Relationship between Thymidylate synthase and p53 and response versus taxane adjuvant chemotherapy for breast carcinoma
2011
Many drugs can be used for adjuvant therapy of breast cancer, including anthracyclines, cyclophosphamide, 5-fluorouracil (5-fU) and, recently, taxanes (TXT) have shown promising results. 5-FU blocks thymidylate synthase (TS) which cross-links p53 mRNA, inhibiting its synthesis. TS overexpression is one of the main mechanisms involved in 5-FU drug resistance. Enough p53 mutations can confer resistance to chemotherapy using anthracyclines and 5-FU, while are associated with improved responses to TXT. The aim of this study was to examine the TS and p53 levels in tumor samples and to compare the efficacy of FEC (5-FU, epirubicin, cyclophosphamide) and TXT chemotherapy in a group of patients wit…
Bypass of G1 arrest induced by DNMT1 posttranscriptional silencing triggers aneuploidy in human cells.
2010
Aneuploidy is a major source of genomic instability in cancer, resulting from chromosome segregation errors caused by defects in genes controlling correct mitotic spindle assembly, centrosome duplication and cell cycle checkpoints. Interestingly in aneuploid cells some of these genes, although not mutated, were underexpressed suggesting the involvement of epigenetic alterations. DNA methylation and histone modifications are the main epigenetic modifications occurring in cells. DNA methyl-transferase 1 (Dnmt1) is known to restore DNA methylation patterns during cell divisions. We investigated the effects of DNMT1 silencing by RNA-interference on the generation of aneuploidy in primary human …
Digital control circuitry for the p53 dynamics in cancer cell and apoptosis
2010
Experimental work and theoretical models deduce a "digital" response of the p53 transcription factor when genomic integrity is damaged. The mutual influence of p53 and its antagonist, the Mdm2 oncogene, is closed in feedback. This paper proposes an aerospace architecture for translating the p53/Mdm2/DNA damage network into a digital circuitry in which the optimal control theory is applied for obtaining the requested dynamic evolutions of some considered cell species for repairing a DNA damage. The purpose of this paper is not to improve the analysis of the actual mathematical models but to demonstrate the usefulness of such digital circuitry design to detect and predict the cell species dyn…
The edition of the ms. London, British Library, Harley 3535, cc. 95v-156v is important for the study of Sicilian vulgarisations of farriery treaties. The witness, dati…
The edition of the second treatise of the ms. London, Harley 3535, cc. 41v-95r could be a new contribution to the study of Middle Age vulgarisations…
2018
After the fall of Rhodes in 1522, the Order of St. John needed to recover new headquarters (which were finally established in Malta in 1530) and its reputation as militia Christi. This Religion in arms had now to redraft its chivalrous ideals within the new context of the Reformation and the Catholic Renewal. However, Hospitallers met this challenge partially and late, although they represented themselves as belonging to this glorious Religion. Such a dialectic between practice and theory is proved, for example, by an apologetic book published in 1619 in Italy and Spain, as well as by unpublished instructions in the late seventeenth century directed at the chaplains providing religious supp…
Molecular analysis of TP53, Ki-Ras and P16 methylation status in tissue and plasma of subjects affected by gastrointestinal cancer (GIC)
2007
BACKGROUND: Despite the improvement in detection and surgical therapy in the last years, the outcome of patients affected by colorectal carcinoma (CRC) remains limited by metastatic relapse. The aim of this study was to investigate the presence of free tumor DNA in the plasma of CRC patients in order to understand its possible prognostic role. PATIENTS AND METHODS: Ki-Ras, TP53 mutations and p16(INK4A) methylation status were prospectively evaluated in tumor tissues and plasma of 66 CRC patients. RESULTS: In 50 of the 66 primitive tumor cases (76%) at least one significant alteration was identified in Ki-Ras and/or TP53 and/or p16(INK4A) genes. Eighteen of the 50 patients presented the same…
