Type V collagen-induced upregulation of capn2 (large subunit of m-calpain) gene expression and DNA fragmentation in 8701-BC breast cancer cells

2011

Abstract Type V collagen is known to be over-deposited in the stroma of ductal infiltrating carcinomas of the breast. When used as a substrate, type V collagen restrains growth and invasion, and affects gene expression of 8701-BC ductal infiltrating carcinomas cells. Here we supplement existing data by demonstrating type V collagen dependent upregulation of capn2 gene expression in 8701-BC cells through differential display-PCR and Western blot assays. Furthermore, we suggest that our data obtained by centrifugal sedimentation and electrophoresis strongly suggest a correlation between calpain overproduction and DNA fragmentation, since the incubation with calpain inhibitor partly reverts th…

Paternity testing of endangered species of birds by DNA fingerprinting and random amplified polymorphic DNA fingerprinting

1994

Since 1985 DNA fingerprinting is the method of choice to solve forensic problems such as paternity testing (Jeffreys et al., 1986) or identification of immigration cases (Jeffreys et al., 1985 a, b). Its applicability became evident not only in man but also in many biological problems. Jeffreys’probes 33.15 and 33.6 or Epplen’s oligonucleotide probes were used in DNA fingerprinting of dogs and cats (Jeffreys & Morton, 1987), non-human primates (Weiss, 1989), birds (Longmire et al., 1991, Miyaki et al., 1993), fishes (Schartl et al., 1993) or even plants (Weising et al., 1991). Especially its application in paternity testing turned to account in many species of animals (Lubjuhn et al., 1991,…

EZH2 mutations are frequent and represent an early event in follicular lymphoma

2013

Gain of function mutations in the H3K27 methyltransferase EZH2 represent a promising therapeutic target in germinal center lymphomas. In this study, we assessed the frequency and distribution of EZH2 mutations in a large cohort of patients with follicular lymphoma (FL) (n = 366) and performed a longitudinal analysis of mutation during the disease progression from FL to transformed FL (tFL) (n = 33). Mutations were detected at 3 recurrent mutation hot spots (Y646, A682, and A692) in 27% of FL cases with variant allele frequencies (VAF) ranging from 2% to 61%. By comparing VAF of EZH2 with other mutation targets (CREBBP, MLL2, TNFRSF14, and MEF2B), we were able to distinguish patients harbori…

Identification of subspecies and parentage relationship by means of DNA fingerprinting in two exemplary of Pan troglodytes (Blumenbach, 1775) (Mammal…

2018

Four chimpanzee subspecies (Mammalia Hominidae) are commonly recognised: the Western Chimpanzee, P. troglodytes verus (Schwarz, 1934), the Nigeria-Cameroon Chimpanzee, P. troglodytes ellioti, the Central Chimpanzee, P. troglodytes troglodytes (Blumenbach, 1799), and the Eastern Chimpanzee, P. troglodytes schweinfurthii (Giglioli, 1872). Recent studies on mitochondrial DNA show the incorporation of P. troglodytes schweinfurthii in P. troglodytes troglodytes, suggesting the existence of only two subspecies: P. troglodytes troglodytes in Central and Eastern Africa and P. troglodytes verus-P. troglodytes ellioti in West Africa. The aim of the present study is twofold: first, to identify the cor…

Phenol Profiling and Nutraceutical Potential of

2019

In recent years, agricultural and industrial residues have attracted a lot of interest in the recovery of phytochemicals used in the food, pharmaceutical, and cosmetic industries. In this paper, a study on the recovery of phenol compounds from Lycium spp. leaves is presented. Ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE) have been used with alcoholic and hydroalcoholic solvents. Methanolic UAE was the most successful technique for extracting phenols from Lycium leaves, and we used on leaves from L. barbarum and L. chinense cultivated in Italy. The extracts were then characterized as regards to the antioxidant properties by in vitro assays and the phenol profil…

Methodological advances in the functional profiling of genomic studies

2013

In this thesis we present bioinformatic tools and algorithms for the analysis of genomic data such as those generated by microarray devices or next generation sequencing techniques. Particularly, we develop new approaches to gene set analysis. The described procedures should be useful in practice to tackle complex biological experiments, but hopefully will also be methodologically relevant, as they introduce new ways of conceptualizing genomic functional profiling. Our very flexible approach allows for the inclusion of not just one kind of genomic measurement but many. It makes possible, for instance, to analyze expression measurement and genomic variation data at a time. This multidimensio…

Testing chemical carcinogenicity by using a transcriptomics HepaRG-based model?

2014

The EU FP6 project carcinoGENOMICS explored the combination of toxicogenomics and in vitro cell culture models for identifying organotypical genotoxic- and non-genotoxic carcinogen- specific gene signatures. Here the performance of its gene classifier, derived from exposure of metabolically competent human HepaRG cells to prototypical non-carcinogens (10 compounds) and hepatocarcinogens (20 compounds), is reported. Analysis of the data at the gene and the pathway level by using independent biostatistical approaches showed a distinct separation of genotoxic from non-genotoxic hepatocarcinogens and non-carcinogens (up to 88 % correct prediction). The most characteristic pathway responding to …

The Ferroxidase Hephaestin in Lung Cancer: Pathological Significance and Prognostic Value

2021

AbstractIron is a fundamental nutrient utilized by living cells to support several key cellular processes. Despite its paramount role to sustain cell survival, excess of labile iron availability can inflict severe cell damage via reactive oxygen species generation which, in turn, can promote neoplastic transformation. The lung is particularly sensitive to iron-induced oxidative stress, given the high oxygen tensions herein present. Moreover, cigarette smoke as well as air pollution particulate can function as vehicles of iron supply, leading to an iron dysregulation condition shown to be crucial in the pathogenesis of several respiratory diseases including lung cancer. Hephaestin (HEPH) bel…
