Target Analogue Imprinted Polymers with Affinity for Folic Acid and Related Compounds

Two approaches to synthesize molecularly imprinted polymers with affinity for folic acid and other substituted pteridines have been compared. In the first approach, the folic acid analogue methotrexate was used as template and functional monomers capable of generating selective binding sites were searched in a miniaturized screening system based on binding assessment in the batch mode. Highest selectivity was seen using 2-vinylpyridine as functional monomer, which was confirmed in the chromatographic mode for a batch synthesized on a gram scale. Die zweifache unterdrückung der polymerisation einiger bis(methacrylsäureester) mittels radikalischer additionsreaktionen

Aus Hydrochinon, Resorcin, Dihydroxydiphenylen und Dihydroxynaphthalinen wurden mit Methacrylsaurechlorid Bis(methacrylsaureester) hergestellt. Setzte man diese in siedendem Benzol mit groser Verdunnung gelosten Ester dem Angriff von Radikalen aus α.α′-Azoisobuttersauredinitril (Primarradikale) aus, dessen Uberschus in Beziehung zum Ester mindestens zwanzigfach molar war, so erhielt man mittels saulenchromatographischer Trennung Verbindungen, bei denen je olefinische Doppelbindung zwei Primarradikale addiert waren (unterdruckte Polymerisation). Highly cross-linked network polymers prepared by molecular imprinting catalyzed enantioselectively the hydrolysis of N-tert-butoxycarbonyl phenylalanine-p-nitrophenyl ester (BOCPheONP). The templates were designed to allow incorporation of the key catalytic elements, found in the proteolytic enzyme chymotrypsin, into the polymer active sites. Three model systems were evaluated. To obtain a carrier polymer for pharmacologically active components, linear polyethyleneimine (LPEI) was chemically modified by reactions with acrylic acid (1a), acrylamide (1b), acrylic acid esters (1c and 1d), sodium chloroacetate, and hydrochloric acid. All product polymers 2a – 2e are soluble in water. Among these, 2a and 2e, β- and α-amino acid derivatives, respectively, show no acute toxicity in mice up to dosages of 1 g/kg i. v. Polyacroleine werden durch verdunntes Wasserstoffperoxyd vollstandig abgebaut. Als Abbauprodukte konnten Kohlendioxyd, Ameisensaure und Essigsaure identifiziert werden. Daneben bilden sich noch unbekannte nicht-fluchtige Saren. Polyacrylsaure erleidet unter den gleichen Bedingungen ebenfalls einen Abbau. Die Zusammensetzung der Abbauprodukte ist eine andere. Durch Peressigsaure werden Polyacroleine zu polymeren Sauren oxydiert. Diese gleichen bezuglich der Loslichkeit, der Reaktionen mit Schwermetallsalzen und dem IR- Spektrum weitgehend der Polyacrylsaure. In constrast with recent molecular orbital calculations on the decarboxylation of O-protonated 2-oxetanone, this experimental work indicates that no decarboxylation of this cation occurs in sulphuric acid solution up to 150°C, but instead a clean isomerization to protonated acrylic acid takes place. Parallel theoretical work shows that the gas-phase model is too crude to account successfully for the experimental facts obtained in acidic media. However, the latter are well reproduced when the effect of the solvent is taken into account. Abstract The interactions between some acrylic and sulphonic polyanions and some protonated amines (diamines NH 2 -(CH 2 ) x -NH 2 , x =2,…,10; linear tri-, tetra-, penta- and hexa-amines) were studied potentiometrically in aqueous solution, at 25°C. For both types of polyanions AL 2 H i (L − , monomer of polyanion, A, amine) species are formed, with i =1,…, n ( n =number of amino groups in the amine). Aus Styrol und Bisacrylsaurephenylester durch radikalische, vernetzende Polymerisation erhaltene und in Toluol sowie Dioxan noch losliche Copolymere wurden durch alkalische Hydrolyse gezielt in die linear strukturierten Bestandteile zerlegt. Diese enthielten neben sehr vielen Styrolbausteinen wenige aus dem Vernetzer stammende Acrylsaurebausteine. UV-Spektren zeigten, das die Vernetzerreste durch den hydrolytischen Abbau vollstandig abgespalten waren. Von den linearen Baustucken wurden viskosimetrisch, osmometrisch und z.
