Individual Differences in DNA Repair Capacities in Man

After metabolic activation of benzo[a]pyrene to the 7,8-dihydrodiol- 9,10-epoxide, this ultimate carcinogen preferentially binds to the exocyclic amino group of guanine. In order to investigate possible interindividual differenes in the capacity of repair of the DNA adducts formed from benzo[a]- pyrene 7,8-dihydrodiol 9,10-epoxide, their persistence in freshly isolated lymphocytes of several donors was studied. The results show a surprisingly large interindividual variation in DNA adduct formation and their persistence in freshly isolated lymphocytes. Synthesis of Benzo[b]thioxanthenes Benzo[b]thioxanthenes, heterocyclic compounds related to tetracenes and tetracyclines can be obtained by the reaction of 2H-benzo[b]thiete (1) and 1,4-naphthoquinones (3a–g). The primary cycloadducts 4a–g undergo an autoxidation process leading to the quinones 6a–g. The dihydroxy compound 4e shows an additional isomerization by a tetrafold H transfer (4e 5e′). Another preparative route to benzo[b]thioxanthenes makes use of the cycloadditon reaction of 1 and 1,4-epoxynaphthalenes (7a–d). Abstract By treatment with a catalytic amount of DBU in DCM, N-p-toluenesulphonyl carbamates 6a–c , prepared starting from the corresponding Baylis–Hillman adducts, gave (E)-2-(p-toluenesulphonylaminomethyl)propenoates 3a–c , exclusively. On the contrary, changing DABCO for DBU, 2-methylene-3-p-toluenesulfonylamino esters 4a–f were obtained in good yield starting from N-p-toluenesulphonyl carbamates 6a–f . In analogy, N-acyl carbamates 9a–f were treated with DABCO in DCM to give the 2-methylene-3-acylamino esters 5a–f .

Addition reactions of 1,2-dimethylpyrrole and 1-methyl-2-carbomethoxypyrrole with C-acetyl-N-phenylnitrilimine, have been investigated. 1,2-Dimethylpyrrole gives three different types of adducts: i.e. bis-cycloadducts (Vc) and (VIc), spirocycloadduct (IX), and non cyclic bis-adduct (XII). On the other hand, 1-methyl-2-carbomethoxypyrrole gives the bis-cycloadduct (VIb) only. Scandium-catalyzed preparation of cytotoxic 3-functionalized quinolin-2-ones: Regioselective ring enlargement of isatins or imino isatins

Trimethylsilyldiazomethane in the presence of catalytic amounts of Sc(OTf) 3 smoothly promotes the ring expansion of isatins or imino isatins to efficiently afford 3-functionalized quinolin-2-ones through controlled ring enlargement. Whereas the ring-expansion reaction of azetidine-2,3-diones led to the adduct resulting from migration of the carbonyl group, the ring-expansion reaction of oxindole derivatives gave the adduct resulting from migration of the aryl group. To rationalize the experimental observations, theoretical studies have been performed. Steady-state fluorescence emission studies on polyazacyclophane macrocyclic receptors and on their adducts with hexacyanocobaltate(III)

The steady-state fluorescence emission spectra of the azacyclophanes 2,5,8,11-tetraaza[12] paracyclophane (L1), 2,6,9,13-tetraaza[14]paracyclophane (L2), 14,15,17,18-tetramethyl-2,5,8,11-tetraaza[12]paracyclophane (L3) and 16,17,19,20-tetramethyl-2,6,9,13-tetraaza[14]paracyclophane (L4) as a function of pH have been measured. The fully protonated species of each cyclophane gives the highest fluorescence-emission quantum yield. The shapes of the titration curves have been explained by the existence of an electron-transfer quenching effect from a non-protonated amine to the benzene chromophore. Bonding Trends in Lewis Acid Adducts of S4N4 — X-Ray Structure of TeCl4×S4N4.

Tetrasulfur tetranitride and tellurium tetrachloride react in dichloromethane to form a 1:1 adduct TeCl4·S4N4 (1). The crystal structure of 1 shows that TeCl4 is bonded to the S4N4 ring through a Te–N linkage. As a consequence, the transannular S···S bonds in S4N4 are broken and the molecule assumes an open, monocyclic conformation. The Te–N bond of 2.16(1) A is slightly longer than the single bond. The S–N bonds span a range of 1.55(1)–1.67(1) A. The adduct 1 was also characterized by mass spectrometry and Raman spectroscopy. ChemInform Abstract: Aza-Henry Reaction of Isatin Ketimines with Methyl 4-Nitrobutyrate en Route to Spiro[piperidine-3,3′-oxindoles].

A new enantioselective route to spiro[piperidine-3,3′-oxindoles] from isatin ketimines is described. The aza-Henry reaction of N-Boc-isatin ketimines with methyl 4-nitrobutyrate in the presence of a Ph2BOX-CuBr2 complex provided the corresponding nitro amino esters with good diastereoselectivity and excellent enantioselectivity (up to >99% ee). The aza-Henry adducts were transformed into spiro[piperidine-3,3′-oxindoles] after reduction of the nitro group to oxime, and cleavage of the N-Boc group and lactamisation.
