Clinical and Molecular-Based Approach in the Evaluation of Hepatocellular Carcinoma Recurrence after Radical Liver Resection
2021
Background: Hepatic resection remains the treatment of choice for patients with early-stage HCC with preserved liver function. Unfortunately, however, the majority of patients develop tumor recurrence. While several clinical factors were found to be associated with tumor recurrence, HCC pathogenesis is a complex process of accumulation of somatic genomic alterations, which leads to a huge molecular heterogeneity that has not been completely understood. The aim of this study is to complement potentially predictive clinical and pathological factors with next-generation sequencing genomic profiling and loss of heterozygosity analysis. Methods: 124 HCC patients, who underwent a primary hepatic …
ABCL-366: Mosunetuzumab (Mosun) Monotherapy for Elderly/Unfit Patients with First-Line Diffuse Large B-Cell Lymphoma (DLBCL) Continues to Show Promis…
2021
Context Mosun, a full-length, humanized, IgG1 CD20×CD3 bispecific antibody, demonstrated efficacy/safety in relapsed/refractory DLBCL (NCT02500407; Schuster, et al. ASH 2019). The phase I/II multicenter GO40554 (NCT03677154) study showed similar results for elderly/unfit patients with first-line DLBCL intolerant of full-dose chemoimmunotherapy (CIT) (Olszewski, et al. ASH 2020). Objective To describe updated GO40554 study data. Methods Two safety-evaluation cohorts were assessed (Mosun 13.5 mg and 30 mg), then a 30 mg expansion phase. Patients were aged ≥80 years; 60–79 years with impairment in ≥1 activity of daily living (ADL); instrumental ADL; or impaired cardiac, renal, or liver functio…

The disease and treatment of patients with head and neck cancer can lead to multiple late and long-term sequelae. Especially pain, psychosocial problems, and voice issues can have a high impact on patients’ health-related quality of life. The aim was to show the feasibility of implementing an electronic Patient-Reported Outcome Measure (PROM) in patients with head and neck cancer (HNC). Driven by our department’s intention to assess Patient-Reported Outcomes (PRO) based on the International Classification of Functioning during tumor aftercare, the program “OncoFunction” has been implemented and continuously refined in everyday practice. The new version of “OncoFunction” was evaluated by 20 …
Refining Prognosis in Chemoembolization for Hepatocellular Carcinoma: Immunonutrition and Liver Function
2021
A combination of albumin-bilirubin (ALBI) grading and the Prognostic Nutritional Index (PNI) was identified recently as a highly predictive tool for patients with hepatocellular carcinoma (HCC) undergoing tumor ablation. The present study evaluated this combination in patients undergoing transarterial chemoembolization (TACE). Between 2010 and 2020, 280 treatment-naïve patients were retrospectively identified. The influence of ALBI grade, PNI and the novel ALBI-PNI on the median overall survival (OS) was assessed. In the next step, the prognostic ability of the combined approach was compared to established scoring systems. Both ALBI grade 2−3 and a low PNI were highly predictive for median …
Clinical and therapeutic features of acute cholecystitis in diabetic patients
2021
The present study aimed to compare the clinical, paraclinical, intraoperative findings, and postoperative complications in acute cholecystitis in diabetic patients vs. non-diabetic patients. A 2-year retrospective study was performed on the patients who underwent emergency cholecystectomy for acute cholecystitis between 2017 and 2019 at the 4th Department of Surgery, Emergency University Hospital Bucharest. The diabetic subgroup numbered 46 eligible patients and the non-diabetic one 287 patients. Demographics, the severity of the clinical forms, biological variables (including white cell count, urea, creatinine, coagulation and liver function tests) comorbidity status, surgical approach, po…
Osteo-Oto-Hepato-Enteric Syndrome (O2HE) is caused by loss of function mutations in UNC45A
2017
AbstractDespite the rapid discovery of genes for rare genetic disorders, we continue to encounter individuals presenting with hitherto unknown syndromic manifestations. Here, we have studied four affected people in three families presenting with cholestasis, congenital diarrhea, impaired hearing and bone fragility, a clinical entity we have termed O2HE (Osteo-Oto-Hepato-enteric) syndrome. Whole exome sequencing of all affected individuals and their parents identified biallelic mutations in Unc-45 Myosin Chaperone A (UNC45A), as a likely driver for this disorder. Subsequent in vitro and in vivo functional studies of the candidate gene indicated a loss of function paradigm, wherein mutations …
Fulminant hepatic failure requiring liver transplantation in 22q13.3 deletion syndrome.
2010
We report on a 4-year-old girl with severe developmental delay, absent speech, and chromosome 22q13.3 deletion (Phelan-McDermid syndrome), karyotype 46,XX.ish del(22)(q13.31qter)(ARSA-,N85A-,SHANK3-). At the age of 3 years, she needed an emergency liver transplantation because of fulminant hepatic failure, most likely caused by hyperacute autoimmune hepatitis triggered by a viral infection. This is the second report of a patient with 22q13.3 deletion and fulminant liver failure. By array-CGH we identified in this patient a 5.675 Mb terminal deletion (22q13.31 --> qter; including approximately 55 genes; from NUP50 to RABL2B) and in the previous patient a 1.535 Mb deletion (22q13.32 --> qter;…
Simulation studies of gas-liquid transitions in two dimensions via a subsystem-block-density distribution analysis
1993
The finite-size scaling analysis of the density distribution function of subsystems of a system studied at constant total density is studied by a comparative investigation of two models: (i) the nearest-neighbor lattice gas model on the square lattice, choosing a total lattice size of 64×64 sites. (ii) The two-dimensional off-lattice Lennard-Jones system (truncated at a distance of 2.5 σ, σ being the range parameter of the interaction) withN=4096 particles, applying the NVT ensemble. In both models, the density distribution functionPL(ρ) is obtained forL×L subsystems for a wide range of temperaturesT, subblock linear dimensionsL and average densities . Particular attention is paid to the qu…

Shear Effects in Elastic Nanobeams
2020
Small-scale, shear deformable nanobeams, subjected to quasi-static loads, are analyzed by a nonlocal (integral) elasticity model with the main goal to evaluate the influence of shear deformation on size effects. To this aim a warping parametric model is considered in order to obtain a continuous family of shear deformable beam models which span from the Euler-Bernoulli to the Thimoshenko beam model, passing from the Reddy model. The strain difference based nonlocal elasticity theory is applied under the hypotheses of small displacements and isotropic material. The results, obtained by analysing a cantilever nonlocal nanobeam, indicate that shear deformation has a considerable influence upon…
