Proliferation and MHC-unrestricted bystander lysis by virus-specific cytotoxic T cells following antigen self-presentation.
1998
Cytotoxic T cells (CTL) not only act as effector cells, but can also serve as antigen-presenting cells (APC) for other CTL due to their expression of major histocompatibility complex (MHC) class I molecules. In the present study we show that independently derived CTL lines (CTLL) with specificity for an L(d)-presented nonapeptide corresponding to amino acids 168-176 of the immediate-early 1 (IE1) protein of murine cytomegalovirus not only lyse syngeneic but also allogeneic target cells, if the peptide is present during the cytolytic assay. Whereas a short peptide pulse is sufficient to render syngeneic cells susceptible to lysis, continued presence of soluble peptide is mandatory for the ly…
Mobilisation of plasmid-mediated blaVEB-1 gene cassette into distinct genomic islands of Proteus mirabilis after ceftazidime exposure
2021
ABSTRACT Objectives We sought to integrate a VEB-1-encoding gene cassette into the integron of the MDR region of genomic islands (GIs) harboured by Proteus mirabilis strains after antibiotic exposure. Methods An IncP1 plasmid from Achromobacter xylosoxidans carrying the cassette array dfrA14–blaVEB-1–aadB was introduced by conjugation into five strains of P. mirabilis: PmBRI, PmABB, PmSCO and Pm2CHAMA harbouring Salmonella GI 1 and PmESC harbouring Proteus GI 1. Circular intermediates of the cassettes were amplified by PCR. blaVEB-harbouring P. mirabilis were exposed to increasing concentrations of ceftazidime each day. Presence of blaVEB-1 in the GI was assessed by PCR. The complete MDR re…
Flow cytometric assay for estimating fungicidal activity of Amphotericin B in human serum
1992
We describe a simple and rapid bioassay for estimating fungicidal activity of Amphotericin B in human serum using flow cytometry. The method exploits the fact that Candida albicans damaged by Amphotericin B show a decrease in size and take up propidium iodide to exhibit a red fluorescence after deoxycholate treatment. These phenomena display characteristic dose dependencies, and their assessment permits serum fungicidal activity to be broadly grouped into three categories: (1) subfungicidal; (2) fungicidal; and (3) strongly fungicidal. In normal human serum, these three categories correspond to Amphotericin B concentrations of 0 less than or equal to 0.5 micrograms/ml, 0.75-1.5 micrograms/m…

THE RELATIONSHIP BETWEEN INFERENTIAL PROCESSING AND TEXT PROCESSING: A DEVELOPMENTAL STUDY
2012
The research reported here was designed to investigate the critical role played by certain factors implicated in the mental representation of text, and to establish whether their role varies significantly as a function of developmental age. Specifically, it was decided to analyse, in a sample of 180 subjects was selected from three different age groups (7, 10 and 18 years of age respectively), the role of such factors in mediating and influencing the generation of the inferences needed to understand a piece of text characterised by a sequence of information which flows in a logical order, but leads to a conclusion which is contrary to the expectations evoked by the text. In line with this o…
Influence of temperature on the calibration curves in IRMA for neuron specific enolase and its physicochemical interpretation
2009
Abstract Background immunoradiometric assay (IRMA) is one of the principal methods used for the analytical determination of neuron specific enolase (NSE) concentration. We studied the influence of temperature on the calibration curves obtained by this method, and a physicochemical justification based on two theoretical models is proposed. Material and methods we used a commercially available RIA kit for NSE and a gamma counter. Data was analysed using Statistical software. Results and discussion activity bound to the antibody increases with temperature, producing results that are consistent with two modifications to the four parameter and Langmuir equations. Conclusions the two models used …
Anisakid Nematodes and Potential Risk of Human Anisakiasis through the Consumption of Hake, Merluccius spp., Sold Fresh in Spanish Supermarkets
2022
Nematode parasite species belonging to the Anisakis simplex complex are the most important cause of human anisakiasis through the consumption of (mainly) undercooked, previously not frozen, or conveniently treated fish. In Spain, the consumption of hake has been recognized as an important source of this parasitosis. With the aim of shedding light on the risk factors that can influence the potential risk of human anisakiasis in Spain through the consumption of fresh hake sold by nationwide supermarket chains, a total of 536 small hake specimens belonging to the species Merluccius bilinearis caught off the Northeast American coasts and Merluccius merluccius caught in the Northeast Atlantic an…
