Nuclear calcium signaling by inositol trisphosphate in GH3 pituitary cells
2008
It has been proposed that nuclear and cytosolic Ca2+ ([Ca2+]N and [Ca2+]C) may be regulated independently. We address here the issue of whether inositol trisphosphate (IP3) can, bypassing changes of [Ca2+]C, produce direct release of Ca2+ into the nucleoplasm. We have used targeted aequorins to selectively measure and compare the changes in [Ca2+]C and [Ca2+]N induced by IP3 in GH3 pituitary cells. Heparin, an IP3 inhibitor that does not permeate the nuclear pores, abolished the [Ca2+]C peaks but inhibited only partly the [Ca2+]N peaks. The permeant inhibitor 2-aminoethoxy-diphenyl-borate (2-APB) blocked both responses. Removal of ATP also inhibited more strongly the [Ca2+]C than [Ca2+]N pe…
Tubuloreticular structures (TRS) and cylindric confronting cisternae (CCC) in childhood dermatomyositis.
1989
Tubuloreticular structures (TRS) and cylindric confronting cisternae (CCC) have been observed in circulating lymphocytes and in the muscle of six children with dermatomyositis. The presence of TRS was seen in all cases investigated, the number of CCC increased in various cells with the severity of the disease. Extensive formation of TRS and CCC in childhood dermatomyositis probably reflects local or systemic alpha-interferon production and suggests that some viral factor is responsible for the disease.
Experimental study of species invasion : early population dynamics and role of disturbance in invasion success
2020
Much of our understanding of natural invasions is retrospective, based on data acquired after invaders become established. As a consequence, we know little about the characteristics of the early population growth and habitat use of the invaders during establishment. Here we report on experimental introductions of guppies into natural streams in which we conducted monthly censuses of each population. Two of the four introductions were in streams with thinned canopies, which mimics a common form of habitat disturbance. We conducted similar censuses of natural populations to characterize natural population densities and generate a null distribution against which we could test a priori hypothes…
Efficient finance regulator as supporter for financial stability of inhabitants
2020
Recent developments in banking sector in Latvia especially related with PNB Bank where bank accounts of many retired people were located leads to requirements for proficiency of Finance Regulator which has to be efficiently financed, stable and professional. In different countries Finance Regulators have different models of their financing. Aim of the study is to investigate different approaches for Finance Regulator financing and analyse expert views on possible development of Finance Regulator financing model in Latvia. Research methods applied: scientific publications and previous conducted research results analysis and expert survey for possible financing model changes in Latvia. In exp…
Impact of
2018
Drosophila melanogaster has been for over a century the model of choice of several neurobiologists to decipher the formation and development of the nervous system as well as to mirror the pathophysiological conditions of many human neurodegenerative diseases. The rare disease Friedreich’s ataxia (FRDA) is not an exception. Since the isolation of the responsible gene more than two decades ago, the analysis of the fly orthologue has proven to be an excellent avenue to understand the development and progression of the disease, to unravel pivotal mechanisms underpinning the pathology and to identify genes and molecules that might well be either disease biomarkers or promising targets for therap…
Exploring Determinants of Early Retirement Among Saudi Medical Staff
2021
The aim of this research was to explore the relationship between the push, pull, anti-push, and anti-pull factors vs. early retirement intention among Saudi medical staff, and to investigate whether there are gender differences in the early retirement intention. To this end, we designed a correlational and cross-sectional study, for which data were collected through an online survey. A total of 680 responses were gathered, of which 221 valid responses constituted the final sample for the analysis. Logistics regression was used to test the hypotheses of the study. The results showed that approximately 58% of the respondents indicated early retirement intention. The significant factors in pre…
AUTOMATIC RETINA EXUDATES SEGMENTATION WITHOUT A MANUALLY LABELLED TRAINING SET
2011
International audience; Diabetic macular edema (DME) is a common vision threatening complication of diabetic retinopathy. In a large scale screening environment DME can be assessed by detecting exudates (a type of bright lesions) in fundus images. In this work, two new methods for the detection of exudates are presented. The methods do not require a lesion training set so the need to ground-truth data is avoided with significant time savings and independence from human error. We evaluate our algorithm with a new publicly available dataset from various ethnic groups and levels of DME. Also, we compare our results with two recent exudate segmentation algorithms on the same dataset. In all of …
Suppression and Replacement Gene Therapy for Autosomal Dominant Disease in a Murine Model of Dominant Retinitis Pigmentosa
2011
For dominantly inherited disorders development of gene therapies, targeting the primary genetic lesion has been impeded by mutational heterogeneity. An example is rhodopsin-linked autosomal dominant retinitis pigmentosa with over 150 mutations in the rhodopsin gene. Validation of a mutation-independent suppression and replacement gene therapy for this disorder has been undertaken. The therapy provides a means of correcting the genetic defect in a mutation-independent manner thereby circumventing the mutational diversity. Separate adeno-associated virus (AAV) vectors were used to deliver an RNA interference (RNAi)-based rhodopsin suppressor and a codon-modified rhodopsin replacement gene res…
24S-hydroxycholesterol and cholesterol-24S-hydroxylase (CYP46A1) in the retina: from cholesterol homeostasis to pathophysiology of glaucoma.
2011
http://www.sciencedirect.com/; International audience; Free cholesterol is the predominant form of cholesterol in the neural retina. The vertebrate neural retina exhibits its own capacity to synthesize cholesterol and meets its demand also by taking it from the circulation. Defects in cholesterol synthesis and trafficking in the neural retina has detrimental consequences on its structure and function, highlighting the crucial importance of maintaining cholesterol homeostasis in the retina. Our purpose was to give a review on the functioning of the retina, the role of cholesterol and cholesterol metabolism therein, with special emphasis on cholesterol-24S-hydroxylase (CYP46A1). Similar to th…
