Lipidomic profiling identifies signatures of metabolic risk

Background: Metabolic syndrome (MetS), the clustering of metabolic risk factors, is associated with cardiovascular disease risk. We sought to determine if dysregulation of the lipidome may contribute to metabolic risk factors. Methods: We measured 154 circulating lipid species in 658 participants from the Framingham Heart Study (FHS) using liquid chromatography-tandem mass spectrometry and tested for associations with obesity, dysglycemia, and dyslipidemia. Independent external validation was sought in three independent cohorts. Fabry disease (FD) is an inherited metabolic disorder caused by partial or full inactivation of the lysosomal hydrolaseα-galactosidase A (α-GAL). The impairment ofα-GAL results in the accumulation of undegraded glycosphingolipids in lysosomes and subsequent cell and microvascular dysfunctions. This study reports the clinical, biochemical, and molecular characterization of 15 members of the same family. Eight members showed the exonic mutation M51I in the GLA gene, a disease-causing mutation associated with the atypical phenotype. Abstract We report a novel heteroplasmic point mutation G8299A in the gene for mitochondrial tRNA Lys in a patient with progressive external ophthalmoplegia complicated by recurrent respiratory insufficiency. Biochemical analysis of respiratory chain complexes in muscle homogenate showed a combined complex I and IV deficiency. The transition does not represent a known neutral polymorphism and affects a position in the tRNA acceptor stem which is conserved in primates, leading to a destabilization of this functionally important domain. CNS myelination by oligodendrocytes requires directed transport of myelin membrane components and a timely and spatially controlled membrane expansion. In this study, we show the functional involvement of the R-solubleN-ethylmaleimide-sensitive factor attachment protein receptor (R-SNARE) proteins VAMP3/cellubrevin and VAMP7/TI-VAMP in myelin membrane trafficking. Evapotranspiration Estimation with the S-SEBI Method from Landsat 8 Data against Lysimeter Measurements at the Barrax Site, Spain

Evapotranspiration (ET) is a variable of the climatic system and hydrological cycle that plays an important role in biosphere–atmosphere–hydrosphere interactions. In this paper, remote sensing-based ET estimates with the simplified surface energy balance index (S-SEBI) model using Landsat 8 data were compared with in situ lysimeter measurements for different land covers (Grass, Wheat, Barley, and Vineyard) at the Barrax site, Spain, for the period 2014–2018. Daily estimates produced superior performance than hourly estimates in all the land covers, with an average difference of 12% and 15% for daily and hourly ET estimates, respectively. This study shows that high methylmercury concentrations are cytotoxic for Dicentrarchus labrax leucocytes, whereas subletal concentrations affect leucocyte phagocytosis and cells morphology in a dose dependent fashion. Although the serum hemoagglutinating activity was not inhibited by the metal, the activity of purified serum F-lectin fraction and mucus was significantly decreased by relevant methylmercury concentrations.

Summary Using a combination of advanced RNA synthesis techniques and single molecule spectroscopy, the deconvolution of individual contributions of posttranscriptional modifications to the overall folding and stabilization of human mitochondrial tRNA Lys is described. An unexpected destabilizing effect of two pseudouridines on the native tRNA folding was evidenced.
