Conformational clamping by a membrane ligand activates the EphA2 receptor

2021

AbstractThe EphA2 receptor is a promising drug target for cancer treatment, since EphA2 activation can inhibit metastasis and tumor progression. It has been recently described that the TYPE7 peptide activates EphA2 using a novel mechanism that involves binding to the single transmembrane domain of the receptor. TYPE7 is a conditional transmembrane (TM) ligand, which only inserts into membranes at neutral pH in the presence of the TM region of EphA2. However, how membrane interactions can activate EphA2 is not known. We systematically altered the sequence of TYPE7 to identify the binding motif used to activate EphA2. With the resulting six peptides, we performed biophysical and cell migratio…

Clustering induces a lateral redistribution of α2β1 integrin from membrane rafts to caveolae and subsequent protein kinase C-dependent internalization

2004

Integrin alpha 2 beta 1 mediates the binding of several epithelial and mesenchymal cell types to collagen. The composition of the surrounding plasma membrane, especially caveolin-1- and cholesterol-containing membrane structures called caveolae, may be important to integrin signaling. On cell surface alpha 2 beta 1 integrin was located in the raft like membrane domain, rich in GPI-anchored proteins, rather than in caveolae. However, when antibodies were used to generate clusters of alpha 2 beta 1 integrin, they started to move laterally on cell surface along actin filaments. During the lateral movement small clusters fused together. Finally alpha 2 beta 1 integrin was found inside caveolae …

Cell Susceptibility to Baculovirus Transduction and Echovirus Infection Is Modified by Protein Kinase C Phosphorylation and Vimentin Organization

2013

ABSTRACT Some cell types are more susceptible to viral gene transfer or virus infection than others, irrespective of the number of viral receptors or virus binding efficacy on their surfaces. In order to characterize the cell-line-specific features contributing to efficient virus entry, we studied two cell lines (Ea.hy926 and MG-63) that are nearly nonpermissive to insect-specific baculovirus (BV) and the human enterovirus echovirus 1 (EV1) and compared their characteristics with those of a highly permissive (HepG2) cell line. All the cell lines contained high levels of viral receptors on their surfaces, and virus binding was shown to be efficient. However, in nonpermissive cells, BV and it…

B7/CD28 costimulation of T cells induces a distinct proteome pattern.

2005

Effective immune strategies for the eradication of human tumors require a detailed understanding of the interaction of tumor cells with the immune system, which might lead to an optimization of T cell responses. To understand the impact of B7-mediated costimulation on T cell activation comprehensive proteome analysis of B7-primed T cell populations were performed. Using this approach we identified different classes of proteins in T cells whose expression is either elevated or reduced upon B7-1- or B7-2-mediated CD28 costimulation. The altered proteins include regulators of the cell cycle and cell proliferation, signal transducers, components of the antigen processing machinery, transporters…

Dynamic Test Methods for COTS SRAMs

2014

International audience; In previous works, we have demonstrated the importance of dynamic mode testing of SRAM components under ionizing radiation. Several types of failures are difficult to expose when the device is tested under static (retention) mode. With the purpose of exploring and defining the most complete testing procedures and reveal the potential hazardous behaviors of SRAM devices, we present novel methods for the dynamic mode radiation testing of SRAMs. The proposed methods are based on different word address accessing schemes and data background: Fast Row, Fast Column, Pseudorandom, Adjacent (Gray) and Inverse Adjacent (Gray). These methods are evaluated by heavy ion and atmos…

Sub-Finsler Horofunction Boundaries of the Heisenberg Group

2020

We give a complete analytic and geometric description of the horofunction boundary for polygonal sub-Finsler metrics---that is, those that arise as asymptotic cones of word metrics---on the Heisenberg group. We develop theory for the more general case of horofunction boundaries in homogeneous groups by connecting horofunctions to Pansu derivatives of the distance function.

Perturbed Bernstein-type operators

2018

The present paper deals with modifications of Bernstein, Kantorovich, Durrmeyer and genuine Bernstein-Durrmeyer operators. Some previous results are improved in this study. Direct estimates for these operators by means of the first and second modulus of continuity are given. Also the asymptotic formulas for the new operators are proved.

A priori bounds and multiplicity of solutions for an indefinite elliptic problem with critical growth in the gradient

2019

Let $\Omega \subset \mathbb R^N$, $N \geq 2$, be a smooth bounded domain. We consider a boundary value problem of the form $$-\Delta u = c_{\lambda}(x) u + \mu(x) |\nabla u|^2 + h(x), \quad u \in H^1_0(\Omega)\cap L^{\infty}(\Omega)$$ where $c_{\lambda}$ depends on a parameter $\lambda \in \mathbb R$, the coefficients $c_{\lambda}$ and $h$ belong to $L^q(\Omega)$ with $q>N/2$ and $\mu \in L^{\infty}(\Omega)$. Under suitable assumptions, but without imposing a sign condition on any of these coefficients, we obtain an a priori upper bound on the solutions. Our proof relies on a new boundary weak Harnack inequality. This inequality, which is of independent interest, is established in the gener…

Common fixed points for α-ψ-φ-contractions in generalized metric spaces

2014

We establish some common fixed point theorems for mappings satisfying an α-ψ-ϕcontractive condition in generalized metric spaces. Presented theorems extend and generalize manyexisting results in the literature.
Erratum to “Common fixed points for α-ψ-φ-contractions in generalized metric spaces”
In Example 1 of our paper [V. La Rosa, P. Vetro, Common fixed points for α-ψ-ϕcontractions in generalized metric spaces, Nonlinear Anal. Model. Control, 19(1):43–54, 2014] a generalized metric has been assumed. Nevertheless some mistakes have appeared in the statement. The aim of this note is to correct this situation.
