Repression of Human Papillomavirus Oncogene Expression under Hypoxia Is Mediated by PI3K/mTORC2/AKT Signaling

Oncogenic HPV types are major human carcinogens. Under hypoxia, HPV-positive cancer cells can repress the viral E6/E7 oncogenes and induce a reversible growth arrest. This response could contribute to therapy resistance, immune evasion, and tumor recurrence upon reoxygenation. Here, we uncover evidence that HPV oncogene repression is mediated by hypoxia-induced activation of canonical PI3K/mTORC2/AKT signaling. AKT-dependent downregulation of E6/E7 is only observed under hypoxia and occurs, at least in part, at the transcriptional level. Abstract The collapse of unsaturated soils consists of the reduction in volume upon wetting at constant total stress. Several studies at the macro scale outline the influence of initial void ratio, confining pressure and matric suction on the onset of collapse of standard laboratory specimens. Conversely, few observations at the micro scale are available in the literature, although the influence of the particles arrangement and intergranular bonding has been formerly supposed. Micelles as containers for self-assembled nanodevices: a fluorescent sensor for lipophilicity

Potentiometric titrations, fluorescence versus pH titrations, dynamic light scattering and fluorescence polarization anisotropy studies demonstrate that inside the nanodimensioned Triton X-100 micelles, 1-pyrenecarboxylic acid, PCOO-, forms an apical complex with the Zn2+ cation encircled by a lipophilic cyclen ligand and hugely increasing its fluorescence. The ability of the Zn2+-cyclen-PCOO- complex plus its micellar container to act as a fluorescent sensor to evaluate the lipophilicity of molecular species is demonstrated on the fatty acid series CH 3(CH2)xCOOH (x=0-16). Small cyclic peptides have been employed to elucidate the performance of novel sorbents as stationary phases in capillary electrochromatography (CEC). In this paper chain length dependencies for ordinary liquid chromatographic sorbents are reported together with findings acquired on beads specifically designed to suit CEC. Abstract An enzyme pretreatment and negative-pressure cavitation extraction (E-NPCE) method was investigated for extraction genistein and apigenin from pigeon pea roots. The important parameters involved in E-NPCE process were optimized by single-factor experiments and then critical parameters were investigated by a 2 3 full factorial central composite design (CCD) to optimize extract conditions. Under optimal conditions, the yields of genistein and apigenin were 0.628 mg/g and 0.359 mg/g, which represented an increase of 44.70% and 53.05%, respectively, compared to standard NPCE. Abstract A fungal endophyte MD89 with obvious antioxidant activities was isolated from pigeon pea and identified as Chaetomium globosum by ITS sequence. Different fractions from MD89 culture were compared and evaluated by total phenol (TP) content, total flavonoid (TFL) content, DPPH radical scavenging, reducing power and lipid peroxidation assays, respectively. Results showed that EtOAc extracts had high content of TP and TFL, and strong antioxidant activities (IC 50 value was 6.87, 15.19, 16.78 μg/mL, respectively).
