Gene regulatory networks elucidating Huanglongbing disease mechanisms

Next-generation sequencing was exploited to gain deeper insight into the response to infection by Candidatus liberibacter asiaticus (CaLas), especially the immune disregulation and metabolic dysfunction caused by source-sink disruption. Previous fruit transcriptome data were compared with additional RNA-Seq data in three tissues: immature fruit, and young and mature leaves. Four categories of orchard trees were studied: symptomatic, asymptomatic, apparently healthy, and healthy. Dynamical Models of Interrelation in a Class of Artificial Networks

The system of ordinary differential equations that models a type of artificial networks is considered. The system consists of a sigmoidal function that depends on linear combinations of the arguments minus the linear part. The linear combinations of the arguments are described by the regulatory matrix W. For the three-dimensional cases, several types of matrices W are considered and the behavior of solutions of the system is analyzed. The attractive sets are constructed for most cases. The illustrative examples are provided. The list of references consists of 12 items.

Assessing the impact of copy number variants on miRNA genes in autism by Monte Carlo simulation.

Autism Spectrum Disorders (ASDs) are childhood neurodevelopmental disorders with complex genetic origins. Previous studies have investigated the role of de novo Copy Number Variants (CNVs) and microRNAs as important but distinct etiological factors in ASD. We developed a novel computational procedure to assess the potential pathogenic role of microRNA genes overlapping de novo CNVs in ASD patients. Here we show that for chromosomes # 1, 2 and 22 the actual number of miRNA loci affected by de novo CNVs in patients was found significantly higher than that estimated by Monte Carlo simulation of random CNV events. MAP kinase p38 and its relation to T cell anergy and suppressor function of regulatory T cells

Diverse regulatory T cell populations (Treg) are important for the control of self tolerance and immune homeostasis. These include naturally occurring CD4+CD25+ Treg (nTreg) and induced Treg (iTreg). Tolerogenic dendritic cells, modulated by IL-10, are able to convert peripheral T cells into iTreg. These are anergic and characterized by a G(1) cell cycle arrest, dependent on elevated levels of the cdk inhibitor p27(Kip1). A closer look at the cholesterol sensor

Abstract Transport of the sterol regulatory element-binding protein (SREBP) cleavage-activating protein (SCAP)–SREBP complex from the endoplasmic reticulum (ER) to the Golgi is the central event mediating the cholesterol-feedback process in mammalian cells. A conformational change in SCAP is a crucial step; when cholesterol levels are high, the conformation of SCAP enables the SCAP–SREBP complex to associate with an insulin-induced gene (INSIG) retention protein in the ER. By contrast, when cholesterol levels are low, SCAP switches to a conformation that enables the dissociation of the retention protein and the association of SCAP–SREBP with COP II vesicles.

Objective— Liver X receptors (LXRs) modulate cholesterol and fatty acid homeostasis as well as inflammation. This study aims to decipher the role of LXRs in the regulation of polyunsaturated fatty acid (PUFA) synthesis in macrophages in the context of atherosclerosis. Approach and Results— Transcriptomic analysis in human monocytes and macrophages was used to identify putative LXR target genes among enzymes involved in PUFA biosynthesis.
