We have reviewed the safety and tolerability of β3-adrenoceptor agonists, specifically mirabegron and solabegron, a newly emerging drug class for the treatment of the overactive bladder syndrome. We discuss them mechanistically in the context of expression and other preclinical data.Based on a systematic PubMed search, incidence of overall adverse events, hypertension, dry mouth, and constipation are comparable between mirabegron or solabegron and placebo. Hypertension is the most frequently observed adverse event, but has a similar incidence with mirabegron and placebo. Late-onset or nonclassic hyperandrogenic congenital adrenal hyperplasia (CAH) is an attenuated deficiency of 21-hydroxylase, 3β-ol-hydroxysteroid dehydrogenase or 11β-hydroxylase which presents during childhood or adolescence and leads to an increased secretion of adrenal androgens. Many reviews of the genetic or hormonal characteristics of these syndromes have been published, but relatively little attention has been paid to the pathogenesis and treatment of hirsutism which, in most young women, is the main complaint. For the first time it has been proven that cholest-4-ene-3-one is an endogenous constituent of adrenal tissue. Concentrations of cholestenone in quartered rat adrenals after 3 hours of in vitro incubation were comparable to those of progesterone. However, from specific radioactivities of tissue cholesterol and progesterone and from those of tissue cholestenone and progesterone after incubations with either 4-14C-cholesterol or 4-14C-cholestenone it has been concluded that, at best, cholestenone plays a minimal role as a C27-intermediate of corticosteroid biosynthesis.
Laparoscopy versus open adrenalectomy in patients with solid tumor metastases: results of a multicenter European study

Background: The outcome of adrenalectomy carried out by laparoscopy or open surgery for solid tumor metastases was assessed. Methods: A total of 317 patients with histologically confirmed adrenal metastatic disease collected from 30 centres in Europe underwent adrenalectomy by laparoscopy (n=146) or open laparotomy (n=171). Differences between laparoscopic and open adrenalectomy were assessed by a single Cox analysis for both procedures. Results: The median overall survival was 24.0 [95% confidence interval (CI): 21.4-26.6] months for open adrenalectomy and 45.0 (95% CI: 22.6-67.4) for laparoscopic adrenalectomy (P=0.008). Pētījuma ietvaros tiek analizēta arī juridiskās adreses izpratne Latvijas Republikas tiesību sistēmā un tās regulējuma ietekme uz kapitālsabiedrību pārvaldes institūciju sapulču un sēžu organizēšanu.
