The Controversy Between Erich Hückel and Linus Pauling over the Benzene Problem

2009

Shortly after Huckel’s quantum-theoretical work on the problem of aromatic compounds was published, the first paper on the same subject by the American Linus Pauling also appeared. It was the fifth installment of a total of seven that Pauling published between April 1931 and July 1933 under the general title The Nature of the Chemical Bond. This fifth part was the first coauthored with his pupil George Wheland, a National Research Fellow in Pasadena. In their quantum mechanical treatment of benzene, naphthaline and free organic radicals, they applied a “VB” approximation slightly different from Huckel’s “first method” yet sharing some resemblance with it. The two subsequent parts of Pauling’…

Click modification of multifunctional liposomes bearing hyperbranched polyether chains.

2014

Aiming at controlled modification of liposomal surface structures, we describe a postpreparational approach for surface derivatization of a new type of multifunctional, sterically stabilized liposomes. Application of dual centrifugation (DC) resulted in high encapsulation efficiencies above 50% at very small batch sizes with a total volume of 150 μL, which were conductive to fast and efficient optimization of variegated surface modification reactions. Cholesterol-polymer amphiphiles, including complex hyperbranched polyether structures bearing 1-4 terminal alkynes, were used in DC formulations to provide steric stabilization. The alkyne moieties were explored as anchors for the conjugation …

Single Molecules Probing the Freezing of Polymer Melts: A Molecular Dynamics Study for Various Molecule-Chain Linkages

2010

8 pages; International audience; We present molecular dynamics simulations of coarse-grained model systems of a glassforming polymer matrix containing fluorescent probe molecules. These probe molecules are either dispersed in the matrix or covalently attached to the center or the end of a dilute fraction of the polymer chains. We show that in all cases the translational and rotational relaxation of the probe molecules is a faithful sensor for the glass transition of the matrix as determined from a mode-coupling analysis or Vogel-Fulcher analysis of their R-relaxation behavior. Matrix and dumbbell related relaxation processes show a clear violation of the Stokes-Einstein-Debye laws. In accor…

Prospective Cancer Therapies Using Stimuli‐Responsive DNA Nanostructures

2021

Financial support by the Emil Aaltonen Foundation, the Sigrid Jusélius Foundation, the Magnus Ehrnrooth Foundation, Academy of Finland (grants no. 317042 and 331151), the Jane and Aatos Erkko Foundation and the Vilho, Yrjö and Kalle Väisälä Foundation of the Finnish Academy of Science and Letters is gratefully acknowledged Nanostructures based on DNA self-assembly present an innovative way to address the increasing need for target-specific delivery of therapeutic molecules. Currently, most of the chemotherapeutics being used in clinical practice have undesired and exceedingly high off-target toxicity. This is a challenge in particular for small molecules, and hence, developing robust and ef…

Synthesis of fluorinated first generation starburst molecules containing a triethanolamine core and 1,2,4-oxadiazoles

2006

Abstract 5-Pentafluorophenyl-1,2,4-oxadiazoles, differently substituted at C(3), have been synthesized and used to functionalize the aliphatic primary hydroxyl functionalities of triethanolamine, under mild conditions. UV–visible absorption and emission spectra are reported, for both tri- and difunctionalised molecules, showing their potential applications as light-emitters in optoelectronic devices.

Cellular Models and Assays to Study NLRP3 Inflammasome Biology

2020

The NLRP3 inflammasome is a multi-protein complex that initiates innate immunity responses when exposed to a wide range of stimuli, including pathogen-associated molecular patterns (PAMPs) and danger-associated molecular patterns (DAMPs). Inflammasome activation leads to the release of the pro-inflammatory cytokines interleukin (IL)-1β and IL-18 and to pyroptotic cell death. Over-activation of NLRP3 inflammasome has been associated with several chronic inflammatory diseases. A deep knowledge of NLRP3 inflammasome biology is required to better exploit its potential as therapeutic target and for the development of new selective drugs. To this purpose, in the past few years, several tools have…

Morpholino knockdown of the ubiquitously expressed transmembrane serine protease TMPRSS4a in zebrafish embryos exhibits severe defects in organogenes…

2011

AbstractOver the past years the members of the type II transmembrane serine protease (TTSP) family have emerged as new players in mammalian biology. TMPRSS4 (transmembraneprotease/serine) is overexpressed in several human cancer tissues, promoting invasion, migration, and metastasis. However, the physiological function has not yet been elucidated. Here, we present morpholino knockdown studies targeting TMPRSS4a, a homolog of human TMPRSS4 in zebrafish embryos. By RT-PCR, we could demonstrate an expression of this protease already 5 h post-fertilization, suggesting important functions in the early stages of embryonic development. Indeed,in vivogene silencing caused severe defects in tissue d…
