Zymomonas mobilis elpošanas ķēdes mutantu skābekļa un temperatūras stresa tolerance

Zymomonas mobilis Zm6 celms un astoņi tā elpošanas ķēdes mutantu celmi tika audzēti četros dažādos apstākļos: 30oC aerobi (kratītāja kolbās), 30oC mikroaerobos (bez kratīšanas), 42oC aerobi un 42oC mikroaerobi, lai salīdzinātu oksidatīvā un termālā stresa ietekmi uz katru no šiem celmiem. Tika noteikta celmu augšana, etanola produkcija un skābekļa patēriņš. Citohroma c peroksidāzes pārekspresija uzlaboja augšanu 30oC mikroaerobos apstākļos. Tomēr citohroma c peroksidāzes un NADH dehidrogenāzes negatīvajam dubultmutantam augšana 42oC aerobos apstākļos ir samazināta, salīdzinot ar NADH dehidrogenāzes negatīvo mutantu. Recently, mitochondrial aldehyde dehydrogenase (ALDH-2) was reported to reduce ischemic damage in an experimental myocardial infarction model. ALDH-2 activity is redox-sensitive. Therefore, we here compared effects of various electrophiles (organic nitrates, reactive fatty acid metabolites, or oxidants) on the activity of ALDH-2 with special emphasis on organic nitrate-induced inactivation of the enzyme, the biochemical correlate of nitrate tolerance.
