Cytotoxicity of ungeremine towards multi-factorial drug resistant cancer cells and induction of apoptosis, ferroptosis, necroptosis and autophagy

2018

Abstract Background Successful cancer chemotherapy is hampered by resistance of cancer cells to established anticancer drugs. Numerous natural products reveal cytotoxicity towards tumor cells. Purpose The present study was aimed to determine the cytotoxicity of a betaine-type alkaloid, ungeremine, towards 9 cancer cell lines including various sensitive and drug-resistant phenotypes. The mode of action of this compound was further investigated. Methods The cytotoxicity, ferroptotic and necroptotic cell death were determined by the resazurin reduction assay. Caspase activation was evaluated using the caspase-Glo assay. Flow cytometry was applied for the analysis of cell cycle analysis (PI sta…

8,8-bis-(Dihydroconiferyl)-diferulate displayed impressive cytotoxicity towards a panel of human and animal cancer cells

2020

Abstract Background Recalcitrant cancers appear as a major obstacle to chemotherapy, prompting scientists to intensify the search for novel drugs to tackle the cell lines expressing multi-drug resistant (MDR) phenotypes. Purpose The purpose of this study was to evaluate the antiproliferative potential of a ferrulic acid derivative, 8,8-bis-(dihydroconiferyl)-diferulate (DHCF2) on a panel of 18 cancer cell lines, including various sensitive and drug-resistant phenotypes, belonging to human and animals. The mode of induction of cell death by this compound was further studied. Methods The antiproliferative activity, autophagy, ferroptotic and necroptotic cell death were evaluated by the resazu…

Programmed cell death (PCD) associated with the stilbene motif of arotinoids: discovery of novel apoptosis inducer agents possessing activity on mult…

2000

Considering that the stereochemistry of the C9-C10 alkenyl portion of natural 9-cis-RA, as the one of the olefinic moiety of the previously described isoxazole retinoid 4, seems of particular importance for their apoptotic activity, we prepared a novel class of TTNPB analogues bearing both the cis or trans configuration of the alkenyl portion. The compounds were evaluated in vitro for their cytotoxic and apoptotic activities. We discovered that the cis-TTNPB 9c possesses apoptotic activity comparable with that of the retinoid 4. Moreover, the amino arotinoid 16c showed potent apoptotic activity in HL60 promyelocytic leukemia cells. Interestingly, 16c proved to be a particularly potent apopt…

Antiproliferative Properties of a Few Auranofin-Related Gold(I) and Silver(I) Complexes in Leukemia Cells and their Interferences with the Ubiquitin …

2020

A group of triethylphosphine gold(I) and silver(I) complexes, structurally related to auranofin, were prepared and investigated as potential anticancer drug candidates. The antiproliferative properties of these metal compounds were assessed against two leukemia cell lines, i.e., CCRF-CEM and its multidrug-resistant counterpart, CEM/ADR5000. Interestingly, potent cytotoxic effects were disclosed for both series of compounds against leukemia cells, with IC50 values generally falling in the low-micromolar range, the gold derivatives being on the whole more effective than the silver analogues. Some initial structure-function relationships were drawn. Subsequently, the ability of the study compo…

Exposure to natalizumab throughout pregnancy: effectiveness and safety in an Italian cohort of women with multiple sclerosis.

2022

ObjectiveAssessing the risk of clinical and radiological reactivation during pregnancy and post partum in women with multiple sclerosis (MS) treated with natalizumab (NTZ) throughout pregnancy (LONG_EXP) compared with women interrupting treatment before (NO_EXP) and within >−30 days and ≤90 days from conception (SHORT_EXP), and describing newborns’ outcomes.MethodsMaternal clinical and radiological outcomes and obstetric and fetal outcomes were retrospectively collected and compared among groups (NO_EXP, SHORT_EXP, LONG_EXP). Predictors of clinical and radiological reactivation were investigated through univariable and multivariable analysis.Results170 eligible pregnancies from 163 women…

Supporting public decision process in buildings energy retrofitting operations: the application of a Multiple Criteria Decision Aiding model to a cas…

2020

Abstract The challenge of promoting sustainable cities and reaching the objectives developed by the European Green Deal includes the renovation of the building sector, as it is responsible for 40% of energy consumption in Europe. Regional or local public administrations have to allocate their financial resources for improving the energy performances of their building stock and to face a multidimensional problem, where different aspects – such as energy efficiency, financial-economic feasibility and environmental protection – have to be harmonized. The present study proposes a Multiple Criteria Decision Aiding model, which includes the ELECTRE TRI-nC method, for supporting the public decisio…

Synchronous Two Distinct Neuroendocrine Lung Cancer Lesions

2015

Synchroniczne nowotwory płuc są rzadką chorobą, a prezentowany przypadek to pierwszy opublikowany opis jednoczesnego wystąpienia dwóch pierwotnych neuroendokrynnych raków płuca zlokalizowanych w jednym płacie.

Differential changes in purine nucleotides after Doxorubicin treatment of human cancer cells in vitro

2002

The present investigation was performed to elucidate the role of purine nucleotides as potential indicators of chemosensitivity of malignant tumors. Drug-sensitive (s) and -resistant (r) tumor cell lines grown as monolayers (s: T47D, MCF-7 wild-type; r: NCI/ADR-RES, MCF-7/MDR) or as multicellular spheroids (T47D; NCI/ADR-RES) were exposed to 0.1, 1.0, and 10.0 microM Doxorubicin for up to 24 h. Purine nucleotides were assayed using HPLC and with some selected spheroids using imaging bioluminescence. The data show that in the time frame of the experiments reproducible and statistically significant changes in the nucleotides only occur at the highest drug concentration investigated. Under the…

Combining super-orthogonal space-frequency trellis coding, constellation shaping by shell mapping, and OFDM for high data rate broadband mobile commu…

2014

In this paper, we seamlessly combine three transmission techniques to design an improved performance broadband mobile communication system with two transmit antennas. In so doing, we took inspiration from the V.34 data modem toolbox designed for public switched telephone network. We modified the constellation shaping by shell mapping algorithm to apply it only to the orthogonal frequency division multiplexing (OFDM) frame sent by the first antenna, in such a way that the peak-to-average power ratio (PAPR) is also reduced for the second antenna. We use a 24-point quadrature amplitude modulation (QAM) signal constellation on each subcarrier, while the reference system employs 16 QAM for the s…
