2009

The combination of irinotecan with 5-fluorouracil demonstrates efficacy with tolerable safety in the first-line treatment of metastatic gastroesophageal cancer (mGC). This randomized phase II trial compared for the first time capecitabine with irinotecan or cisplatin in this setting.Patients were randomly assigned to receive 3-week cycles of capecitabine 1000 mg/m(2), twice daily for 14 days, with on day 1 either irinotecan 250 mg/m(2) (XI) or cisplatin 80 mg/m(2) (XP). The primary end point was overall response rate (ORR) and secondary end points included progression-free survival (PFS), overall survival (OS) and safety.Of 118 patients recruited, 112 were eligible for safety analysis and 1…

Induction of apoptosis in human retinoblastoma cells by topoisomerase inhibitors

1998

PURPOSE:To examine the apoptotic effect induced in human retinoblastoma Y79 cells by camptothecin, etoposide, and amsacrine, to examine the effect of these drugs on the expression of many apoptosis-related modulators, and to test the antiapoptotic effect exerted by insulin-like growth factor-I (IGF-I). METHODS:Morphologic features of apoptosis were demonstrated using acridine orange- ethidium bromide staining and electron microscopy. DNA fragmentation was determined by means of an in situ cell detection procedure (TdT-dUTP terminal nick-end labeling [TUNEL]) or by electrophoresis on agarose gels and was quantified by enzyme-linked immunosorbent assay. The expression of apoptosis-related mod…

Apoptotic effects of different drugs on cultured retinoblastoma Y79 cells

1998

This paper deals with the apoptotic effect exerted in human retinoblastoma Y79 cells by a number of compounds. A remarkable effect was observed after treatment with DNA-damaging agents, such as camptothecin, etoposide, cisplatin and carboplatin; camptothecin was found to be the most efficacious. Treatment with these compounds induced the appearance of morphological features of apoptosis in the cells together with the distinct fragmentation of DNA, as shown by agarose gel electrophoresis. These effects were also accompanied by a remarkable increase in the level of p53. Many other compounds, which are not DNA-damaging agents, induced the morphological features of apoptosis but none of them we…

Continuously manufactured magnetic polymersomes--a versatile tool (not only) for targeted cancer therapy.

2013

Micromixer technology was used to prepare polymeric vesicles (Pluronic® L-121) dual loaded with the anti-cancer drug camptothecin and magnetic nanoparticles. Successful incorporation of the magnetic nanoparticles was confirmed by transmission electron microscopy. Dynamic light scattering measurements showed a relatively narrow size distribution of the hybrid polymersomes. Camptothecin polymersomes reduced the cell viability of prostate cancer cells (PC-3) measured after 72 h significantly, while drug-free polymersomes showed no cytotoxic effects. Covalent attachment of a cancer targeting peptide (bombesin) as well as a fluorescent label (Alexa Fluor® 647) to the hybrid polymersomes was perf…

Effects of eutrophication and organic loading on the occurrence of profundal harpacticoids in a lake in southern Finland.

1992

Harpacticoids made up 19 and 30% of the meiobenthos proper by number and 39 and 55% by biomass at maximum depths and a depth of 20 m, respectively, in Lake Paijanne. Harpacticoids accounted for higher proportions of the total meiobenthos in oligotrophic and unpolluted areas than in polluted areas. Seven species were identified, with Canthocamptus staphylinus confined to semi-lotic areas influenced by organic pollution from a pulp mill. Other species exhibited somewhat different distributions in oligotrophic and mesotrophic areas. Harpacticoid densities were most closely correlated with oxygen content and phytoplankton biomass, and correlations with environmental values were closer at maximu…

The apoptotic effects of cisplatin and carboplatin in retinoblastoma Y79 cells.

1998

This study demonstrated that cisplatin and carboplatin stimulate apoptosis in human retinoblastoma Y79 cells, cisplatin being the most effective compound. The apoptotic effect appeared after 8 h and then increased in a time-dependent manner. Treatment with cisplatin and carboplatin also provoked an increase in the level of p53 and p21, and a lowering in Bcl-2. The prolonged exposure of Y79 cells to cisplatin induced resistance to cisplatin, carboplatin and etoposide. The basal level of p53 was in resistant cells higher than in untreated cells, while Bcl-2 was not modified. p53 and Bcl-2 levels did not change after treating of resistant cells with cisplatin, carboplatin or etoposide. However…

Lacustrine profundal meiobenthos as an environmental indicator

1992

Organic loading and eutrophy is indicated at profundal depths by large numbers of resting copepodid stages of cyclopoid copepods, by the occurrence of the naidid oligochaete species, Amphichaeta leydigii and Specariajosinae, and the harpacticoid species Canthocamptus staphylinus, and by a low meiobenthos/ macrobenthos biomass ratio. An oligotrophic environment is indicated by the occurrence of the aeolo-somatid oligochaetes Aeolosoma quaternarium, A. hemprichi and Rheomorpha neiswestnovae, the naidid oligochaete Chaetogaster langi and the harpacticoid species Attheyella crassa and a high meiobenthos/ macrobenthos biomass ratio.
