Abstract Liquid-liquid extraction (LLE), solid phase extraction (SPE) and microwave-assisted extraction are evaluated for Fourier transform infrared (FTIR) spectrometric determination of oil and grease in polluted waters. A fast microwave-assisted quantitative procedure is proposed for the total extraction of oil and grease into CCl 4 from water samples, with an irradiation time of 1 min at an exit power of 520 W using closed PTFE vessels. A simple and flexible system, employing a column switching technique, has been designed to allow the analysis of peptides and proteins smaller than 15 kDa by molecular weight in filtered urine samples by performing a direct on-column injection utilising simultaneous sample clean-up and trace enrichment. The positively charged peptides and small proteins in the sample are attracted to the inner, negatively charged pore structure of the RAM-SCX column while the larger proteins and uncharged or negatively charged compounds are excluded. A simple and sensitive method has been developed for the liquid chromatographic determination of short-chain aliphatic amines in water. Analytes are retained in solid-phase extraction (SPE) cartridges, and then derivatized by drawing an aliquot of the fluorogeneic reagent 9-fluorenylmethyl chloroformate (FMOC) through the cartridges. After a certain reaction time the derivatives formed are desorbed with acetonitrile. The extraction behaviour of 10 μg samples of five pesticides and some related compounds from glass wool with supercritical CO2 has been investigated under several conditions (10 MPa, 20 MPa extraction pressure, 313 K, dry and water saturated CO2). The extraction fluid was decompressed over a line of little columns, filled with 3μ Si60 or RP18, and the amount of deposited material was analyzed by HPLC for each of these columns. Determination of triazines and organophosphorus pesticides in water samples using solid-phase extraction.

Abstract Octadecyl (C 18 )-bonded porous silica was evaluated for the extraction of triazine and organophosphorus pesticides from natural water. The extraction results showed an effective performance when 1 l of water was passed through small glass columns containing 500 mg of 50–100-μm C 18 bonded porous silica. The absorbed compounds were removed with ethyl acetate, evaporated to 200 μl and determined by gas chromatography. The overall average recoveries were greater than 85% except for dimethoate and trichlorfon. Application of this procedure to the analysis of natural water samples gave results that agree well with those obtained by solvent extraction methods.

Abstract Dansylation of amines (primaries and secondaries) using C18 solid-phase supports is described. The time of analysis and the handing sample have been markedly improved with respect to those required for solution dansylation. The dansylated amine derivates were injected onto the liquid chromatography system and fluorescence detected. The sensitivity and selectivity is better than the other dansylation procedures described in the literature and also better than other derivatization reagents such 1,2-napthoquinone 4-sulfonate (NQS) or 3,5-dinitrobenzoyl chloride (3,5-DNB). A rapid and cost-efficient DMSO-based method for isolating DNA from cultured lichen photobionts

We have developed a simple and fast procedure for the purification of PCR-quality DNA from cultured lichen photobionts. This new one-step method uses the solvent dimethyl sulfoxide (DMSO) combined with heat treatment to chemically breakdown algal and plant tissues. The DMSO-DNA extracts may be directly precipitated and purified by standard techniques in a total time of approximately 30 min. Compared to other DNA extraction protocols, the DMSO-based method suppresses the need for liquid nitrogen, any extraction buffer, grinding, phenol, and long incubation or centrifugation steps, thereby considerably reducing the possibility of contamination. Graphitized Carbon Black (GCB) extractive cartridges are evaluated for on-line coupling with a C8 analytical column to determine eleven carbamates and one carboximide pesticide from spiked deionized water at the 1.2 μg/L level. Several experiments were carried out to ascertain whether GCB saturation, pesticide degradation on the surface, existence of by-pass channels, mobility among the bulk cartridge, or strong retention on the surface interfere with the determination of pesticides.
