Construction of an agglutination tool: recombinant Fab fragments biotinylated in vitro.

The pComb3H vector system is used for constructing and panning recombinant antibody libraries. It allows for expression of monovalent Fab fragments, either on the surface of M13 phage, or in the form of soluble proteins secreted into the periplasmic space of bacteria. We constructed a modified pComb3H vector containing cDNA encoding for a 23-amino acid fragment of the Escherichia coli biotin carboxy carrier protein (BCCP), which is an acceptor sequence for biotinylation. The vector was used to express the Fab fragment recognizing human glycophorin A. Effect of α-factor on individual wall mannoproteins fromSaccharomyces cerevisiae acells

Treatment of Saccharomyces cerevisiae a cells with α-factor partially inhibits mannosylation of the high Mr mannoproteins, although there is an increase in the total amount of these molecules present in the wall. They show a similar mobility in SDS-acrylamide gels to those from untreated mnn2 cells. No other significant effects on wall mannoproteins have been observed, except a decrease in the amount of the 29 kDa species.

Grammaticalization in Slavic languages

Abstract This article examines the grammaticalisation developments in Slavic languages. The functions of the past tenses lost in northern Slavic are only partially covered by the younger opposition of perfective and imperfective aspect. The only new classes of morphemes that arose in some sub-areas of Slavic are the definite and the indefinite article, both with preliminary, not-yet-grammaticalised stages in some more Slavic varieties. In sum, in Slavic grammaticalization, phenomena have occurred predominantly in the realm of verbal categories; only very few phenomena are related to the noun phrase.

Im Hautwaschwasser von 28 gesunden Personen wurde mittels des Agglutinationshemmungstests bei uber der Halfte der Probanden Albumin und γ-Globulin nachgewiesen. Das deutet darauf hin, das die menschliche Haut im gesunden Zustand auch fur Makromolekule durchgangig ist, welche bei neutralem pH in wasrige Losungen ubertreten konnen.

The carbohydrate specificities of Ascidia malaca serum hemagglutinins were determined by hemagglutination inhibition tests. Analysis of agglutinins against rabbit and human A, B, O erythrocytes suggests that the size of the combining site corresponds to a disaccharide with a specificity for saccharides containing a D-galacto configuration (D-melibiose, D-raffinose, D-galactose, alpha-lactose, lactulose, L-arabinose). No anomeric specificity was observed with oligosaccharides. Circulating diphtheria antitoxin levels in children aged 11-14 years in relation to the vaccinal history.

Diphtheria antitoxin level in serum samples obtained from 204 healthy children aged 11-14 years was determined by means of an indirect haemagglutination technique and related to the vaccinal history of the subjects. Irrespective of the time since the last toxoid inoculation, the mean antitoxin titre per ml of serum in the individuals who had received incomplete/irregular anti-diphtheria vaccination in childhood was significantly higher when the number of toxoid doses was higher (from two to more than four doses); the same was not observed for individuals given primary vaccination (three toxoid doses) according to the schedule for childhood vaccination in Italy (regular vaccinees) and one or…

Sponge aggregation factor and sponge hemagglutinin: possible relationships between two different molecules.

Abstract A lectin from the marine sponge GEODIA CYDONIUM was isolated and characterized. GEODIA lectin (GL) agglutinates human red blood cells irrespective of the ABO blood group and precipitates with a variety of D -galactose containing glycosubstances, i.e. certain snail galactans, bovine erythrocyte glycoprotein and PNEUMOCOCCUS type XIV polysaccharide. The only simple sugars inhibiting the GL-mediated hemagglutination were lactose and n -acetyl- D -galactosamine. A novel galactose- and arabinose-specific lectin from the sponge Pellina semitubulosa: isolation, characterization and immunobiological properties.

A new lectin from the sponge Pellina semitubulosa is derived which was extracted and purified to homogeneity. The purified lectin is probably a hexamer of polypeptide chains (each M(r) 34,000) which are covalently linked via disulfide linkages; the isoelectric point is 6.1. The lectin displays the following specificities: D-galactose (50% inhibition of hemagglutination at 0.2 mM) = L-arabinose (0.2 mM) greater than D-fucose (1.5 mM) greater than D-glucose (3.0 mM). It precipitates human erythrocytes (A1, A2, A1B, B, and O) with a titer between 2(8) and 2(11) and erythrocytes from sheep and rabbits with a titer between 2(5) and 2(10).
