Cancer Stem Cells: From Birth to Death
2019
Abstract Conspicuous investigations have proven the role of cancer stem cells (CSCs) in the onset and progression of a plethora of liquid and solid neoplasms. CSCs are endowed with the capability of initiating tumor growth and becoming dormant at distant organ sites just waiting for optimal conditions amenable for metastatic outgrowth. This cancer subpopulation is inherently resistant to anticancer therapeutics, and its targeting could avoid metastatic disease, which is largely incurable, and clinical relapses. CSCs are considered the Achilles heel of cancer. However, many efforts are necessary to identify univocal CSC markers as well as specific CSC biomarkers of therapeutic response. Here…
Hype and Hopes of Stem Cell Research in Neurodegenerative Diseases
2017
Hope from the regeneration promoting effects of stem cells have provided new insights for understanding diseases that were previously thought to have a limited prognostic improvement upon medical intervention. This is especially indicated in neurodegenerative diseases, which until the discovery and research in stem cells were thought to have minimal regenerative capabilities. This review covers various treatment modalities involving different types of stem cells, such as human embryonic stem cells, induced pluripotent stem cells, mesenchymal stem cells and neural stem cells, which have been tested for various neurodegenerative disorders such as Multiple Sclerosis, Alzheimer’s disease, Parki…
Hematopoietic Stem Cell Mobilization for Gene Therapy: The Combination of G-CSF+Plerixafor in Patients with Beta-Thalassemia Major Provides High Yiel…
2015
Abstract Hematopoietic stem cell engineering is a promising therapy to cure b-thalassemia, in particular for patients who lack a suitable BM donor for allogeneic transplantation. Since the engrafted gene-corrected stem cells will not have any selective advantage over the unmodified ones, the effectiveness of the therapy in this setting largely depends on the infusion of high numbers of gene-modified cells and on the conditioning regimen. The quality of the infused cells is also crucial for the clinical outcome and the duration of the therapeutic effect. HSPCs mobilization, particularly when G-CSF and plerixafor are used in combination, has been proved to be the optimal approach to harvest a…

Impact of DLI after In Vivo T-Cell-Depletion: Prophylactic CD8 Depleted or Preemptive CD3pos DLI?
2016
Abstract Introduction: The combination of reduced-intensity conditioning (RIC) with in vivo T-cell depletion by alemtuzumab prior to hematopoietic stem cell transplantation (HSCT) has demonstrated efficient engraftment and reduced graft-versus-host disease (GVHD). However, this regimen is associated with slow lymphocyte recovery leading to a delayed anti-infectious and anti-malignant immunity. DLI can be used to improve immune reconstitution. Here we investigate on the impact of different DLI: prophylactic CD8-depleted DLI vs preemptive non-depleted DLI. Methods: 256 patients with different hematologic malignancies were planned for treatment with DLI after allogeneic HSCT following reduced …
A perspective analysis: microRNAs, glucose metabolism, and drug resistance in colon cancer stem cells
2021
Metabolism sustains the stemness of Cancer Stem Cells (CSCs), affecting, in turn, tumor heterogeneity, metastatic potential, and therapy resistance. Therefore, it is appealing to target CSCs metabolism as a new therapeutic approach. Consequently, we paid considerable attention to the anti-apoptotic microRNA miR-483-3p, that we reported being regulated by glucose metabolism in liver cancer cells. We investigated the therapeutic potential of targeting miR-483-3p by using the anti-glucose metabolism 2-deoxyglucose (2-DG) molecule in tumor Xenograft mouse model originating from two different Colon-Cancer Stem Cell lines (CCSC lines). We show that 2-DG treatment does not affect CCSCs during tumo…
Adipose stromal cells promote the transition of colorectal cancer cells toward a mesenchymal-like phenotype
2021
ABSTRACT Colon cancer progression is among the risks that increase with obesity. We have recently unveiled the molecular mechanism by which adipose tissue-released molecules, HGF and IL-6, make colorectal cancer (CRC) cells acquiring mesenchymal traits. Targeting of adipose-derived factors abrogate the metastatic potential of CRC stem cells (CR-CSCs) in obese patients.
Effective targeting of breast cancer stem cells by combined inhibition of Sam68 and Rad51
2022
AbstractBreast cancer (BC) is the second cause of cancer-related deceases in the worldwide female population. Despite the successful treatment advances, 25% of BC develops resistance to current therapeutic regimens, thereby remaining a major hurdle for patient management. Current therapies, targeting the molecular events underpinning the adaptive resistance, still require effort to improve BC treatment. Using BC sphere cells (BCSphCs) as a model, here we showed that BC stem-like cells express high levels of Myc, which requires the presence of the multifunctional DNA/RNA binding protein Sam68 for the DNA-damage repair. Analysis of a cohort of BC patients displayed that Sam68 is an independen…
