Influence of metal ions on thermal aggregation of bovine serum albumin: aggregation kinetics and structural changes

2009

Metal ions are implicated in protein aggregation processes of several neurodegenerative pathologies. In this work the effects of Cu(II) and Zn(II) ions on heat-induced structural modifications of bovine serum albumin (BSA) were studied, with the aim of delineating the role of these ions in the early stages of proteins aggregation kinetics. A joint application of different techniques was used. The aggregate growth was followed by dynamic light scattering measurements, whereas the conformational changes occurring in the protein structure were monitored by Raman and IR spectroscopy. Both in absence and in presence of metal ions, heating treatment gave rise to b-structures to the detriment of a…

Inulin-Ethylenediamine Coated SPIONs Magnetoplexes: A Promising Tool for Improving siRNA Delivery.

2015

An inulin based polycation (Inu-EDA) has been synthesized by the grafting of ethylenediamine molecules onto inulin backbone. The obtained inulin copolymer has been though to coat SPIONs (IC-SPIONs) and obtain stable magnetoplexes by complexation of IC-SPIONs with a model duplexed siRNA, for improving oligonucleotide transfection efficiency. The physical-chemical characteristics of IC-SPIONs and IC-SPIONs/siRNA magnetoplexes have been investigated by scanning and transmission electron microscopies, dynamic light scattering, FT-IR and qualitative surface elementary analysis. Cell compatibility and internalization in vitro of IC-SPIONs have been evaluated by MTS and fluorescence microscopy resp…

Two-Step Nucleation Process of Calcium Silicate Hydrate, the Nanobrick of Cement

2018

Despite a millennial history and the ubiquitous presence of cement in everyday life, the molecular processes underlying its hydration behavior, like the formation of calcium–silicate–hydrate (C–S–H), the binding phase of concrete, are mostly unexplored. Using time-resolved potentiometry and turbidimetry combined with dynamic light scattering, small-angle X-ray scattering, and cryo-TEM, we demonstrate C–S–H formation to proceed via a complex two-step pathway. In the first step, amorphous and dispersed spheroids are formed, whose composition is depleted in calcium compared to C–S–H and charge compensated with sodium. In the second step, these amorphous spheroids crystallize to tobermorite-typ…

From Spherical Mesopores to Worm-Shaped Mesopores : Morphology Transition in Titania–Polystyrene-b-poly(ethylene oxide) Composite Films with Increasi…

2013

A morphology transition from spherical mesopores to worm-shaped mesopores within titania block copolymer composite thin films has been observed by varying the sol–gel reaction time from 40 min to 48 h in the four-component templating system of polystyrene-$\mathit{b}$-poly(ethylene oxide) (PS-$\mathit{b}$-PEO), 1,4-dioxane, concentrated HCl, and titanium tetraisopropoxide (TTIP) with a PS-$\mathit{b}$-PEO mass concentration of 0.25 wt.-%. The impact of the sol–gel reaction time on the local structure, long-range lateral structure, and vertical structure of the as-prepared, calcined, and UV-degraded thin films as well as the structural changes in solution have been systematically investigate…

Supramolecular assembly using helical peptides

1997

Abstract We investigated supramolecular assemblies of various hydrophobic helical peptides. The assemblies were formed at the air/water interface or in aqueous medium. The hexadecapeptide, Boc-(Ala-Aib)8-OMe (BA16M), was reported to take α-helical structure by X-ray analysis. Several derivatives were prepared, which have the repeating sequence of Ala-Aib, Lys(Z)-Aib or Leu-Aib, or have the terminal chemically modified. CD spectra of the peptides indicated helical conformation in ethanol solution. The surface pressure-area isotherms of the peptide monolayers showed an inflection at the surface area corresponding to the cross section along the helix axis, and the monolayers were collapsed by …

Aggregation versus Biological Activity in Gold(I) Complexes. An Unexplored Concept

2021

The aggregation process of a series of mono- and dinuclear gold(I) complexes containing a 4-ethynylaniline ligand and a phosphane at the second coordination position (PR3-Au-C≡CC6H4-NH2, complexes 1-5, and (diphos)(Au-C≡CC6H4-NH2)2, complexes 6-8), whose biological activity was previously studied by us, has been carefully analyzed through absorption, emission, and NMR spectroscopy, together with dynamic light scattering and small-angle X-ray scattering. These experiments allow us to retrieve information about how the compounds enter the cells. It was observed that all compounds present aggregation in fresh solutions, before biological treatment, and thus they must be entering the cells as a…

A Multipurpose Instrument To Measure the Vitreous Properties of Charged Colloidal Solids

2001

We present a new high-precision light-scattering setup to study the properties of colloidal solids. It combines static light scattering, dynamic light scattering, and torsional resonance spectroscopy with a flexible and reliable preparational procedure. All three experiments can be performed quasi-simultaneously on the same mechanically undisturbed sample. Thus, unequivocal identification of glassy behavior in a comprehensive characterization dependent on interaction parameters becomes possible. A detailed description of the mechanics is given. We thoroughly tested the new apparatus on dilute colloidal samples and against commercial reference instruments. We performed a first systematic ser…

"Sweet Chemistry": a Green Way for Obtaining Selenium Nanoparticles Active against Cancer Cells

2017

We present an environment friendly synthesis of selenium nanoparticles and the study of their cytotoxic activity against uterine sarcoma cancer and fibroblasts cells. Amorphous selenium (a-SeNPs) and trigonal selenium (t-SeNPs) were synthesized using D-fructose as the reducing agent and characterized by high-resolution transmission electron microscopy (HRTEM), energy-dispersive X-ray spectroscopy (EDX), powder X-ray diffraction analysis (XRD), inductively coupled plasma optical emission spectrometry (ICP OES), dynamic light scattering (DLS) to obtain zeta potential values and cyclic voltammetry (CV). Particularly, a-SeNPs presented high toxicity toward the resistant cancer cell line MES-SA/…

Miniaturized liquid chromatography coupled on-line to in-tube solid-phase microextraction for characterization of metallic nanoparticles using plasmo…

2018

Abstract This tutorial aims at providing guidelines for analyzing metallic nanoparticles (NPs) and their dispersions by using methods based on miniaturized liquid chromatography with diode array detection (MinLC-DAD) and coupled on-line to in-tube solid-phase microextraction (IT-SPME). Some practical advice and considerations are given for obtaining reliable results. In addition, this work outlines the potential applications that set these methodologies apart from microscopy-related techniques, dynamic light scattering, single particle ICP-MS, capillary electrophoresis, field-flow fractionation and other chromatographic configurations, which are discussed and mainly seek to accomplish size …

Role of the co-surfactant nature in soybean w/o microemulsions.

2008

Abstract The influence of the co-surfactant on physicochemical properties of w/o soybean oil microemulsions (MEs) has been studied. In spite of the similarity in phase diagrams, the MEs display remarkable differences when examined by electrical conductivity, dynamic light scattering (DLS), small angle X-ray scattering (SAXS) and linear voltammetry. When different short-chain alcohols were employed as co-surfactants, together with sodium dodecyl sulfate (SDS) as surfactant, the DLS results indicated the systems to be monodisperse. Both the electrical conductivity of the MEs and the hydrodynamic radii of the droplets (RH) increased with water content while RH diminished as temperature increas…
