based on the IC50 (50% inhibitory concen…
