Pseudovirions as Specific Tools for Investigation of Virus Interactions With Cells

This chapter outlines the generation and application of human papillomavirus type 33 (HPV33) pseudovirions. The method describes (1) the construction of vaccinia viruses recombinant for the major and minor HPV capsid proteins, L1 and L2, respectively; (2) the transfection of Cos7 cells with a marker plasmid replicating to high copy numbers; (3) the expression of L1 and L2 using the vaccinia virus expression system; (4) the extraction, purification, and analysis of HPV33 pseudovirions; and (5) their use in pseudoinfection assays. During the analysis of the La (SS-B) autoantigen for catalytic activities an ATP-dependent double-stranded RNA unwinding activity was detected. Both native and recombinant La proteins from different species displayed this activity, which could be inhibited by monospecific anti-La antibodies. La protein was able to melt dsRNA substrates with either two 3'-overhangs or a single 3'- and a 5'-overhang. Double-stranded RNAs with two 5'-overhangs were not unwound, indicating that at least one 3'-overhang is required for unwinding. Tomato yellow leaf curl disease (TYLCD) is well known in Mediterranean countries, where it has been causing severe losses in tomato crops for decades. Until recently, two viruses (with several isolates) in the genus Begomovirus, family Geminiviridae, have been associated with the epidemics: Tomato yellow leaf curl virus (TYLCV) and Tomato yellow leaf curl Sardinia virus (TYLCSV). Since the lampreys died massively from early February onwards, they were investigated in March and May to clarify the cause of the deaths. The symptoms in lampreys resembled those of lipid-related thiamine (vitamin B1) deficiency of salmonines, called the M74 syndrome in the Baltic Sea area. Tässä artikkelissa tarkastellaan pedagogisia käytänteitä, jotka tukevat kiinnittymistä sekä kielen ja sisältöjen rinnakkaista oppimista ammatillisessa koulutuksessa. Tiimietnografinen haastattelu- ja havainnointiaineisto kerättiin ammatillisessa oppilaitoksessa kahtena lukuvuonna. Osallistujat ovat myöhään suomalaiselle koulupolulle tulleita maahanmuuttotaustaisia nuoria aikuisia. Näiden seitsemän opiskelijan lisäksi haastateltiin neljäätoista opetus- ja ohjaushenkilökunnan jäsentä. Volcanic gases give information on magmatic processes. In particular, anomalous releases of carbon dioxide precede volcanic eruptions. Up to now, this gas has been measured in volcanic plumes with conventional measurements that imply the severe risks of local sampling and can last many hours. The electrical conductivity of glass fiber-reinforced plastic (GFRP) with epoxy matrix modified by multiwall carbon nanotubes (MWCNT) was studied. The electrical conductivity of nanomodified lamina and multi-layered GFRP was investigated on several levels using a structural approach. Components of the electrical conductivity tensor for unidirectional-reinforced monolayer were calculated similarly as in micromechanics using the conductivity of the nanomodified matrix. Desde la perspectiva sociojurídica de los estudios de la gobernabilidad, se analiza la evolución de las estrategias de provisión de seguridad, desde su orientación neoliberal hasta el predominio de la austeridad desde la crisis de 2008. Se delimita así un escenario en el que la desigualdad y la aporofobia son elementos fundamentales del marco axiológico hegemónico. Esto es relevante para la configuración y efectividad de los derechos sociales de distintos grupos vulnerables. El artículo se aproxima finalmente a la situación de las empleadas de hogar y de quienes acceden a una vivienda mediante la ocupación de inmuebles vacíos en España.
