WRN protects against topo I but not topo II inhibitors by preventing DNA break formation

The Werner syndrome helicase/3′-exonuclease (WRN) is a major component of the DNA repair and replication machinery. To analyze whether WRN is involved in the repair of topoisomerase-induced DNA damage we utilized U2-OS cells, in which WRN is stably down-regulated (wrn-kd), and the corresponding wild-type cells (wrn-wt). We show that cells not expressing WRN are hypersensitive to the toxic effect of the topoisomerase I inhibitor topotecan, but not to the topoisomerase II inhibitor etoposide. This was shown by mass survival assays, colony formation and induction of apoptosis. Aim: Kawasaki disease (KD) is a systemic vasculitis with unknown etiology. In addition to cardiovascular system involvement, it can also have other multiple organs involved. This study is aimed at investigating the correlation between anti-cardiolipin antibody (ACA)/D dimer/C reactive protein (CRP) and coronary artery lesions (CAL)/multiple-organ lesions in children with KD.Methods: Retrospective analysis was performed in 284 KD/IKD patients from May 2015 to April 2016. Single- and Double-Strand Breaks of Dry DNA Exposed to Protons at Bragg-Peak Energies

International audience; Ultrathin layers (<20 nm) of pBR322 plasmid DNA were deposited onto 2.5 μm thick polyester films and exposed to proton Bragg-peak energies (90–3000 keV) at various fluences. A quantitative analysis of radio-induced DNA damage is reported here in terms of single- and double-strand breaks (SSB and DSB, respectively). The corresponding yields as well as G-values and the cross sections exhibit fairly good agreement with the rare available data, stemming from close experimental conditions, namely, based on α particle irradiation. Flooding damage appraisal can been obtained by interpolating real damage data caused by historical flooding events or accounting the effects of a flood in terms of the depreciation of assets. Most often, the expected damage is evaluated by means of damage functions describing the relationship occurring between the damage and hydraulic characteristics of flood. The essay focuses on the damage caused by the loss of the parental relationship and, in particular, its consequences from a compensation point of view. Taking as its starting point a recent ruling of the Corte di Cassazione, the paper highlights how, in the Italian legal system, this type of damage has different, and significant, consequences for the injured party. In the second part of the essay, it will be seen how, in the English legal system, the so called damage for bereavement has only recently been introduced in the Fatal Accident Act 1976 and how its consequences differ considerably from what happens in the Italian legal system.

On September 1st 1726 Palermo was hit by a violent earthquake. Although it did not bring about the destruction of the city, the earthquake caused widespread and extensive damage to the architectural heritage. The local insti- tutions carried out a series of actions involving representatives of the technical culture. These actions affected the subsequent appearance of the urban pattern and influenced the construction methods of the time. Crossing the data taken from chronicles of the time, from maps produced after the earthquake and above all copious archival documents, the research aims to reconstruct the state in which Palermo was after that tragic event, highlighting a number of aspects. …
