DNA oxidation products determined with repair endonucleases in mammalian cells: Types, basal levels and influence of cell proliferation

Purified repair endonucleases such as Fpg protein, endonuclease III and IV allow a very sensitive quantification of various types of oxidative DNA modifications in mammalian cells. By means of these assays, the numbers of base modifications sensitive to Fpg protein, which include 8-hydroxyguanine (8-oxoG), were determined to be less than 0.3 per 10(6) bp in several types of untreated cultured mammalian cells and human lymphocytes and less than 10 per 10(6) bp in mitochondrial DNA from rat and porcine liver. Propyl substituted 4-arylimino-1,2,3-trihydroacridylnickel complexes: Their synthesis, characterization and catalytic behavior toward ethylene

Propyl substituted 4-arylimino-1,2,3-trihydroacridylnickel dihalide complexes were designed and prepared by metal-induced template reaction with NiCl2 center dot 6H(2)O or (DME)NiBr2. They were characterized by infrared spectroscopy and elemental analysis. Single crystal X-ray crystallography of representative complex Ni3 revealed a distorted trigonal bipyramidal geometry around nickel. The catalytic activities of the title nickel complexes were negatively affected by propyl substituent on their backbone when comparing with the results by unsubstituted ones. Abstract We present simulation of the UV–visible spectra of acridine orange, a widely used photosensitizer for in vivo studies due to its highly environment-dependent spectroscopic properties. This dye has been investigated both in its protonated and neutral forms, either isolated or embedded in a pumpkin-shaped macromolecular cycle (cucurbit-7-uril), using time-dependent density functional theory techniques. Sea urchin embryos as a model system for studying autophagy induced by cadmium stress

It is well known that sea urchin embryos are able to activate different defense strategies against stress. We previously demonstrated that cadmium treatment triggers the accumulation of metal in embryonic cells and the activation of defense systems depending on concentration and exposure time, through the synthesis of heat shock proteins and/or the initiation of apoptosis. Here we show that Paracentrotus lividus embryos exposed to Cd adopt autophagy as an additional stratagem to safeguard the developmental program. Abstract The spectrophotometric and fluorimetric determination of 9-amino acridine is carried out by means of the FIA procedures and on the basis of its own color or fluorescence. The methanol used as a carrier stream resulted in a significative increase in sensitivity and sample throughput. The method is applied to the drug determination in pharmaceutical formulations.

A potent inhibitor of serine/threonine kinases, staurosporine exerts antiproliferative and apoptotic effects in many cancer cells, although the exact mechanism of its action is still unclear. This study examines the effects of staurosporine on Chang liver cells, an immortalized non-tumor cell line, in comparison with those caused in HuH-6 and HepG2 cells, two human hepatoma cell lines. Our results provide evidence that staurosporine promotes apoptosis in Chang liver cells as observed by flow cytometric analysis and acridine orange/ethidium bromide staining.
