Moderated Redactable Blockchains: A Definitional Framework with an Efficient Construct

Blockchain is a multiparty protocol to reach agreement on the order of events, and to record them consistently and immutably without centralized trust. In some cases, however, the blockchain can benefit from some controlled mutability. Examples include removing private information or unlawful content, and correcting protocol vulnerabilities which would otherwise require a hard fork. Two approaches to control the mutability are: moderation, where one or more designated administrators can use their private keys to approve a redaction, and voting, where miners can vote to endorse a suggested redaction. Abstract Detection and mapping of modified nucleotides in RNAs is a difficult and laborious task. Several physico-chemical approaches based on differential properties of modified nucleotides can be used, however, most of these methods do not allow high-throughput analysis. Here we describe in details a method for mapping of rather common 1-methyladenosine (m1A) residues using high-throughput next generation sequencing (NGS). Directional high-throughput sequencing of RNAs without gene-specific primers.

Ribosomal RNA analysis is a useful tool for characterization of microbial communities. However, the lack of broad-range primers has hampered the simultaneous analysis of eukaryotic and prokaryotic members by amplicon sequencing. We present a complete workflow for directional, primer-independent sequencing of size-selected small subunit ribosomal RNA fragments. The library preparation protocol includes gel extraction of the target RNA, ligation of an RNA oligo to the 5′-end of the target, and cDNA synthesis with a tailed random-hexamer primer and further barcoding.
