LC–MS Based Cleavage Site Profiling of the Proteases ADAM10 and ADAM17 Using Proteome-Derived Peptide Libraries
2014
A Disintegrin and Metalloproteinase 10 (ADAM10) and ADAM17 catalyze ectodomain shedding of a number of cell surface proteins important for embryonic development and tissue homeostasis. Changes in the expression levels or dysregulated proteolytic activity of ADAM10 and ADAM17 have been shown to play important roles in multiple diseases such as inflammation, cancer, and neurodegenerative disorders. Despite the well documented substrate repertoire of ADAM10 and ADAM17, little is known about their cleavage site specificity. We optimized Q-PICS (Quantitative Proteomics for the Identification of Cleavage Sites) to elucidate the cleavage site specificity of recombinant murine ADAM10 and ADAM17. Tw…
The substrate degradome of meprin metalloproteases reveals an unexpected proteolytic link between meprin β and ADAM10
2012
The in vivo roles of meprin metalloproteases in pathophysiological conditions remain elusive. Substrates define protease roles. Therefore, to identify natural substrates for human meprin α and β we employed TAILS (terminal amine isotopic labeling of substrates), a proteomics approach that enriches for N-terminal peptides of proteins and cleavage fragments. Of the 151 new extracellular substrates we identified, it was notable that ADAM10 (a disintegrin and metalloprotease domain-containing protein 10)—the constitutive α-secretase—is activated by meprin β through cleavage of the propeptide. To validate this cleavage event, we expressed recombinant proADAM10 and after preincubation with meprin…

On symplectically rigid local systems of rank four and Calabi–Yau operators
2013
AbstractWe classify all Sp4(C)-rigid, quasi-unipotent local systems and show that all of them have geometric origin. Furthermore, we investigate which of those having a maximal unipotent element are induced by fourth order Calabi–Yau operators. Via this approach, we reconstruct all known Calabi–Yau operators inducing an Sp4(C)-rigid monodromy tuple and obtain closed formulae for special solutions of them.
Hadamard-type theorems for hypersurfaces in hyperbolic spaces
2006
Abstract We prove that a bounded, complete hypersurface in hyperbolic space with normal curvatures greater than −1 is diffeomorphic to a sphere. The completeness condition is relaxed when the normal curvatures are bounded away from −1. The diffeomorphism is constructed via the Gauss map of some parallel hypersurface. We also give bounds for the total curvature of this parallel hypersurface.
Some fourth order CY-type operators with non symplectically rigid monodromy
2012
We study tuples of matrices with rigidity index two in $\Sp_4(\mathbb{C})$, which are potentially induced by differential operators of Calabi-Yau type. The constructions of those monodromy tuples via algebraic operations and middle convolutions and the related constructions on the level differential operators lead to previously known and new examples.
Improved Bounds for Hermite–Hadamard Inequalities in Higher Dimensions
2019
Let $\Omega \subset \mathbb{R}^n$ be a convex domain and let $f:\Omega \rightarrow \mathbb{R}$ be a positive, subharmonic function (i.e. $\Delta f \geq 0$). Then $$ \frac{1}{|\Omega|} \int_{\Omega}{f dx} \leq \frac{c_n}{ |\partial \Omega| } \int_{\partial \Omega}{ f d\sigma},$$ where $c_n \leq 2n^{3/2}$. This inequality was previously only known for convex functions with a much larger constant. We also show that the optimal constant satisfies $c_n \geq n-1$. As a byproduct, we establish a sharp geometric inequality for two convex domains where one contains the other $ \Omega_2 \subset \Omega_1 \subset \mathbb{R}^n$: $$ \frac{|\partial \Omega_1|}{|\Omega_1|} \frac{| \Omega_2|}{|\partial \Ome…
The hermeneutic self-overcoming of culture in interculturalism. Toward an intercultural reading of Hans-Georg Gadamer’s philosophical hermeneutics
2017
ABSTRACT The objective of the article is to analyze the philosophical hermeneutics of HansGeorg Gadamer in order to explore the possibility of an intercultural reading based on his proposa…
