Stilbenes and resveratrol metabolites improve mitochondrial fatty acid oxidation defects in human fibroblasts
2014
International audience; Background: Inborn enzyme defects of mitochondrial fatty acid beta-oxidation (FAO) form a large group of genetic disorders associated to variable clinical presentations ranging from life-threatening pediatric manifestations up to milder late onset phenotypes, including myopathy. Very few candidate drugs have been identified in this group of disorders. Resveratrol (RSV) is a natural polyphenol with anti-oxidant and anti-inflammatory effects, recently shown to have beneficial metabolic properties in mice models. Our study explores its possible effects on FAO and mitochondrial energy metabolism in human cells, which are still very little documented. Methods: Using cells …
Identification and diagnosis of patients with familial chylomicronaemia syndrome (FCS): Expert panel recommendations and proposal of an "FCS score".
2018
Familial chylomicronaemia syndrome (FCS) is a rare, inherited disorder characterised by impaired clearance of triglyceride (TG)-rich lipoproteins from plasma, leading to severe hypertriglyceridaemia (HTG) and a markedly increased risk of acute pancreatitis. It is due to the lack of lipoprotein lipase (LPL) function, resulting from recessive loss of function mutations in the genes coding LPL or its modulators. A large overlap in the phenotype between FCS and multifactorial chylomicronaemia syndrome (MCS) contributes to the inconsistency in how patients are diagnosed and managed worldwide, whereas the incidence of acute hypertriglyceridaemic pancreatitis is more frequent in FCS. A panel of Eu…
Cyclopropane Fatty Acid Synthase from Oenococcus oeni: Expression in Lactococcus lactis subsp. Cremoris and Biochemical Characterization
2015
Bacterial cyclopropane fatty acid synthases (CFA synthases) catalyze the transfer of a methyl group from S-adenosyl-L-methionine (AdoMet) to the double bond of a lipid chain, thereby forming a cyclopropane ring. CFAs contribute to resistance to acidity, dryness, and osmotic imbalance in many bacteria. This work describes the first biochemical characterization of a lactic acid bacterium CFA synthase. We have overexpressed Oenococcus oeni CFA synthase in E. coli in order to purify the enzyme. The optimum cyclopropanation activity was obtained at pH 5.6 and 35.8 °C. The high K(m) (AdoMet) value obtained (2.26 mM) demonstrates the low affinity of O. oeni enzyme toward the L. lactis subsp. cremo…
Regulation of the synthesis of aryl metabolites by phospholipid sources in the white-rot fungus Bjerkandera adusta
1999
The white-rot basidiomycete Bjerkandera adusta was cultivated in a liquid medium enriched with l-phenylalanine and various phospholipid sources (lecithin, egg yolk and asolectin). Three aromatic metabolites (benzaldehyde, benzyl alcohol and benzoic acid) were produced under these culture conditions. High concentrations of benzaldehyde (404 mg l–1) were obtained when the cultures were supplemented with 10 g lecithin l–1. Benzyl alcohol production was promoted when the strain was grown with 5 or 10 g lecithin l–1. In the absence of or with a low concentration of lecithin (2.5 g l–1), benzoic acid was the major aryl metabolite synthesized. The results presented here indicate that aryl alcohol …
Assessment of the toxicity of cholesterol oxides on retinal pigment epithelial cultured cells
2006
International audience; Purpose: Among the retinal lipid deposits found in AMD (age related macular degeneration), esterified and unesterified cholesterol have been identified. Cholesterol that accumulates in Bruch's membrane could be oxidized and lead to cholesterol oxides known as oxysterols. These compounds have cytotoxic activities on many vascular cell types, and we conducted this study to evaluate the cytotoxics effects of oxysterols on retinal pigment epithelial (RPE) cells. Methods: Human RPE cells (ARPE–19) were cultured and treated by 7–ketocholesterol (7–keto), 7–betahydroxycholesterol (7–beta) and 25–hydroxycholesterol (25–OH). ARPE–19 cells viability was determined with MTT ana…

Biostimulant Potential of Humic Acids Extracted From an Amendment Obtained via Combination of Olive Mill Wastewaters (OMW) and a Pre-treated Organic …
2018
Olive mill wastewaters (OMW) detain contain significant levels of phenolic compounds with antimicrobial/phytotoxic activity and high amounts of undecomposed organic matter that may a high pollutant load that exerts negative effects on soil biology because of significant levels in phenolic compounds with antimicrobial/phytotoxic activity and limited biodegradability, and high amounts of undecomposed organic matter. Among OMW detoxification techniques, those focusing on oxidative degradation of phenolic compounds are relevant those focusing on oxidative degradation of phenolic compounds to reduce their toxicity. The composting (bio-oxidation) process in particular, exploits exothermic oxidati…
The young hard active Sun: soft X-ray irradiation of tryptophan in water solutions
2010
AbstractThe X-ray emission of the young Sun was much harder and intense than today and might have played a significant role in the evolution of complex organics in protoplanetary environments. We investigate the effects of soft X-rays on tryptophan molecules in aqueous solutions at room temperature. As results of the irradiation experiments we detect several light species indicative of fragmentation, together with large molecular structures such as tryptophan dipeptide and tripeptide. Complexification is more evident in H2O solution than in D2O, probably due to isotopic effects. The abundances of peptides depend on the irradiation dose and decrease with increasing energy deposition. Radical…
Enrichment of amino acids by supported liquid membrane extraction using Aliquat 336 as a carrier
1998
Abstract The possible application of Aliquat 336 (trioctylmethyl ammonium chloride) as a carrier for liquid membrane extraction of amino acids was investigated. The extraction is performed from an aqueous donor phase with pH > 11 to an acceptor phase containing a salt solution through a supported liquid membrane with Aliquat 336 as a carrier in the membrane to facilitate amino acid transport. Counter-coupled transport of chloride anions from the acceptor phase to the donor phase is the driving force of the mass transfer in this system. The extraction efficiency depends on the donor phase pH, the carrier concentration in the liquid membrane, the composition of the acceptor phase and the hydr…
Hemp Flour Particle Size Affects the Quality and Nutritional Profile of the Enriched Functional Pasta
2023
The rheological and chemical quality of pasta samples, which were obtained using the durum wheat semolina fortified with the hemp seed solid residue, after oil extraction, sieved at 530 μm (Hemp 1) or 236 μm (Hemp 2) at different percentages of substitution (5%, 7.5%, and 10%, were evaluated. The total polyphenolic content in hemp flour was quantified in the range of 6.38–6.35 mg GAE/g, and free radical scavenging was included in the range from 3.94–3.75 mmol TEAC/100 g in Hemp 1 and Hemp 2, respectively. The phenolic profiles determined by UHPLC-ESI/QTOF-MS showed that cannabisin C, hydroxycinnamic and protocatechuic acids were the most abundant phenolic compounds in both…
