Identification of an Immunogenic Medulloblastoma-Specific Fusion Involving EPC2 and GULP1

2021

Medulloblastoma is the most common malignant brain tumor in children. Immunotherapy is yet to demonstrate dramatic results in medulloblastoma, one reason being the low rate of mutations creating new antigens in this entity. In tumors with low mutational burden, gene fusions may represent a source of tumor-specific neoantigens. Here, we reviewed the landscape of fusions in medulloblastoma and analyzed their predicted immunogenicity. Furthermore, we described a new in-frame fusion protein identified by RNA-Seq. The fusion involved two genes on chromosome 2 coding for the enhancer of polycomb homolog 2 (EPC2) and GULP PTB domain containing engulfment adaptor 1 (GULP1) respectively. By qRT-PCR …

Cell Systems Bioelectricity: How Different Intercellular Gap Junctions Could Regionalize a Multicellular Aggregate

2021

Simple Summary Electric potential patterns across tissues are instructive for development, regeneration, and tumorigenesis because they can influence transcription, migration, and differentiation through biochemical and biomechanical downstream processes. Determining the origins of the spatial domains of distinct potential, which in turn decide anatomical features such as limbs, eyes, brain, and heart, is critical to a mature understanding of how bioelectric signaling drives morphogenesis. We studied theoretically how connexin proteins with different voltage-gated gap junction conductances can maintain multicellular regions at distinct membrane potentials. We analyzed a minimal model that i…

Heterogeneous Photocatalysis and Photoelectrocatalysis: From Unselective Abatement of Noxious Species to Selective Production of High-Value Chemicals

2015

Heterogeneous photocatalysis and photoelectrocatalysis have been considered as oxidation technologies to abate unselectively noxious species. This article focuses instead on the utilization of these methods for selective syntheses of organic molecules. Some promising reactions have been reported in the presence of various TiO2 samples and the important role played by the amorphous phase has been discussed. The low solubility of most of the organic compounds in water limits the utilization of photocatalysis. Dimethyl carbonate has been proposed as an alternative green organic solvent. The recovery of the products by coupling photocatalysis with pervaporation membrane technology seems to be a…

Shining castles and humans of metal/floral appearance ? metaphorical language in the Palaiologan romances Kallimachos and Velthandros

2019

About eight centuries after Heliodorus, the Greek novel resurfaced in the twelfth century, in Komnenian Byzantium, and again two centuries later under the Palaiologan dynasty. This latter literary revival was due to the political stability of the imperial Byzantine government, which promoted cultural production, rhetorical education, and patronage networks. Kallimachos and Velthandros, two Palaiologan romances presented as court literature, combine ancient and medieval tropes with rhetorical artistry to blur the boundaries between the artificial and the natural. Castles and objects made of precious metals thus resemble living, natural spaces, and human characters are portrayed in metallurgi…

Can the plasma PD-1 levels predict the presence and efficiency of tumor-infiltrating lymphocytes in patients with metastatic melanoma?

2019

Background: The immune response in melanoma patients is locally affected by presence of tumor-infiltrating lymphocytes (TILs), generally divided into brisk, nonbrisk, and absent. Several studies have shown that a greater presence of TILs, especially brisk, in primary melanoma is associated with a better prognosis and higher survival rate. Patients and Methods: We investigated by enzyme-linked immunosorbent assay (ELISA) the correlation between PD-1 levels in plasma and the presence/absence of TILs in 28 patients with metastatic melanoma. Results: Low plasma PD-1 levels were correlated with brisk TILs in primary melanoma, whereas intermediate values correlated with the nonbrisk TILs, and hig…

Association of Maternal Secretor Status and Human Milk Oligosaccharides With Milk Microbiota: An Observational Pilot Study

2019

Background and Objectives: Breast milk contains several bioactive factors including oligosaccharides (HMO) and microbes that shape the infant gut microbiota. HMO profile is determined by secretor status, however their influence on milk microbiota is still uncovered. This study is aimed to determine the impact of the FUT2 genotype on the milk microbiota during the first month of lactation and the association with HMO. Methods: Milk microbiota from 25 healthy lactating women was determined by quantitative PCR and 16S gene pyrosequencing. Secretor genotype was obtained by PCR-RFLPs and by HMO identification and quantification. Results: The most abundant bacteria were Staphylococcus and Strepto…

Microbial deposits in the aftermath of the end-Permian mass extinction: A diverging case from the Mineral Mountains (Utah, USA)

2015

40 pages; International audience; The Lower Triassic Mineral Mountains area (Utah, USA) preserves diversified Smithian and Spathian reefs and bioaccumulations that contain fenestral-microbialites and various benthic and pelagic organisms. Ecological and environmental changes during the Early Triassic are commonly assumed to be associated with numerous perturbations (productivity changes, acidifica-tion, redox changes, hypercapnia, eustatism and temperature changes) post-dating the Permian–Triassic mass extinction. New data acquired in the Mineral Mountains sediments provide evidence to decipher the relationships between depositional environments and the growth and distribution of microbial …

Containment of a Large SARS-CoV-2 Outbreak Among Healthcare Workers in a Pediatric Intensive Care Unit.

2020

Objective Healthcare workers (HCWs) are particularly exposed SARS-CoV-2 because they are critical in preventing viral transmission and treating COVID-19 patients. Within HCWs, personnel of intensive care units (ICUs) are at the forefront of treating patients with a severe course of COVID-19 infection and therefore represent an extremely vulnerable group. Thus, our objective is to contribute to establish means of infection control protecting HCWs in the frontline of the current pandemic. Design An outbreak of SARS-CoV-2 was detected and contained in a pediatric ICU (PICU). The first positive case was identified with a point-of-care diagnostic system on site. Real-time PCR-based testing syste…
