Therapeutic-like properties of a dopamine uptake inhibitor in animal models of amphetamine addiction.

2010

N-substituted benztropine (BZT) analogs are molecules that display high affinity for the dopamine transporter (DAT), therapeutic-like effects in animal models of cocaine abuse, and psychopharmacological characteristics consistent with those of a substitute medication for cocaine addiction. Since amphetamine (Amph) and cocaine share mechanisms of action at the DAT, we evaluated the effectiveness of a BZT analog in animal models of Amph addiction. We tested in mice and rats the effects of the BZT derivative, 3α-[bis(4-fluorophenyl)methoxy]-tropane (AHN-1055), on Amph-induced conditioned place preference (CPP), locomotor activity, sensitization, self-administration and ΔFosB accumulation in th…

Effects of aging in salt spray conditions on flax and flax/basalt reinforced composites: Wettability and dynamic mechanical properties

2016

Abstract In the last years, the industrial policies are more attentive to issues concerning sustainability, recycling and environmental care. Therefore, the use of natural fibres in composite materials has spread more and more. This paper deals with flax and basalt fibres within an epoxy matrix by investigating the wettability and the dynamic mechanical properties of the resulting composites, subjected to long-term aging tests in critical environmental conditions. The first laminate was constituted by stacking ten layers of bidirectional flax fabrics. The second one was produced by replacing two external flax layers with two layers of basalt mat, for each side of the laminate. Both laminate…

Effect of temperature on ammonium removal in Scenedesmus sp

2015

The effect of temperature on microalgal ammonium uptake was investigated by carrying out four batch experiments in which a mixed culture of microalgae, composed mainly of Scenedesmus sp., was cultivated under different temperatures within the usual temperature working range in Mediterranean climate (15-34 ºC). Ammonium removal rates increased with temperature up to 26 ºC and stabilized thereafter. Ratkowsky and Cardinal Temperatures models successfully reproduced the experimental data. Optimum (31.3 ºC), minimum (8.8 ºC) and maximum (46.1 ºC) temperatures for ammonium removal by Scenedesmus sp. under the studied conditions were obtained as model parameters. These temperature-related paramet…

RELATIONSHIP BETWEEN MAXIMAL FAT OXIDATION AND OXYGEN UPTAKE: COMPARISON BETWEEN PATIENTS WITH TYPE 2 DIABETES AND HEALTHY SEDENTARY SUBJECTS

2014

The contribution of fat oxidation to energy production during exercise is influenced by intensity of exercise. The aim of this study was to assess the relationship between the highest value of fat oxidation rate (FATmax) and the oxygen uptake (VO2) in sedentary type 2 diabetes (T2D) patients vs healthy sedentary subjects. Sedentary T2D patients and healthy sedentary subjects were evaluated to a graded exercise test, and oxygen uptake and fat oxidation rate were detected. Data show that in T2D patients fat oxidation rate is not impaired and the positive linear correlation between FATmax and both VO2 and VO2max suggests that even in T2D patients the muscle oxidative capacity might increase in…

Effect of ion exchange capacity and water uptake on hydroxide transport in PSU-TMA membranes: A DFT and molecular dynamics study

2020

Abstract Anion Exchange Membranes (AEMs) are nowadays used in a very wide range of different applications, from electrodialytic systems for water treatment, to Fuel Cells for energy generation. In many of these cases, the AEM is in contact with water molecules, either in aqueous or gas phase, which can strongly interact with the AEM polymer, thus affecting membrane properties. The role of water in fully swollen AEM and its effect on the diffusion of hydroxide is the focus of this work. Density Functional Theory (DFT) based calculations were merged with Molecular Dynamics (MD) simulations coming to a more accurate description of the ionic exchange capacity effect on the hydroxide ion diffusi…

Effects of arbuscular mycorrhizal symbiosis on growth and N2 fixation of Trifolium alexandrinum under late drought stress conditions.

2010

Several experiments, mostly carried out under controlled conditions, have shown that arbuscular mycorrhizal (AM) symbiosis enhances nutrient uptake and improves drought tolerance of host plants. The present research, carried out in a typical Mediterranean environment, evaluated the effect of AM symbiosis on berseem clover (Trifolium alexandrinum L.) grown in the field under both late drought stress and well-watered conditions. The crop was subjected to repeated defoliation. N2 fixation was estimated using the 15N dilution method and using ryegrass (Lolium multiflorum var. Westervoldicum) as the reference crop. In late drought conditions, AM symbiosis resulted in a significant increase in bi…

Estimation of ambient GABA levels in layer I of the mouse neonatal cortex in brain slices

2010

GABAergic synapses on Cajal–Retzius neurons in layer I of the murine neocortex experience GABAB receptor (GABABR)-mediated tonic inhibition. Extracellular GABA concentration ([GABA]o) that determines the strength of GABABR-mediated inhibition is controlled by GABA transporters (GATs). In this study, we hypothesized that the strength ofpresynaptic GABABR activation reflects [GABA]o in the vicinity of synaptic contacts. Slices obtained from two age groups were used, namely postnatal days (P)2–3 and P5–7. GABAergic postsynaptic currents (IPSCs) were recorded using the whole-cell patch-clamp technique. Minimal electrical stimulation in layer I was applied to elicit evoked IPSCs (eIPSCs) using a…

Uridine uptake inhibition as a cytotoxicity test for a human hepatoma cell line (HepG2 cells): comparison with the neutral red assay

2001

International audience; This study describes a sensitive microassay for measuring cytotoxicity based on the degree of inhibition of RNA synthesis in HepG2 cells. RNA synthesis is measured by the kinetic uptake of radiolabeled uridine. A large number of compounds were tested in a wide range of concentrations. The concentration required to induce 50% inhibition of HepG2 uridine uptake rates (IC50) was determined for each compound and used to rank its potency. These IC50s were compared with IC50s measured with the neutral red assay. 2-acetylaminofluorene, benzo[a]pyrene and methylnitrosourea were not cytotoxic in the neutral red assay. Uridine uptake was always inhibited at lower concentrations…

Genotyping analysis and 18FDG uptake in breast cancer patients: a preliminary research

2013

Background: Diagnostic imaging plays a relevant role in the care of patients with breast cancer (BC). Positron Emission Tomography (PET) with 18F-fluoro-2-deoxy-D-glucose (FDG) has been widely proven to be a clinical tool suitable for BC detection and staging in which the glucose analog supplies metabolic information about the tumor. A limited number of studies, sometimes controversial, describe possible associations between FDG uptake and single nucleotide polymorphisms (SNPs). For this reason this field has to be explored and clarified. We investigated the association of SNPs in GLUT1, HIF-1a, EPAS1, APEX1, VEGFA and MTHFR genes with the FDG uptake in BC. Methods: In 26 caucasian individu…
