Abstract We have studied the subcellular localization of pentachlorophenol 4-monooxygenase (PCP4MO) in Sphingobium chlorophenolicum ATCC 39723 during induction by pentachlorophenol (PCP). Using a monoclonal antibody CL6 specific to the native and recombinant PCP4MO, the enzyme was primarily found soluble as determined by immunoblot and ELISA analyses of cellular fractions. However, the enzyme was observed both in the soluble and membrane-bound forms during induction for 2–4 h, suggesting its translocation out from the cytoplasm. International audience; 8-Hydroxypyrene-1,3,6-trisulfonic acid (pyranine) can be used as a vital intracellular pH (pH(i)) indicator. In the yeast Yarrowia lipolytica, a partial efflux of the probe was detected by using the pH-independent wavelength of 415 nm. A simplified correction of the fluorescent signals was applied, enabling to show for this species a good near-neutral pH(i) maintenance capacity in a pH 3.9 medium. Octanoic acid, which is known to have toxic effects on yeast, decreased the pH(i) and increased the 260-nm-absorbing compounds leakage. Ribonucleotide reductase (RNR) is an essential enzyme required for DNA synthesis and repair. Although iron is necessary for class Ia RNR activity, little is known about the mechanisms that control RNR in response to iron deficiency. In this work, we demonstrate that yeast cells control RNR function during iron deficiency by redistributing the Rnr2–Rnr4 small subunit from the nucleus to the cytoplasm. Our data support a Mec1/Rad53-independent mechanism in which the iron-regulated Cth1/Cth2 mRNA-binding proteins specifically interact with the WTM1 mRNA in response to iron scarcity, and promote its degradation. Maģistra darba tēma: Darba vidi ietekmējošie faktori ķirurģijas aprūpes māsu klīniskajā vidē. Tēmas aktualitāte: darba vide ieņem nozīmīgu lomu, un māsas ir pakļautas daudzveidīgu faktoru kombinētai iedarbībai. Pētījums tika veikts vienā no Latvijas X stacionāriem, kurā tika anketētas 100 ķirurģijas aprūpes profila māsās, ar darba mērķi izpētīt darba vides ietekmējošos faktorus ķirurģijas aprūpes māsu klīniskajā vidē. Iegūtie rezultāti apliecina, ka vairākums ķirurģijas aprūpes māsu, kā neapmierinātības iemeslus ar darba apstākļiem, min kaitīgos darba vidi ietekmējošos faktorus - psiholoģisko spriedzi un darbinieku trūkumu. Veikti secinājumi un izstrādāti ieteikumi.
