Study of the role of “gatekeeper” mutations V654A and T670I of c-kit kinase in the interaction with inhibitors by means mixed molecular dynamics/dock…

2011

The over-expression of c-kit proto-oncogene has been reported in hematopoietic cells, small cell lung cancer, and gastrointestinal stromal tumors. The clinical importance of c-kit expression in tumors focused the research towards inhibitors of this tyrosine kinase. Imatinib (Gleevec®) was the first compound used in therapy, but mutations on c-kit led to reduced effectiveness or ineffectiveness of this treatment. Other compounds are likely to be effective against mutants, such as Sunitinib (Sutent®), but the need for new and most effective inhibitors against mutants is still critical. We report mixed Molecular Dynamics/Docking study with the aim to unveil the molecular mechanism involved in …

A Regulatory Mechanism Involving TBP-1/Tat-Binding Protein 1 and Akt/PKB in the Control of Cell Proliferation

2011

Abstract TBP-1 /Tat-Binding Protein 1 (also named Rpt-5, S6a or PSMC3) is a multifunctional protein, originally identified as a regulator of HIV-1-Tat mediated transcription. It is an AAA-ATPase component of the 19S regulative subunit of the proteasome and, as other members of this protein family, fulfils different cellular functions including proteolysis and transcriptional regulation. We and others reported that over expression of TBP-1 diminishes cell proliferation in different cellular contexts with mechanisms yet to be defined. Accordingly, we demonstrated that TBP-1 binds to and stabilizes the p14ARF oncosuppressor increasing its anti-oncogenic functions. However, TBP-1 restrains cell…

Noetherian type in topological products

2010

The cardinal invariant "Noetherian type" of a topological space $X$ (Nt(X)) was introduced by Peregudov in 1997 to deal with base properties that were studied by the Russian School as early as 1976. We study its behavior in products and box-products of topological spaces. We prove in Section 2: 1) There are spaces $X$ and $Y$ such that $Nt(X \times Y) < \min\{Nt(X), Nt(Y)\}$. 2) In several classes of compact spaces, the Noetherian type is preserved by the operations of forming a square and of passing to a dense subspace. The Noetherian type of the Cantor Cube of weight $\aleph_\omega$ with the countable box topology, $(2^{\aleph_\omega})_\delta$, is shown in Section 3 to be closely related …

An epistatic mini-circuitry between the transcription factors Snail and HNF4α controls liver stem cell and hepatocyte features exhorting opposite reg…

2011

Preservation of the epithelial state involves the stable repression of epithelial-to-mesenchymal transition program, whereas maintenance of the stem compartment requires the inhibition of differentiation processes. A simple and direct molecular mini-circuitry between master elements of these biological processes might provide the best device to keep balanced such complex phenomena. In this work, we show that in hepatic stem cell Snail, a transcriptional repressor of the hepatocyte differentiation master gene HNF4α, directly represses the expression of the epithelial microRNAs (miRs)-200c and-34a, which in turn target several stem cell genes. Notably, in differentiated hepatocytes HNF4α, p…

Constrained differential inclusions with nonlocal initial conditions

2017

International audience; We show existence for the perturbed sweeping process with nonlocal initial conditions under very general hypotheses. Periodic, anti-periodic, mean value and multipoints conditions are included in this study. We give abstract results for differential inclusions with nonlocal initial conditions through bounding functions and tangential conditions. Some applications to differential complementarity systems and to vector hysteresis are given.

Characterization of the Clarke regularity of subanalytic sets

2017

International audience; In this note, we will show that for a closed subanalytic subset $A \subset \mathbb{R}^n$, the Clarke tangential regularity of $A$ at $x_0 \in A$ is equivalent to the coincidence of the Clarke's tangent cone to $A$ at $x_0$ with the set \\$$\mathcal{L}(A, x_0):= \bigg\{\dot{c}_+(0) \in \mathbb{R}^n: \, c:[0,1]\longrightarrow A\;\;\mbox{\it is Lipschitz}, \, c(0)=x_0\bigg\}.$$Where $\dot{c}_+(0)$ denotes the right-strict derivative of $c$ at $0$. The results obtained are used to show that the Clarke regularity of the epigraph of a function may be characterized by a new formula of the Clarke subdifferential of that function.

Differential inclusions involving normal cones of nonregular sets in Hilbert spaces

2017

This thesis is dedicated to the study of differential inclusions involving normal cones of nonregular sets in Hilbert spaces. In particular, we are interested in the sweeping process and its variants. The sweeping process is a constrained differential inclusion involving normal cones which appears naturally in several applications such as elastoplasticity, electrical circuits, hysteresis, crowd motion, etc. This work is divided conceptually in three parts: Study of positively alpha-far sets, existence results for differential inclusions involving normal cones and characterizations of Lyapunov pairs for the sweeping process. In the first part (Chapter 2), we investigate the class of positivel…

Optimal recovery of a radiating source with multiple frequencies along one line

2020

We study an inverse problem where an unknown radiating source is observed with collimated detectors along a single line and the medium has a known attenuation. The research is motivated by applications in SPECT and beam hardening. If measurements are carried out with frequencies ranging in an open set, we show that the source density is uniquely determined by these measurements up to averaging over levelsets of the integrated attenuation. This leads to a generalized Laplace transform. We also discuss some numerical approaches and demonstrate the results with several examples.

MODY izraisošo mutāciju detektēšana, izmantojot eksomu sekvenēšanu

2015

MODY (Maturity Onset Diabetes of the Young) ir monogēnā cukura diabēta veids, kam raksturīga autosomāli dominanta pārmantošana un simptomu izpausme pirms 25 gadu vecuma. Mutācija kādā no trim gēniem – HNF1α, HNF4α un GCK, sastāda 70-80% MODY gadījumu, tomēr pastāv vēl vismaz 10 gēni, kuros radušās mutācijas spēj izraisīt MODY simptomus.
