Lymphomas and thyroid: Bridging the gap

2018

The thyroid gland is often involved in the development of neoplastic diseases, including lymphoproliferative disorders. The aim of this paper is to provide a comprehensive overview of the multiple interactions between lymphoma and thyroid. Through an extensive research among the literature, the relationship between lymphomas and thyroid can be established at various levels, and the possible interconnections are here summarized in 5 points: (1) the greater risk of lymphoma development in some thyroid diseases; (2) the primary thyroid lymphoma, with focus on issues related to the diagnosis, differential diagnosis, and treatment; (3) the incidence of thyroid involvement in primitive nodal lymp…

Cancer of the Thyroid

2021

Thyroid cancers are the most frequent endocrine neoplasms whose incidence is globally increasing. Thyroid carcinomas basically derived from follicular (papillary, follicular, anaplastic and poorly differentiated) or parafollicular cells (medullary). The diagnostic evaluation of thyroid cancers is mainly based on neck ultrasonography along with fine needle aspiration. First initial risk evaluation should be carried out postoperatively, while a dynamic risk stratification should be continually assessed to personalize treatment. Whereas combination of surgery, adjuvant radioactive iodine (RAI) ablation, and hormone therapy enables high rates of cure in differentiated tumors (DTC), surgical res…

Immune cells in colorectal cancer: prognostic relevance and therapeutic strategies.

2008

During the last two decades, considerable efforts have been made to improve the prevention, early diagnosis and therapy of colorectal cancer by gaining enhanced insights into disease-specific pathogenesis. Along these lines, tumor-infiltrating immune cells turned out to be critical indicators for an efficient antitumor immune response and the number and type of tumor-infiltrating immune cells determined the resulting tumor prognosis. This review aims to describe the prognostic relevance of the different subsets of tumor-infiltrating immune cells and highlights their specific function in the complex process of immune system-mediated rejection of colorectal cancer cells. Considering the clini…

The Role of HSP70 in the Diagnosis of HCC

2015

ABSTRACT: Hepatocellular carcinoma (HCC) is a common tumor with a fatal clinical outcome if not diagnosed at an early stage. To date there is still a lack of reliable tumor biomarkers to detect asymptomatic precursor lesions in early HCC. Conventional pathological analysis also often fails to achieve sufficient sensitivity and specificity to diagnose early HCC. Genetic, proteomic, immunohistochemical, and liquid biopsy analysis indicate a role for members of the heat-shock protein 70 (HSP70) family as potential tumor-specific markers for HCC. The aim of this review is to revisit the existing literature and to specifically explore HSP70 as a molecular tumor biomarker for the detection of HCC…

Detection and Differential Diagnosis of Prekallikrein Deficiency: Genetic Study of New Families and Systematic Review of the Literature

2018

Abstract Introduction. Prekallikrein (PK) and high-molecular-weight kininogen (HK) deficiencies are ultra-rare, autosomal-recessive defects of the contact system caused by biallelic mutations in the KLKB1 and KNG1 genes, respectively. Since affected subjects do not manifest a bleeding phenotype, a correct diagnosis is essential to prevent the administration of prohemostatic agents or plasma and to avoid delay of surgery. We describe a new case of PK deficiency identified at UMC Mainz. In addition, we performed a systematic review of the literature in order to i) collect blood material for genetic studies of reported PK deficient cases lacking this information, and ii) perform a comprehensiv…

Head and Neck Lymphomas in HIV Patients: a Clinical Perspective

2017

Introduction Because of the many HIV-related malignancies, the diagnosis and treatment of lymphoma in patients infected with human immunodeficiency virus are challenging. Objective Here, we review current knowledge of the pathogenesis, epidemiology, symptomatology, diagnosis, and treatment of head and neck lymphomas in HIV patients from a clinical perspective. Data Synthesis Although Hodgkin's lymphoma is not an AIDS-defining neoplasm, its prevalence is ten times higher in HIV patients than in the general population. NHL is the second most common malignancy in HIV patients, after Kaposi's sarcoma. In this group of patients, NHL is characterized by rapid progression, frequent extranodal inv…

Early diagnosis, clinical management, and follow-up of cardiovascular events with ponatinib

2020

Chronic myeloid leukemia (CML) is a myeloproliferative disorder characterized by neoplastic transformation of pluripotent cells due to a typical cytogenetic and molecular mutation known as Philadelphia (Ph) chromosome. In 2001, the introduction of the tyrosine kinasis inhibitor (TKI) imatinib as a therapeutic strategy for CML with PH chromosome mutation represented an important step towards treatment of these patients, and nowadays, this drug represents the gold therapeutic standard in this clinical setting. A second generation of TKIs (dasatinib, nilotinib, and bosutinib) showed an effective action in all patients with mutations resistant to imatinib. Ponatinib is a third-generation TKI an…
