MDMA Administration and Heat Shock Proteins Response: Foreseeing a Molecular Link
2010
Molecular and cellular mechanisms of MDMA-induced toxicity have been extensively studied in a number of experimental models. Nevertheless, only few studies investigated the involvement of HSPs ("molecular chaperones") in MDMA organs toxicity. In the present minireview we highlight this subject analysing the results of these studies conducted especially on brain tissue. Despite of it seems obvious that HSPs overexpression is a protective reaction against MDMA treatment, the molecular mechanisms for exerting their action are far to be undiscovered. At the same time, we need of comprehensive studies concerning the whole range of Hsps/chaperones expressions in all organs after acute and chronic…

The Role of HSP70 in the Diagnosis of HCC
2015
ABSTRACT: Hepatocellular carcinoma (HCC) is a common tumor with a fatal clinical outcome if not diagnosed at an early stage. To date there is still a lack of reliable tumor biomarkers to detect asymptomatic precursor lesions in early HCC. Conventional pathological analysis also often fails to achieve sufficient sensitivity and specificity to diagnose early HCC. Genetic, proteomic, immunohistochemical, and liquid biopsy analysis indicate a role for members of the heat-shock protein 70 (HSP70) family as potential tumor-specific markers for HCC. The aim of this review is to revisit the existing literature and to specifically explore HSP70 as a molecular tumor biomarker for the detection of HCC…
Bacillus thuringiensis treatment modulate the HSP70 expression in larva and adult brain of Rhynchophorus ferrugineus
2013
To study the pathogen-host relationship, we used the model of the entomopathogenic bacterium Bacillus thuringiensis (Bt) and Rhynchophorus ferrugineus, a quarantine pest that attacks palm trees. In particular, we focused on the Bt stress-induced infections. We studied the effect of Bt on larval and adult growth, and on the expression of the heat shock proteins (HSPs), rapidly synthesized in the cell after exposure to stress including pathogens. Bt has negative effects on larval and adult growth, on total hemocytes counts and on the hemocyte type. HSP70 was evaluated in the supernatant of the brain lysate obtained from larvae and adults fed with sublethal doses of Bt. HSP70 expression was mo…
Bacillus thuringiensis treatment alters larval growth, hemocytes and modulation of Hsp70 in Rhynchophorus ferrugineus
2011
To study the pathogen-host relationship, we used the model of the entomopathogenic bacterium Bacillus thuringiensis (Bt) and Rhynchophorus ferrugineus, a quarantine pest that attacks the palm trees. In particular, we focused on the Bt stress-induced infections. We studied the effect of Bt on larval growth, on hemocytes and on the expression of the heat shock proteins (Hsp70). HSPs are rapidly synthesized in the cells after a stress exposition including pathogens. The Hsp70 was evaluated in the supernatant of the hemocyte lysate (HLS) obtained from larvae fed with Bt. This is the first time that the presence of Hsp70 has been recorded in R. ferrugineus. Bt has negative effects on larval grow…

Mouse mesoangioblasts release Hsp70 in a controlled manner through membrane vesicle shedding
2009
Mesoangioblaststs (Mabs) are mesodermal stem cells associated with vessels which can differentiate into different mesoderm cell types. They have the property to cross endothelial barrier and when injected into circulation they localize into damaged tissues. A6 cells, a clone of mouse Mabs, express Hsp70 protein in physiological conditions and this synthesis may be required to answer to several intra- and/or extra-cellular needs. It was found that Hsp70 may be released outside from several type of cells, but its role remains still undefined. In order to clarify the reason of Hsp70 constitutive expression, we prepared several A6 clones with different levels of Hsp70 expression by stable RNA i…

Constitutive hsp70 is essential to mitosis during early cleavage of Paracentrotus lividus embryos: The blockage of constitutive hsp70 impairs mitosis
1999
Localization of constitutive hsp70 in eggs and early embryos of sea urchin Paracentrotus lividus is shown by means of in situ immunostaining. An accumulation of this protein is shown in the mitotic structures (asters, spindles and centrosomes). Microinjection of anti-hsp70 antibodies into eggs causes impairment of formation of mitotic structures and of cell division. This impairment goes from a complete mitotic block, to irregular mitotic apparatus formation with irregular cleavage, depending upon the antibody concentration. The localization of hsp70 after antibody microinjection is also described. Blockage of mitotic apparatus formation by nocodazole also blocks the concentration of hsp70 …
