Melatonin's transduction mechanisms were investigated using in vitro cultured anterior hemipituitaries. Melatonin inhibited cAMP and 3',5'-cyclic GMP accumulation in neonatal rat anterior pituitary stimulated with LHRH. Maximal inhibitory effect was reached within 25 min and persisted for at least 20 min. Inhibition of cAMP accumulation is specific for melatonin because its analogs N-acetylserotonin and 5-methoxytryptamine are 1000 times less potent. Melatonin effect is age- and time-dependent. Marked inhibition was observed in 5-, 10-, and 14-day-old rats but not in 29-day-old ones. The G protein coupling characteristics of a flag epitope-tagged orexin receptor type 1 (OX1R) was investigated in HEK293 cells. Immunoprecipitation of the OX1R and immunoblotting revealed interactions with Gq/G11 proteins as well as with Gs and Gi proteins. Stimulation with orexin-A did not affect the ability of the OX1R to coprecipitate Gq/G11 proteins, but it robustly elevated the intracellular concentration of Ca2+, [Ca2+]i. No changes in cAMP levels could be detected upon receptor stimulation. The formation and structural features of positively mono-charged aggregates of sodium bis(2-ethylhexyl) sulfosuccinate (AOT) and sodium methane—(MetS), butane—(ButS) and octane—(OctS) sulfonate molecules in the gas phase have been investigated by electrospray ionization mass spectrometry, energy-resolved mass spectrometry and density functional theory (DFT) calculations. The conservation status of amphibians is often assessed using public databases because of a lack of up-to-date field data. However, it is crucial that this kind of data is used carefully, evaluating the reliability and the consistency of the information. In Italy, the conservation status of the painted frog, Discoglossus pictus, was recently assessed using public databases, and the outcome highlighted a worrying situation for the species.
