Reduced graphene oxide decorated with metals nanoparticles electrode as electrochemical sensor for dopamine

2019

Dopamine (DA) is one of the most important neurotransmitters that influences the processes that involve memory, sleep, mood, learning among others [1]. In fact, in the last years, dopamine concentration in human body fluids has been related to some neurodegenerative diseases, such as Parkinson and Alzheimer's diseases [2]. The possibility to have a bio-marker for these disease is of extreme importance because, disease related with dementia, are diagnosed when they are already developed and their management become almost impossible. The possibility to continuously monitor DA level in fluids, such as blood and urine, could accelerate the early diagnosis of these diseases. The principal analyt…

Kontroll-Bedingungen untersucht. Die Verum-Gruppe erhält ein spezifisches sportliches Trainingsprogramm. In der Kontrollgruppe werden lediglich…

Role of physical activity in ameliorating neuropsychiatric symptoms in Alzheimer disease: A narrative review

2019

Objective: Neuropsychiatric symptoms (NPs) affect almost all patients with Alzheimer disease (AD). Because of the complications associated with the pharmacological treatment, nonpharmacological treatment (such as physical activity) can be considered as an additional complementary treatment option for NPs. The aim of this review is to evaluate the impact of physical activity on NPs in patients with AD. Methods: We searched Pubmed and Google Scholar for potential eligible articles until March 1, 2018. Results: Although there are contradictory results showing the impact of physical exercise on NPs, most of them reported that it had a significant effect on depression and sleep disturbances in p…

The neuroprotective actions of corticotropin releasing hormone

2005

Corticotropin-releasing hormone (CRH) modulates the activity of the hypothalamic-pituitary-adrenal (HPA) axis, and has a key role in mediating neuroendocrine effects that occur in response to stressful stimuli. Disruption of the CRH system however has been shown to be closely associated with the progression of Alzheimer's disease (AD), and these observations prompted an investigation into the potential neuroprotective effects of the hormone. In addition to its regulatory affects on the molecular processes that underlie AD i.e., amyloid precursor protein (APP) processing and potentially tau phosphorylation, evidence is provided that the neuroprotective effects of CRH are mediated by a number…

High turnover rate of central histaminergic system in patients with Down syndrome and Alzheimer disease

2013

HIGH TURNOVER RATE OF CENTRAL HISTAMINERGIC SYSTEM IN PATIENTS WITH DOWN SYNDROME AND ALZHEIMER DISEASE Maria Concetta Gueli Dipartimento di Biomedicina Sperimentale e Neuroscienze Cliniche (BioNEC), Università degli Studi di Palermo It is well confirmed that a strong relationship exists between Down’s syndrome (DS) and Alzheimer’s disease (AD). Neurochemical investigations reported that many central neurotransmitter systems are similarly affected in aging Down and in Alzheimer patients, respectively. Airaksinem et al. (1) found numerous neurofibrillary tangles in the tuberomammillary area of the hypothalamus, where cell bodies of histaminergic neurons are located. While Mazurkiewicz-Kwilec…

Impact of the Relationship of Stress and the Immune System in the Appearance of Alzheimer’s Disease

2016

The understanding of how the immune system works, as well as its relationship with the stress level, seems to be important at the start of the Alzheimer’s disease (AD). To analyze this, immunoglobulin A (IgA) and cortisol in saliva were measured using ELISA in patients with mild AD and healthy volunteers, and the production of both biomarkers was compared and correlated. In participants without AD, IgA was higher when cortisol was lower, and the opposite happened in participants with AD, with the quantification in saliva being a suitable method to determine it. Neuronal activity and secreted amyloid β lead to altered amyloid β precursor protein and presenilin 1 interactions.

2013

Deposition of amyloid β (Aβ) containing plaques in the brain is one of the neuropathological hallmarks of Alzheimer's disease (AD). It has been suggested that modulation of neuronal activity may alter Aβ production in the brain. We postulate that these changes in Aβ production are due to changes in the rate-limiting step of Aβ generation, APP cleavage by γ-secretase. By combining biochemical approaches with fluorescence lifetime imaging microscopy, we found that neuronal inhibition decreases endogenous APP and PS1 interactions, which correlates with reduced Aβ production. By contrast, neuronal activation had a two-phase effect: it initially enhanced APP-PS1 interaction leading to increased …

Immunosenescence, inflammation and Alzheimer’s disease

2012

Abstract Ageing impacts negatively on the development of the immune system and its ability to fight pathogens. Progressive changes in the T-cell and B-cell systems over the lifespan of individuals have a major impact on the capacity to respond to immune challenges. The cumulative age-associated changes in immune competence are termed immunosenescence that is characterized by changes where adaptive immunity deteriorates, while innate immunity is largely conserved or even upregulated with age. On the other hand, ageing is also characterized by “inflamm-ageing”, a term coined to explain the inflammation commonly present in many age-associated diseases. It is believed that immune inflammatory p…

Action of low doses of Aspirin in Inflammation and Oxidative Stress induced by aβ

2019

Aspirin has been used as anti-inflammatory and anti-aggregate for decades but the precise mechanism(s) of action after the presence of the toxic peptide Aβ1-42 in cultured astrocytes remains poorly resolved. Here we use low-doses of aspirin (10-7 M) in astrocytes in primary culture in presence or absence of Aβ1-42 toxic peptide. We noted an increase of cell viability and proliferation with or without Aβ1-42 peptide presence in aspirin treated cells. In addition, a decrease in apoptosis, determined by Caspase 3 activity and the expression of Cyt c and Smac/Diablo, were detected. Also, aspirin diminished necrosis process (LDH levels), pro-inflammatory mediators (IL-β and TNF-α) and NF-ᴋB prot…
