Fast UPLC/PDA determination of squalene in Sicilian P.D.O. pistachio from Bronte: Optimization of oil extraction method and analytical characterizati…
2017
Abstract A fast reversed-phase UPLC method was developed for squalene determination in Sicilian pistachio samples that entry in the European register of the products with P.D.O. In the present study the SPE procedure was optimized for the squalene extraction prior to the UPLC/PDA analysis. The precision of the full analytical procedure was satisfactory and the mean recoveries were 92.8 ± 0.3% and 96.6 ± 0.1% for 25 and 50 mg L−1 level of addition, respectively. Selected chromatographic conditions allowed a very fast squalene determination; in fact it was well separated in ∼0.54 min with good resolution. Squalene was detected in all the pistachio samples analyzed and the levels ranged from 5…
Screening of potent phytochemical inhibitors against SARS-CoV-2 protease and its two Asian mutants
2021
Abstract Background COVID-19, declared a pandemic in March 2020 by the World Health Organization is caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). The virus has already killed more than 2.3 million people worldwide. Object The principal intent of this work was to investigate lead compounds by screening natural product library (NPASS) for possible treatment of COVID-19. Methods Pharmacophore features were used to screen a large database to get a small dataset for structure-based virtual screening of natural product compounds. In the structure-based screening, molecular docking was performed to find a potent inhibitor molecule against the main protease (Mpro) of SARS-…
Olaratumab: PDGFR-α inhibition as a novel tool in the treatment of advanced soft tissue sarcomas
2017
Advanced soft tissue sarcomas are aggressive cancers with limited therapeutic options. Recently, inhibition of platelet-derived growth factor receptor (PDGFR)-α by the monoclonal antibody olaratumab showed promising clinical activity. If confirmed, this would be one of the first examples of targeted therapy effective in advanced soft tissue sarcomas therapy independently of the histologic subtype. Here, we reviewed the biology of the PDGF/PDGFR axis, particularly focusing on its role in cancer, and then we discussed on the effects of PDGFR-α inhibition in the therapy of advanced soft tissue sarcomas.
Safety and efficacy of anti-PD-1 inhibitor ABBV-181 in lung and head and neck carcinoma
2019
Abstract Background ABBV-181 is a humanized anti-PD1 monoclonal antibody; dose finding and early safety, PK and pharmacodynamic data have been reported (ESMO18). This report summarizes data from the head and neck squamous cell carcinoma (HNSCC) and non-small cell lung cancer (NSCLC) expansion cohorts in the Ph1 FIH study (NCT03000257). Methods Patients (pts) with previously treated, advanced HNSCC and NSCLC received ABBV-181 IV, 250 mg Q2W or 500 mg Q4W to progression. Response was assessed by RECIST v1.1 and iRECIST. Results As of April 2019, 81 pts were dosed. Table . 1288P n (%) HNSCC n = 41 NSCLC n = 40 Median days on treatment, range 72, 1–407 71, 1–421 Dose: 250 mg/500 mg 31/10 19/21 …
Immunoistochemical expression of PD-1 and PD-L1 in bone marrow biopsies of patients with acute myeloid leukemia
2020
Background. Haematological and non-haematological malignancies are able to escape the host immune by the capacity to hijack the immune check-points. Several immune check-point molecules are known, such as T cell immunoglobulin mucin-3 (TIM-3), cytotoxic T-cell antigen-4 (CTLA-4), programmed death-1 (PD-1) with its ligand PD-L1 and others.1 The function of these immune check-points is to prevent the damage resulting from an excessive activation of the immune response in the setting of chronic antigenic stimulation, thus leading to autoimmune phenomena, as proved in knock-out mice models. PD-1 is normally present on activated T lymphocytes membrane, acting as a negative costimulatory receptor…
A continuous time tug-of-war game for parabolic $p(x,t)$-Laplace type equations
2019
We formulate a stochastic differential game in continuous time that represents the unique viscosity solution to a terminal value problem for a parabolic partial differential equation involving the normalized $p(x,t)$-Laplace operator. Our game is formulated in a way that covers the full range $1<p(x,t)<\infty$. Furthermore, we prove the uniqueness of viscosity solutions to our equation in the whole space under suitable assumptions.
Observation of $Z$ production in proton-lead collisions at LHCb
2014
The first observation of $Z$ boson production in proton-lead collisions at a centre-of-mass energy per proton-nucleon pair of $\sqrt{s_{NN}}=5~\text{TeV}$ is presented. The data sample corresponds to an integrated luminosity of $1.6~\text{nb}^{-1}$ collected with the LHCb detector. The $Z$ candidates are reconstructed from pairs of oppositely charged muons with pseudorapidities between 2.0 and 4.5 and transverse momenta above $20~\text{GeV}/c$. The invariant dimuon mass is restricted to the range $60-120~\text{GeV}/c^2$. The $Z$ production cross-section is measured to be \begin{eqnarray*} _{Z\to^+^-}(\text{fwd})&=&13.5^{+5.4}_{-4.0}\text{(stat.)}\pm1.2\text{(syst.)}~\text{nb} …
Sharp capacity estimates for annuli in weighted R^n and in metric spaces
2017
We obtain estimates for the nonlinear variational capacity of annuli in weighted R^n and in metric spaces. We introduce four different (pointwise) exponent sets, show that they all play fundamental roles for capacity estimates, and also demonstrate that whether an end point of an exponent set is attained or not is important. As a consequence of our estimates we obtain, for instance, criteria for points to have zero (resp. positive) capacity. Our discussion holds in rather general metric spaces, including Carnot groups and many manifolds, but it is just as relevant on weighted R^n. Indeed, to illustrate the sharpness of our estimates, we give several examples of radially weighted R^n, which …
Minimality via second variation for microphase separation of diblock copolymer melts
2017
Abstract We consider a non-local isoperimetric problem arising as the sharp interface limit of the Ohta–Kawasaki free energy introduced to model microphase separation of diblock copolymers. We perform a second order variational analysis that allows us to provide a quantitative second order minimality condition. We show that critical configurations with positive second variation are indeed strict local minimizers of the problem. Moreover, we provide, via a suitable quantitative inequality of isoperimetric type, an estimate of the deviation from minimality for configurations close to the minimum in the L 1 {L^{1}} -topology.
