The Sea Urchin sns5 Insulator Protects Retroviral Vectors From Chromosomal Position Effects by Maintaining Active Chromatin Structure
2009
Silencing and position-effect (PE) variegation (PEV), which is due to integration of viral vectors in heterochromatin regions, are considered significant obstacles to obtaining a consistent level of transgene expression in gene therapy. The inclusion of chromatin insulators into vectors has been proposed to counteract this position-dependent variegation of transgene expression. Here, we show that the sea urchin chromatin insulator, sns5, protects a recombinant gamma-retroviral vector from the negative influence of chromatin in erythroid milieu. This element increases the probability of vector expression at different chromosomal integration sites, which reduces both silencing and PEV. By chr…

New emerging potentials for human Wharton's jelly mesenchymal stem cells: immunological features and hepatocyte-like differentiative capacity.
2010
In recent years, human mesenchymal stem cells (MSC) have been extensively studied. Their key characteristics of long-term self-renewal and a capacity to differentiate into diverse mature tissues favour their use in regenerative medicine applications. Stem cells can be found in embryonic and extra-embryonic tissues as well as in adult organs. Several reports indicate that cells of Wharton's jelly (WJ), the main component of umbilical cord extracellular matrix, are multipotent stem cells, expressing markers of bone marrow mesenchymal stem cells (BM-MSC), and giving rise to different cellular types of both connective and nervous tissues. Wharton's jelly mesenchymal stem cells (WJ-MSC) express …
Collagen-induced differential expression of an RNA polymerase subunit by breast cancer cells
2005
It was previously reported that the stroma of ductal infiltrating carcinoma (DIC) of the human breast contains considerable amount of an embryo-foetal collagen type, OF/LB (onco-foetal/laminin-binding), and that adhesion of 8701-BC DIC cells onto OF/LB collagen substrates selectively promotes cell growth, motility, production of extracellular lytic enzymes and invasion "in vitro" if compared with other collagen species. To detect possible transcriptional differences for regulatory proteins following OF/LB collagen-cell interactions, we submitted RNA preparations from 8701-BC cells grown on collagen type I, IV and OF/LB to "differential display"-PCR in the presence of degenerate C(2)H(2) zin…

A method for measuring mitochondrial mass and activity
2007
Mitochondria, responsible for the energy-generating process essential for the cell metabolism, differ for the number, localization and activity in animal cells and tissues in relation to the energetic needs. Using fluorescent probes specific for mitochondria, Mitotracker Green (MTG) and Orange (MTO), and Confocal Laser-Scanning Microscope (CLSM), we elaborated a method to measure in vivo the mitochondrial mass and activity, in sea urchin Paracentrotus lividus eggs and embryos. The analysis of captured images, revealed a variation of mitochondrial distribution and an increase of activity after fertilization.
Polyphenols from halophytes and modified atmosphere packaging improve sensorial and biochemical markers of quality of common dolphinfish (Coryphaena …
2016
Quality and shelf-life of whole and filleted Coryphaena hippurus, stored with modified atmosphere packaging (MAP) and natural antioxidants from halophytes (HAL), were investigated. Fillets were divided into control, simply sealed in trays; MAP, preserved by modified atmosphere (45% CO2, 50% N-2, 5% O-2); and MAP-HAL, pretreated with antioxidants and preserved by MAP. Whole and filleted fish were stored at -1 +/- 0.5 degrees C for 18days. The quality of the samples was analyzed at the time of packaging and after 3, 6, 9, 12, 15, and 18days. The MAP and MAP-HAL groups maintained the best sensorial profile, pH, and drip loss with respect to the untreated fillets. Higher levels of total volatil…

Evaluation of the qualitative properties and consumer perception of marinated sardine Sardina pilchardus: The effect of fucoxanthin addition
2023
With the increasing consumer awareness and demand for healthy food and natural ingredients; there is an urgent need to develop and propose innovative products to industry. In this study, the combined effect of various time of pre-incubation (30, 60 and 90 min) in vinegar and the addition of fucoxanthin (Fux) extracted from the brown alga Dictyopteris polypodiodes to the covering oil of sardine fillets was investigated during 3 months of refrigerated storage at 4◦C. Compared to fresh sardines, biogenic amines (Bas) decreased significantly in all marinated lots (histamine <0.07 mg/100 g, cadaverine <0.0 mg/100 g) and remained far below the regulatory limit up to the end of storage indic…
