A concise route to MK-4482 (EIDD-2801) from cytidine.

2020

A two-step route to MK-4482 (EIDD-2801, 1) was developed consisting of an esterification and hydroxamination of cytidine. The selective acylation and direct amination eliminate the need for protecting and activating groups and proceed in overall yield of 75%, a significant advancement over the reported yield of 17%. The step count is reduced from five transformations to two, and expensive uridine is replaced with the more available cytidine.

An aryl-fused redox-active tetrathiafulvalene with enhanced mixed-valence and radical-cation dimer stabilities.

Molecular recognition of stable organic radicals is a relatively novel, but important structural binding motif in supramolecular chemistry. Here, we report on a redox-switchable veratrole-fused tetrathiafulvalene derivative VTTF which is ideally suited for this purpose and for the incorporation into stimuli-responsive systems. As revealed by electrochemistry, UV/Vis measurements, X-ray analysis, and electrocrystallisation, VTTF can be reversibly oxidised to the corresponding radical-cation or dication which shows optoelectronic and structural propterties similar to tetrathiafulvalene and tetrakis(methylthio)tetrathiafulvalene. The reaction of [Ni(COD) 2 ] with [Cp* 2 NbTe 2 H] (1; Cp * = η-C 5 Me 5 ) in the presence of Ph 2 PCH 2 PPh 2 (dppm) in boiling toluene gives black-violet [(Cp * 2 Nb) 2 NiTe 4 ] (3). If [Cp' 2 NbTe 2 H] (2; Cp' = tBuC 5 H 4 ) is used under similar conditions dark-brown [(Cp'Nb)2Ni5Te7(dppm)2] (4) is formed. The structures of 3 and 4 have been determined crystallographically. Complex 3 contains a severely distorted NiTe 4 tetrahedron to which two niobocene groups are coordinated. Density functional analysis of the electronic structure of the NiTe 4 building block shows that it is best described as an [Ni(η 2 -Te 2 ) 2 ] 2- fragment. Nucleophilic iodonium interactions (NIIs) in 2-coordinate iodine(i) and silver(i) complexes

The generality of nucleophilic iodonium interactions (NIIs) has been demonstrated by preparing a range of silver(i) and iodonium (I+) complexes and studying their 15N NMR chemical shifts, with the first example of a NII-complex involving a 2-coordinate silver(i) complex being confirmed by X-ray crystallography, and its nucleophilicity studied by DFT calculations.

Structural studies of homoisoflavonoids: NMR spectroscopy, X-ray diffraction, and theoretical calculations

Abstract In this article we present a detailed structural investigation for five homoisoflavonoids, molecules important from the pharmacological point of view. For studying the electron distribution as well as its influence on the physicochemical properties, NMR spectroscopy, X-ray diffraction, and theoretical calculations have been used. Nuclear magnetic shieldings obtained by using DFT calculations for optimized molecular geometries are correlated with the experimentally determined chemical shifts. Abstract Reactions of the [Fe III (CN) 6 ] 3− anion with the [Cu II (tn)] 2+ ion (tn = 1,3-diaminopropane) afford the compounds {[Cu II (tn)] 2 [Fe II (CN) 6 ]}3·[Na 3 Fe III (CN) 6 ]·12H 2 O (1) and {[Cu n (tn)] 2 [Fe II (CN) 6 ]}·KCl·5H 2 O (2). Abstract New iron(II) coordination polymeric neutral chain of formula [Fe(abpt) 2 (μ-M(CN) 4 )], with M = Pt II ( 1 ), Ni II ( 2 ) and abpt = 4-amino-3,5-bis(pyridin-2-yl)-1,2,4-triazole, have been synthesized and characterized by infrared spectroscopy, X-ray diffraction and magnetic measurements. The two compounds are isostructural as deduced from a Rietveld analysis of X-ray powder diffraction data of 2 simulated from the single crystal structure of 1 . A spiro-fused hexa-peri-hexabenzocoronene dimer was synthesized, and its physicochemical properties were studied by UV-Vis absorption and emission spectroscopy as well as cyclic voltammetry. Chemical oxidation of SB-HBC afforded its radical cation and dication derivatives, which could be reversibly reduced to the neutral state.

Structural Tuning and Conformational Stability of Aromatic Oligoamide Foldamers

A series of aromatic oligoamide foldamers with two or three pyridine-2,6-dicarboxamide units as their main folding motifs and varying aromatic building blocks as linkers have been synthetized to study the effects of the structural variation on the folding properties and conformational stability. Crystallographic studies showed that in the solid state the central linker unit either elongates the helices and more open S-shaped conformations, compresses the helices to more compact conformations or acts as a rigid spacer separating the pyridine-2,6-dicarboxamide units, which for their part add the predictability of the conformational properties.
