12S rRNA mitochondrial gene as marker to trace Sicilian mono-species dairy products

Abstract For a rapid, specific and sensitive identification of cows', ewes' and goats' milk in mono-species Sicilian dairy products, species-specific duplex-PCR protocol was applied. DNA samples from blood and experimental cheeses of Sicilian autochthonous breeds were extracted to amplify the 12S rRNA (and part of 16S rRNA in case of Ovis aries ) mitochondrial species-specific gene fragment. The use of species-specific primers for Bos taurus , Capra hircus and Ovis aries species, after electrophoresis on agarose gel, yielded fragments of 256 bp, 326 bp and 172 bp, respectively. Oligonucleotide primers representing promoter and protein motifs in rats and mice were assayed for PCR amplification of ancient DNA from two sheep and one goat. We show preliminary evidence that this type of primers can be used for genomic fingerprinting of ancient DNA at interspecific level and can help in solving some paleoecological promlems.

Summary Climate change along with anthropogenic activities changes biogeochemical conditions in lake ecosystems, modifying the sediment microbial communities. Wastewater effluents introduce nutrients and organic material but also novel microbes to lake ecosystems, simulating forthcoming increases in catchment loadings. This study is focused on the (micro)biogeochemical features of two close geothermal sites (FAV1 and FAV2), both selected at the main exhalative area of Pantelleria Island, Italy. A previous biogeochemical survey revealed high CH4 consumption and the presence of a diverse community of methanotrophs at FAV2 site, whereas the close site FAV1 was apparently devoid of methanotrophs and recorded no CH4 consumption.
