High-resolution crystal structure of an avidin-related protein: insight into high-affinity biotin binding and protein stability.

The chicken avidin gene belongs to an extended gene family encoding seven avidin-related genes (AVRs), of which only avidin is expressed in the chicken. The sequences of AVR4 and AVR5 are identical and the common protein (AVR4) has been expressed both in insect and bacterial systems. The recombinant proteins are similarly hyperthermostable and bind biotin with similarly high affinities. AVR4 was crystallized in the apo and biotin-complexed forms and their structures were determined at high resolution. Molecular mechanisms determining the strength of receptor-mediated intermembrane adhesion

The strength of receptor-mediated cell adhesion is directly controlled by the mechanism of cohesive failure between the cell surface and underlying substrate. Unbinding can occur either at the locus of the specific bond or within the bilayer, which results in tearing the hydrophobic anchors from the membrane interior. In this work, the surface force apparatus has been used to investigate the relationship between the receptor-ligand bond affinities and the dominant mechanism of receptor-coupled membrane detachment. G2(MP2) molecular orbital study of the substituent effect in the H3BPH3−nFn (n=0–3) donor–acceptor complexes

Abstract The complexation energies of H3BPH3−nFn (n=0–3) and the proton affinities of PH3−nFn compounds have been investigated at the G2(MP2) level of theory. G2(MP2) results show that the successive fluorine substitution on the phosphine increases the stability of H3BPH3−nFn complexes although the basicity of the PH3−nFn ligands reduces with this substitution. The NBO partitioning scheme shows that this stability was related to the hyperconjugation effect. It proves also that the shortening of the P–H and P–F bond lengths, upon complexation, is due to an increasing `s' character in these bonds.

A new species of Tococladidae CARPENTER, 1966 from the Permian of France (Insecta: Archaeorthoptera)

Abstract A new forewing venation pattern of the Tococladidae is proposed and the phylogenetic affinities of this family are discussed, based on Tococladus garrici nov. sp. This fossil comes from the Salagou Formation (Permian, Saxonian Group, Lodeve basin, France). The Tococladidae is assigned to the Archaeorthoptera BETHOUX and NEL, 2002 . The biostratigraphic implications of this discovery are briefly discussed, but it is still not possible to infer any definite conclusion about the age of this formation.

Protonation equilibria of some ortho-substituted and annelated aryl and thiophen-2-yl and -3-yl ketones

For some phenyl- (1–8) and thiophen-2-yl (9–11) and thiophen-3-yl (12–14) ketones quantum-mechanical (PM3) calculations have been performed, and for compounds 2, 3, 5–8, 10–14 protonation equilibria have been determined. Phenyl ketones have similar values for the m* parameter and show good linear correlation between the proton affinities calculated in the gas phase and the measured pKBH+ values, which in turn parallel the trend for the calculated carbonyl–phenyl ring dihedral angle. The inhibitory activity of 14 racemic phosphonic acid analogs of phenylglycine, substituted in aromatic rings, towards porcine aminopeptidase N (pAPN) and barley seed aminopeptidase was determined experimentally. The obtained patterns of the inhibitory activity against the two enzymes were similar. The obtained data served as a basis for studying the binding modes of these inhibitors by pAPN using molecular modeling. It was found that their aminophosphonate fragments were bound in a highly uniform manner and that the difference in their affinities most likely resulted from the mode of substitution of their phenyl rings. Novel supramolecular affinity materials based on (−)-isosteviol as molecular templates

The readily available ex-chiral-pool building block (−)-isosteviol was combined with the C3-symmetric platforms hexahydroxytriphenylene and hexaaminotriptycene providing large and rigid molecular architectures. Because of the persistent cavities these scaffolds are very potent supramolecular affinity materials for head space analysis by quartz crystal microbalances. The scaffolds serve in particular as templates for tracing air-borne arenes at low concentration. The affinities of the synthesized materials towards different air-borne arenes were determined by 200 MHz quartz crystal microbalances.
