Capillary enlargement, not sprouting angiogenesis, determines beneficial therapeutic effects and side effects of angiogenic gene therapy.
2010
Aims Currently, it is still unclear which mechanisms drive metabolic benefits after angiogenic gene therapy. The side-effect profile of efficient angiogenic gene therapy is also currently incompletely understood. In this study, the effects of increasing doses of adenoviral (Ad) vascular endothelial growth factor-A (VEGF-A) were evaluated on vascular growth, metabolic benefits, and systemic side effects. Methods and results Adenoviral vascular endothelial growth factor-A or AdLacZ control was injected intramuscularly (109–1011 vp/mL) or intra-arterially (5 × 1011 vp/mL) into rabbit ( n = 102) hindlimb muscles and examined 6 or 14 days later. Blood flow, tissue oedema, metabolic benefits, and…
From the Field to the Laboratory: Quantifying Outdoor Mosquito Landing Rate to Better Evaluate Topical Repellents.
2021
Abstract Vector-borne diseases are a worldwide threat to human health. Often, no vaccines or treatments exist. Thus, personal protection products play an essential role in limiting transmission. The World Health Organization (WHO) arm-in-cage (AIC) test is the most common method for evaluating the efficacy of topical repellents, but it remains unclear whether AIC testing conditions recreate the mosquito landing rates in the field. This study aimed to estimate the landing rate outdoors, in an area of Europe highly infested with the Asian tiger mosquito (Aedes albopictus (Skuse, 1894, Diptera: Culididae)), and to determine how to replicate this rate in the laboratory. To assess the landing ra…
Schistosomiasis reaches Europe
2015
International audience; An outbreak of urogenital schistosomiasis has been detected in Europe, with patients affected in France,1,2 Germany,1,2 and Italy. The infection originated in Corsica, in a river north of Porto-Vecchio, a popular tourist destination (figure). The introduction of schistosomiasis is believed to be associated with infected people arriving from a region endemic for the exclusively African parasite, Schistosoma haematobium and disseminating parasite eggs through their urine into snail breeding sites along the river.
Leishmaniasis in travelers: A literature review.
2014
Leishmaniasis is a vector-borne protozoan infection whose clinical spectrum ranges from asymptomatic infection to fatal visceral leishmaniasis. Over the last decades, an increase in imported leishmaniasis cases in developed, non-endemic countries, have been pointed-out from a review of the international literature. Among the possible causes are increasing international tourism, influx of immigrants from endemic regions and military operations. The main area for the acquisition of cutaneous leishmaniasis, especially for adventure travelers on long-term trips in highly-endemic forested areas, is represented from South America, whereas popular Mediterranean destinations are emerging as the mai…
Coxsackievirus B3 VLPs purified by ion exchange chromatography elicit strong immune responses in mice
2014
Coxsackievirus B3 (CVB3) is an important cause of acute and chronic viral myocarditis, and dilated cardiomyopathy (DCM). Although vaccination against CVB3 could significantly reduce the incidence of serious or fatal viral myocarditis and various other diseases associated with CVB3 infection, there is currently no vaccine or therapeutic reagent in clinical use. In this study, we contributed towards the development of a CVB3 vaccine by establishing an efficient and scalable ion exchange chromatography-based purification method for CVB3 virus and baculovirus-insect cell-expressed CVB3 virus-like particles (VLPs). This purification system is especially relevant for vaccine development and produ…
Expression and cellular localization of the Nef protein from human immunodeficiency virus-1 in stably transfected B-cells.
1992
Nef protein, encoded by the regulatory nef gene of human immunodeficiency virus type 1 (HIV-1), was expressed in the B-cell line Raji. The cells were stably transfected with plasmids containing the nef transcriptional cassette. They expressed Nef with an Mr of 27,000; the yield could be augmented by incubation with the tumor promoter 12-O-tetradecanoylphorbol-13-acetate. The intracellular localization of Nef was analyzed applying immunofluorescence microscopy using a confocal laser scanning microscope. The antigen was stained with a monoclonal antibody directed against the N-terminal part of Nef. The experiments revealed that in non-dividing cells Nef is present both in the cytoplasm and th…
Treatment of Anderson-Fabry Disease
2020
Fabry disease is an X-linked disorder of glycosphingolipid metabolism that results in progressive accumulation of neutral glycosphingolipids, predominantly globotriaosylsphingosine (Gb3) in lysosomes, as well as other cellular compartments of several tissues, causing multi-organ manifestations (acroparesthesias, hypohidrosis, angiokeratomas, signs and symptoms of cardiac, renal, cerebrovascular involvement). Pathogenic mutations lead to a deficiency of the lysosomal enzyme alpha-galactosidase A (GLA). In the presence of high clinical suspicion, a careful physical examination and specific laboratory tests are required. Finally, the diagnosis of Fabry’s disease is confirmed by the demonstrat…
Witt vectors and Fermat quotients
2008
Abstract We give a representation of any integer as a vector of the Witt ring W ( Z p ) and relate it to the Fermat quotient q ( n ) = ( n p − 1 − 1 ) / p . Logarithms are introduced in order to establish an isomorphism between the commutative unipotent groups 1 + p W ( Z p ) and W ( Z p ) .
Study of biogenic amines metabolism in wine lactic acid bacteria
2011
Biogenic amines are indesirable compounds found in fermented products like wine. Lactic acid bacteria from wine, including Oenococcus oeni, the main actor of malolactic fermentation, are able to produce these molecules from nitrogenous precursors. In order to limited biogenic amines accumulation, it is necessary to understand the role of this production by strains responsible for the synthesis of these metabolites in food. That is why the European BiamFood project was put in place. Along my thesis, molecular tools were developed in order to mutate O. oeni genes (encoding decarboxylases), and to express genes of interest. Genetic clusters hdc from L. higardii and odc from O. oeni, responsibl…
