The catalytic properties of DNA G-quadruplexes rely on their structural integrity

2021

International audience; The influence of the G-quartet structural integrity on the catalytic activity of the G-quadruplex (G4) was investigated by comparing the G4-DNAzyme performances of a series of G4s with a G-vacancy site and a G-triplex (G-tri). The results presented herein not only confirm that the structural integrity of the 3’-end G-quartet is necessary for G4s to be catalytically competent but also show how to remediate G-vacancy-mediated catalytic activity losses via the addition of guanine surrogates in an approach referred to as G-vacancy complementation strategy that is applicable to parallel G4s only. Furthermore, this study demonstrates that the terminal G-quartet could act a…

8-Oxo-7,8-dihydroguanine in DNA does not constitute a barrier to transcription, but is converted into transcription-blocking damage by OGG1.

2011

The common DNA base modification 8-oxo-7,8-dihydroguanine (8-oxo-G) affects the efficiency and fidelity of transcription. We constructed plasmid substrates carrying single 8-oxo-G residues, specifically positioned in the transcribed or the non-transcribed DNA strands, to investigate their effects on the expression of an EGFP reporter gene and to explore the role of base excision repair in the mechanism of transcription inhibition. We report that 8-oxo-G does not directly block transcription in cells, since a single 8-oxo-G in the transcribed DNA strand did not reduce the EGFP expression levels in repair-deficient (OGG1-null) mouse embryonic fibroblast cell lines. Rather, inhibition of trans…

Management of Listeria monocytogenes on Surfaces via Relative Air Humidity: Key Role of Cell Envelope

2021

International audience; Although relative air humidity (RH) strongly influences microbial survival, its use for fighting surface pathogens in the food industry has been inadequately considered. We asked whether RH control could destroy Listeria monocytogenes EGDe by envelope damage. The impact of dehydration in phosphate-buffered saline (PBS) at 75%, 68%, 43% and 11% RH on the bacterial envelope was investigated using flow cytometry and atomic force microscopy. Changes after rehydration in the protein secondary structure and peptidoglycan were investigated by infrared spectroscopy. Complementary cultivability measurements were performed by running dehydration–rehydration with combinations o…

On Using Conceptual Modeling for Ontologies

2004

Are database concepts and techniques suitable for ontology design and management? The question has been on the floor for some time already. It gets a new emphasis today, thanks to the focus on ontologies and ontology services due to the spread of web services as a new paradigm for information management. This paper analyzes some of the arguments that are relevant to the debate, in particular the question whether conceptual data models would adequately support the design and use of ontologies. It concludes suggesting a hybrid approach, combining databases and logic-based services.

Trust-enhanced intelligent security model

2012

In this paper we propose a trust-enhancement of access control to protect both integrity and confidentiality based on trustworthiness of users performing operations and documents' content analysis. We propose to utilize trustworthiness opinions from subjective logic and express levels of integrity as levels of trustworthiness. We assign confidentiality levels based on contents of documents and use opinions to express trustworthiness of such assignments.

Ultrascale+ for the new ATLAS calorimeter trigger board dedicated to jet identification

2018

To cope with the expected increase in luminosity at the Large Hadron Collider in 2021, the ATLAS collaboration is planning a major detector upgrade to be installed during Long Shutdown 2. As a part of this, the Level 1 trigger, based on calorimeter data, will be upgraded to exploit the fine granularity readout using a new system of Feature EXtractors (FEXs), which each reconstruct different physics objects for the trigger selection. The Jet FEX (jFEX) is one of three FEXs and has been conceived to identify small/large area jets, large area tau leptons, missing transverse energy and the total sum of the transverse energy. The use of the latest generation Xilinx Field Programmable Gate Array …

Can PBDEs affect the pathophysiologic complex of epithelium in lung diseases?

2020

Brominated flame-retardant (BFRs) exposure promotes multiple adverse health outcomes involved in oxidative stress, inflammation, and tissues damage. We investigated BFR effects, known as polybrominated diphenyl ethers (PBDEs) (47, 99 and 209) in an air-liquid-interface (ALI) airway tissue derived from A549 cell line, and compared with ALI culture of primary human bronchial epithelial cells (pHBEC). The cells, exposed to PBDEs (47, 99 and 209) (0.01-1 mu M) for 24 h, were studied for IL-8, Muc5AC and Muc5B (mRNAs and proteins) production, as well as NOX-4 (mRNA) expression. Furthermore, we evaluated tight junction (TJ) integrity by Trans-Epithelial Electrical Resistance (TEER) measurements, …

Multi-physical analysis of the electrochemical behaviour of OFHC copper surfaces obtained by orthogonal cutting

2020

International audience; As mechanical, physical, and microstructural properties of metals can be significantly affected by the machining process, electrochemical properties and corrosion resistance are consequently altered. Manufacturers should control the impact of cutting conditions on surface integrity, as it will affect the component’s functional performance and life. This study addresses the case of orthogonal cutting of oxygen-free high conductivity (OFHC) copper. A set of experiments was performed and a statistical analysis was conducted to reveal the relationship between the cutting conditions and the surface integrity in terms of residualstresses, microstructure, plastic deformatio…

Selective Brain Network and Cellular Responses Upon Dimethyl Fumarate Immunomodulation in Multiple Sclerosis

2019

Background: Efficient personalized therapy paradigms are needed to modify the disease course and halt gray (GM) and white matter (WM) damage in patients with multiple sclerosis (MS). Presently, promising disease-modifying drugs show impressive efficiency, however, tailored markers of therapy responses are required. Here, we aimed to detect in a real-world setting patients with a more favorable brain network response and immune cell dynamics upon dimethyl fumarate (DMF) treatment. Methods: In a cohort of 78 MS patients we identified two thoroughly matched groups, based on age, disease duration, disability status and lesion volume, receiving DMF (n = 42) and NAT (n = 36) and followed them ove…
