IL-4-mediated drug resistance in colon cancer stem cells

Cancer stem cells are defined as cells able to both extensively self-renew and differentiate into progenitors. Cancer stem cells are thus likely to be responsible for maintaining or spreading a cancer, and may be the most relevant targets for cancer therapy. The CD133 glycoprotein was recently described as a reliable cancer stem-like cell marker in colon carcinoma. CD133+ cells are both necessary and sufficient to initiate tumour growth in animal models. [ES] En el presente trabajo presentamos un proyecto de biología sintética de nivel profesional adaptado a estudiantes de primer curso. Describimos las adaptaciones necesarias para hacerlo asequible para unas prácticas de laboratorio, tanto a nivel pedagógico como en cuanto a la viabilidad económica de los materiales utilizados. El proyecto pretende la motivación de los estudiantes del Grado en Biotecnología mediante la introducción temprana de la actividad profesional real en biomedicina, así como la adquisición de una serie de competencias del trabajo práctico en el laboratorio imprescindibles para el biotecnólogo en formación.

[EN] University students of experimental sciences degrees usually go to the laboratory of practices without having prepared the corresponding activity. This fact leads to a poor preparation of them translated into an effective loss of time available for experimentation, and a reduced acquisition of knowledge and skills, among other aspects. In order to solve this problem, in the present communication, the use of on-line questionnaires using the Socrative platform is described to be used as a prior evaluation system to access the laboratory. [EN] The so-called “threshold concepts” are present in all areas of study. Its acquisition by the student represents a milestone in the advancement of the conceptual understanding of a subject. They are often difficult to learn, but when they are overcome they generate a reconceptualization that allows progress and deepening in the subject of study. We present an activity designed to develop a key threshold concept in pharmacology, the drug discovery process. To do this, we develop a practical laboratory project that will integrate both the extraction and search for active ingredients and the simulation of some aspects of their clinical trials in the phase I.

Obesity is a strong risk factor for cancer progression, posing obesity-related cancer as one of the leading causes of death. Nevertheless, the molecular mechanisms that endow cancer cells with metastatic properties in patients affected by obesity remain unexplored. Here, we show that IL-6 and HGF, secreted by tumor neighboring visceral adipose stromal cells (V-ASCs), expand the metastatic colorectal (CR) cancer cell compartment (CD44v6 + ), which in turn secretes neurotrophins such as NGF and NT-3, and recruits adipose stem cells within tumor mass. Vascular calcification has been recently associated to an increased cardiovascular risk and mortality. In few studies, Fetuin-A showed an association to coronary artery calcification (CAC), although the physiopathological mechanism underlying this association has not been fully established yet. Seventy-four patients with one or more cardiovascular risk factor and asymptomatic for coronary vasculopathy were included in the study. CAC was evaluated by Agatston score. SREBP1 and 2, are cholesterol sensors able to modulate cholesterol-related gene expression responses. SREBPs binding sites are characterized by the presence of multiple target sequences as SRE, NFY and SP1, that can be arranged differently in different genes, so that it is not easy to identify the binding site on the basis of direct DNA sequence analysis. This paper presents a complete workflow based on a one-dimensional Convolutional Neural Network (CNN) model able to detect putative SREBPs binding sites irrespective of target elements arrangements.
